Effects of rat and human calcitonin gene-related peptide (r alpha CGRP and h beta CGRP, respectively) upon uterine contractile force were investigated using uterine horns from nonpregnant rats, r alpha CGRP and h beta CGRP were equipotent (pD2 = 8.85-9.09) in inhibiting spontaneous and electrically evoked uterine contractions. r alpha CGRP was relatively ineffective in inhibiting potassium-induced contractures of preparations from stilbestrol-pretreated rats. The use of selective antagonists established that r alpha CGRP did not release prostanoids, or release or act at receptors for catecholamines and histamine. The effects of the peptides were not significantly modulated by estrogen levels since pD2 values were similar (8.56-8.86) in field-stimulated preparations from rats in proestrus/estrus or metestrus/diestrus. 相似文献

Tachykinins (TKs) are a family of peptides involved in the central and peripheral regulation of urogenital functions through the stimulation of TK NK1, NK2 and NK3 receptors. At the urinary system level, TKs locally stimulate smooth muscle tone, ureteric peristalsis and bladder contractions, initiate neurogenic inflammation and trigger local and spinal reflexes aimed to maintain organ functions in emergency conditions. At the genital level, TKs are involved in smooth muscle contraction, in inflammation and in the modulation of steroid secretion by the testes and ovaries. TKs produce vasodilatation of maternal and fetal placental vascular beds and appear to be involved in reproductive function, stress-induced abortion, and pre-eclampsia. Real synaptic systems consist of a nonuniform population of synapses with a broad spectrum of probability and response distributions varying between synapses, and broad amplitude distributions of postsynaptic unitary responses within a given synapse. A common approach to such systems has been to assume identical synapses and recover apparent quantal parameters by deconvolution procedures from measured evoked (ePSC) and unitary evoked postsynaptic current (uePSC) distributions. Here we explicitly consider nonuniform synaptic systems with both intra (type I) and intersynaptic (type II) response variability and formally define an equivalent system of uniform synapses in which both uePSC and ePSC amplitude distributions best approximate those of the actual nonuniform synaptic system. This equivalent system has the advantage of being fully defined by just four quantal parameters: ?, the number of equivalent synapses;p, the mean probability of quantal release; mu, mean; and sigma(2), variance of the uePSC distribution. We show that these equivalent parameters are weighted averages of intrinsic parameters and can be approximated by apparent quantal parameters, therefore establishing a useful analytical link between the apparent and intrinsic parameters. The present study extends previous work on compound binomial analysis of synaptic transmission by highlighting the importance of the product of p and mu, and the variance of that product. Conditions for a unique deconvolution of apparent uniform synaptic parameters have been derived and justified. Our approach does not require independence of synaptic parameters, such as p and mu from each other, therefore the approach will hold even if feedback (i.e., via retrograde transmission) exists between pre and postsynaptic signals. Using numerical simulations we demonstrate how equivalent parameters are meaningful even when there is considerable variation in intrinsic parameters, including systems where subpopulations of high- and low-release probability synapses are present, therefore even under such conditions the apparent parameters estimated from experiments would be informative. 相似文献

Background
Non-productive binding of enzymes to lignin is thought to impede the saccharification efficiency of pretreated lignocellulosic biomass to fermentable sugars. Due to a lack of suitable analytical techniques that track binding of individual enzymes within complex protein mixtures and the difficulty in distinguishing the contribution of productive (binding to specific glycans) versus non-productive (binding to lignin) binding of cellulases to lignocellulose, there is currently a poor understanding of individual enzyme adsorption to lignin during the time course of pretreated biomass saccharification.Results
In this study, we have utilized an FPLC (fast protein liquid chromatography)-based methodology to quantify free Trichoderma reesei cellulases (namely CBH I, CBH II, and EG I) concentration within a complex hydrolyzate mixture during the varying time course of biomass saccharification. Three pretreated corn stover (CS) samples were included in this study: Ammonia Fiber Expansiona (AFEX?-CS), dilute acid (DA-CS), and ionic liquid (IL-CS) pretreatments. The relative fraction of bound individual cellulases varied depending not only on the pretreated biomass type (and lignin abundance) but also on the type of cellulase. Acid pretreated biomass had the highest levels of non-recoverable cellulases, while ionic liquid pretreated biomass had the highest overall cellulase recovery. CBH II has the lowest thermal stability among the three T. reesei cellulases tested. By preparing recombinant family 1 carbohydrate binding module (CBM) fusion proteins, we have shown that family 1 CBMs are highly implicated in the non-productive binding of full-length T. reesei cellulases to lignin.Conclusions
Our findings aid in further understanding the complex mechanisms of non-productive binding of cellulases to pretreated lignocellulosic biomass. Developing optimized pretreatment processes with reduced or modified lignin content to minimize non-productive enzyme binding or engineering pretreatment-specific, low-lignin binding cellulases will improve enzyme specific activity, facilitate enzyme recycling, and thereby permit production of cheaper biofuels.77.

Despite concerted surveys, the Maloti minnow Pseudobarbus quathlambae (Barnard 1938) had not been recorded in South African waters for almost eighty years since the original collections were made at the type locality in the upper uMkhomazana River in 1938. The species was therefore declared extinct in South Africa, whereas extant populations were considered confined to various rivers in the Lesotho highlands. In April 2017, however, this species was rediscovered in the Mzimkhulu River system in KwaZulu-Natal.
