Extracellular matrix (ECM) molecules constitute a "niche" that modulates the migration, proliferation, and differentiation of neural stem/progenitor cells (NSPCs). The glycoprotein Tenascin-R (TN-R) is an ECM molecule, comprising multiple domains. Either the whole TN-R molecule or its distinct domains has been demonstrated to play a very important role in the developing central nervous system. However, little is known about the effect of the TN-R domain on NSPCs, especially NSPC migration. In the present study, we first show that both TN-R domains epidermal growth factor-like repeat (EGFL) and fibronectin type III (FN)6-8 can inhibit the NSPCs migration from neurospheres in vitro. Furthermore, both the EGFL and FN6-8 domains affect the distribution of neurons generated from neurospheres, indicating that EGFL and FN6-8 domains inhibit the motility of neurons generated from neurospheres. These results suggest that TN-R has an inhibitory effect on NSPCs migration. 相似文献

利用中国唯一的稻麦轮作FACE(free-air carbon dioxide enrichment,开放式空气CO2浓度增高)试验平台,研究大气CO2浓度升高对稻季土壤中小麦秸秆降解速率及其氮素分趋的影响.试验设置Ambient(目前空气对照)和FACE(Ambient+200 μmol·mol-1)两个CO2浓度以及低氮处理(LN,150 kg·hm-2)和高氮处理(HN,250 kg·hm-2)两个氮肥水平,在稻季之初按标记麦秸/土壤重量比0.3%添加15N标记小麦秸秆,根据水稻生长时期依次采样测定秸秆降解速率,并通过分析土壤全氮、植株全氮及其15N丰度来观察已降解秸秆的氮素分趋情况.结果发现,大气CO2浓度升高对高氮处理土壤中小麦秸秆降解速率没有显著影响,但显著促进了低氮处理土壤中小麦秸秆的降解(p < 0.05),使其提高到与高氮处理土壤相当水平;大气CO2浓度升高显著增加了已降解秸秆中氮素的流失,在高氮处理土壤中尤为严重,而对植物吸收已降解秸秆中的氮素没有显著影响.结果表明,大气CO2浓度升高在土壤氮素相对不足时会加速土壤中小麦秸秆的降解,而在土壤氮素相对充足时又会加大降解秸秆中氮素的流失. 相似文献

目的:探讨聚丙烯酰胺水凝胶注射隆胸后取出新方法.方法:对48例注射聚丙烯酰胺水凝胶隆胸患者术前行双乳MRI或彩超检查结合触诊准确定位.全麻小切口开放直视下吸出水凝胶,再用大量生理盐水反复灌洗所有腔隙,直至手感探测不到硬结,盐水中无水凝胶为止.结果:48例患者术后无继发感染和出血等并发症.术前诸症状体征消失,无明显乳房变形,乳房形态术后恢复满意.术后双乳MRI复查未见明显异物残留.术后1 ~12月(6.2±0.3月)45例随访复查MRI亦未见异物残留,乳房修复完好.结论:注射隆乳后腔内大量盐水灌洗辅助取出水凝胶具有创伤小、出血少、操作简单、费用低廉的优点,是一种较好的、值得推广的凝胶取出术式. 相似文献

Objective
The decreased expression of muscle-specific microRNA-1 (miR-1) has been found in many cardiovascular diseases and is considered to contribute to heart failure (HF). Here we investigated the role of miR-1 in myocardium protection by infusion of miR-1 in a cardiac global miRNA-deficient mouse.Methods
We generated a cardiac-selective miRNA-deficient mouse by crossing Dicerflox/flox mice with mice expressing tamoxifen-inducible Cre recombinase under the control of a mouse αMHC promoter. When Dicer gene was removed following tamoxifen injection, the mice were treated with micrONTM mmu-miR-1a-3p agomir (agomir-1). The mice were subjected to echocardiography measurement, and the heart tissue specimens were stained with hematoxylin and eosin (H&E) and Sirius red. Terminal deoxynucleotidyl transferase-mediated dUTP nickend labeling assay and Ki67 immunofluorescence were used to determine apoptosis and proliferation.Results
Dicer deletion resulted in extensive decrease in cardiac miRNAs in the mice. In echocardiography, the mice developed rapid and dramatic left ventricular enlargement. In histology, apparent cardiomyocyte hypertrophy, myofiber disarray, ventricular fibrosis, inflammatory infiltration, and severe ventricular remodeling were exhibited. When the mice were treated with agomir-1, they did not show any significant abnormalities in heart structure and histology in response to Dicer ablation.Conclusion
The proper expression of miRNAs plays vital roles in the maintenance of heart histology and function. Among these miRNAs, miR-1 is critical to inhibit myocyte hypertrophy and extracellular matrix deposition, thereby preventing cardiac remodeling in cardiac-selective Dicer deficient mice. 相似文献56.

构建了马铃薯Y病毒中国株系(PVY-C)蚜传辅助成分(HC-Pro)基因的正义、反义和缺失三种植物表达载体,通过农杆菌介导法转化烟草品种NC89。Southern blot分析表明,HC-Pro基因及其突变体已经整合到烟草染色体中,Western blot分析证明,正义HC-Pro基因及其缺失突变体在转基因烟草中有表达产物,攻毒试验结果表明,转正义,HC-Pro基因及其缺失突变体不仅能够提高T1转基因烟草中PVY-C的病毒积累和致病,而且对异源病毒PVX具有同样的作用,而转反义HC-Pro基因烟草对PVY-C和PVX的致病性无影响,因此,PVY-C HC-Pro基因介导PVX/PVY的协作作用。 相似文献

Dexamethasone (Dex), a synthetic glucocorticoid (GC) with long-lasting treatment effects, has been proved to exert a modulatory effect on osteoblast proliferation and differentiation during embryonic osteogenesis. However, it is still controversial if Dex exposure influences endochondral ossification and the underlying mechanism. In this study, chick embryos in vivo and preosteoblast cell cultures in vitro were utilized to investigate the effects of Dex on osteoblast formation and differentiation during the skeletal development. We first demonstrated that Dex exposure could shorten the long bones of 17-day chick embryos in vivo, and also downregulated the expressions of osteogenesis-related genes. Next, we established that Dex exposure inhibited the proliferation and viability of preosteoblasts-MC3TC-E1 cells, and the addition of insulin-like growth factor 1 (IGF-1) could dramatically rescue these negative effects. On the basis of remarkable changes in the rescue experiments, we next verified the important role of angiogenesis in osteogenesis by culturing isolated embryonic phalanges in Dulbecco's modified Eagle's medium culture or on the chick chorioallantoic membrane (CAM). Then, we transplanted MC3T3-E1 cell masses onto the CAM. The data showed that Dex exposure reduced the vessel density within the developed cell mass, concomitantly with the downregulation of IGF-1 pathway. We verified that the inhibition of blood vessel formation caused by Dex could be rescued by IGF-1 treatment using the CAM angiogenesis model. Eventually, we demonstrated that the shortened length of the phalanges in the presence of Dex could be reversed by IGF-1 addition. In summary, these findings suggested that the inhibition of Igf-1 signal caused by Dex exposure exerts a detrimental impact on the formation of osteoblasts and angiogenesis, which consequently shortens long bones during osteogenesis. 相似文献

单穗升麻的雌蕊群由1~5枚离生心皮组成。子房单室,4~9枚倒生胚珠,双珠被,厚珠心;大孢子四分体呈线形或T形排列,合点端一个具功能。胚囊发育为蓼型。成熟胚囊中3个单核反足细胞,胞质浓密,在球形胚时期尚能观察到其退化痕迹。极核融合早,次生核位于合点端。胚胎发生为柳叶菜型;核型胚乳。雄蕊凋谢后1~2天花柱顶端腹缝线周围出现乳突细胞,雄蕊凋谢后3~5天延长成柱头毛。讨论了单穗升麻与南川升麻柱头可授期与花粉生活力的差异对传粉效果和结籽率的显著影响。 相似文献

社会性动物的个性特征是对环境和社群的适应性结果,对个性的研究是探究群居动物复杂社会关系及其差异的重要内容。相对于个性特征评级法和行为编码法,自然观察法是分析判断野生动物个性特征较为理想的方法之一,最大限度地突出了野生动物的自然属性及个性特征类型。本研究以栖息于安徽黄山的野生短尾猴投食群体鱼鳞坑A1群为研究对象,采用目标动物取样法和全事件记录法,于2015年7月至2016年6月期间采集成年个体自然发生的行为数据,并采用主成分分析方法判断成年个体的个性特征及类型,并分析性别差异。结果表明:成年雄性个体具有5种个性特征,即领导型、内向型、社交型、孤僻型以及受欺凌型,成年雌性个体具有6种个性特征,即领导型、孤僻型、内向型、不安型、神经质型以及友好型。研究结果与已有结果一致,说明自然观察法可能是分析野生动物个性类型的较好
