In a current procedure for periodontal tissue regeneration, enamel matrix derivative (EMD), which is the active component, is mixed with a propylene glycol alginate (PGA) gel carrier and applied directly to the periodontal defect. Exposure of EMD to physiological conditions then causes it to precipitate. However, environmental changes during manufacture and storage may result in modifications to the conformation of the EMD proteins, and eventually premature phase separation of the gel and a loss in therapeutic effectiveness. The present work relates to efforts to improve the stability of EMD-based formulations such as Emdogain™ through the incorporation of arginine, a well-known protein stabilizer, but one that to our knowledge has not so far been considered for this purpose. Representative EMD-buffer solutions with and without arginine were analyzed by 3D-dynamic light scattering, UV-Vis spectroscopy, transmission electron microscopy and Fourier transform infrared spectroscopy at different acidic pH and temperatures, T, in order to simulate the effect of pH variations and thermal stress during manufacture and storage. The results provided evidence that arginine may indeed stabilize EMD against irreversible aggregation with respect to variations in pH and T under these conditions. Moreover, stopped-flow transmittance measurements indicated arginine addition not to suppress precipitation of EMD from either the buffers or the PGA gel carrier when the pH was raised to 7, a fundamental requirement for dental applications. 相似文献

Roger Waldinger's provocative book on migrants’ cross-border lives corroborates many insights already established by transnational migration scholarship. He makes a compelling case for the need to understand migration not just as assimilation – neo, segmented, or otherwise – but also as about how people continue to be simultaneously involved in their homelands, the countries where they settle, and other salient places. I am glad that Waldinger has lent his voice to correct the unproductive divide between immigration and emigration scholarship. But the ‘new’ path he charts forward to right what he sees as wrong with current scholarship is, at times, so selectively formulated and argued that it undercuts its effectiveness. 相似文献

As marine systems are threatened by increasing human impacts, mechanisms to maintain biodiversity and ecosystem functions and services are needed. Protecting areas of conservation importance may serve as a proxy for maintaining these functions, while also facilitating efficient use and management of limited resources. Biodiversity hotspots have been used as surrogates for spatial conservation importance; however, as many protected areas have been established opportunistically and under differing criteria, it is unclear how well they actually protect hotspots. We evaluated how well the current protected area network and priority areas selected through previous systematic conservation planning exercises preserve biodiversity hotspots in the Gulf of California, Mexico. We also determined spatial congruence between biodiversity hotspots based on different criteria, which may determine their ability to be used as surrogates for each other. We focus on the Gulf of California because it is a megadiverse system where limited information regarding species diversity and distribution has constrained development of strategies for conservation and management. We developed a species occurrence database and identified biodiversity hotspots using four different criteria: species richness, rarity, endemism, and threatened species. We interpolated species occurrence, while accounting for heterogeneous sampling efforts. We then assessed overlap of hotspots with existing protected areas and priority areas, and between hotspots derived by distinct criteria. We gathered 286,533 occurrence records belonging to 12,105 unique species, including 6388 species identified as rare, 642 as endemic, and 386 as threatened. We found that biodiversity hotspots showed little spatial overlap with areas currently under protection and previously identified priority areas. Our results highlight the importance of distinct spatial areas of biodiversity and suggest that different ecological mechanisms sustain different aspects of diversity and multiple criteria should be used when defining conservation areas. 相似文献

No-take marine reserves can be powerful management tools, but only if they are well designed and effectively managed. We review how ecological guidelines for improving marine reserve design can be adapted based on an area’s unique evolutionary, oceanic, and ecological characteristics in the Gulf of California, Mexico. We provide ecological guidelines to maximize benefits for fisheries management, biodiversity conservation and climate change adaptation. These guidelines include: representing 30% of each major habitat (and multiple examples of each) in marine reserves within each of three biogeographic subregions; protecting critical areas in the life cycle of focal species (spawning and nursery areas) and sites with unique biodiversity; and establishing reserves in areas where local threats can be managed effectively. Given that strong, asymmetric oceanic currents reverse direction twice a year, to maximize connectivity on an ecological time scale, reserves should be spaced less than 50–200 km apart depending on the planktonic larval duration of target species; and reserves should be located upstream of fishing sites, taking the reproductive timing of focal species in consideration. Reserves should be established for the long term, preferably permanently, since full recovery of all fisheries species is likely to take?>?25 years. Reserve size should be based on movement patterns of focal species, although marine reserves?>?10 km long are likely to protect?~?80% of fish species. Since climate change will affect species’ geographic range, larval duration, growth, reproduction, abundance, and distribution of key recruitment habitats, these guidelines may require further modifications to maintain ecosystem function in the future. 相似文献

The location and activity of a K+-ATPase in mature, dormant peas were investigated using two ultracytochemical techniques, as well as biochemical assays of plasma membrane fractions from separate seed parts. Both the Wachstein and Meisel (1957) and the Ernst (1972) cytochemical methods showed plasma membrane-associated reaction product located primarily on the exterior surfaces of the entire pea embryo, except for the stem apex and tip-most cells of the radicle. No plasma membrane-assocated reaction product was found in the seed coat, which typically consists of cells with degenerating protoplasts. Biochemical results showed the highest specific K+-ATPase activity in the radicles (775 nmol Pi/mg protein/hr), followed by epicotyls (168 nmol Pi/mg protein/hr) and cotyledons (147 nmol Pi/mg protein/hr). It is proposed that the entire pea embryo may function in the active absorption of nutrients during the initial phases of germination. Additional functions of the enzyme may include cell wall loosening prior to cell elongation, regulation of cytoplasmic pH, and the generation of turgor. 相似文献

The genetic diversity among a worldwide collection of 120 strains of Ralstonia solanacearum was assessed by restriction fragment length polymorphism (RFLP) analysis of amplified fragments from the hrp gene region. Five amplified fragments appeared to be specific to R. solanacearum. Fifteen different profiles were identified among the 120 bacterial strains, and a hierarchical cluster analysis distributed them into eight clusters. Each cluster included strains belonging to a single biovar, except for strains of biovars 3 and 4, which could not be separated. However, the biovar 1 strains showed rather extensive diversity since they were distributed into five clusters whereas the biovar 2 and the biovar 3 and 4 strains were gathered into one and two clusters, respectively. PCR-RFLP analysis of the hrp gene region confirmed the results of previous studies which split the species into an “Americanum” division including biovar 1 and 2 strains and an “Asiaticum” division including biovar 3 and 4 strains. However, the present study showed that most of the biovar 1 strains, originating from African countries (Reunion Island, Madagascar, Zimbabwe, and Angola) and being included in a separate cluster, belong to the “Asiaticum” rather than to the “Americanum” division. We analyzed the pathogenicity of chitin synthetase (chs) disruptants of Ustilago maydis obtained with the carboxin-resistant or the hygromycin-resistant cassettes. We found that only chitin synthetase (chs) mutants obtained by gene disruption with the carboxin resistance cassette lost their virulence to maize (Zea mays) seedlings. Carboxin is a systemic fungicide that inhibits respiration by preventing the oxidation of succinate. We demonstrated
that carboxin-resistant transformants were affected in the levels of succinate dehydrogenase and respiratory activities when
compared with hygromycin-resistant disruptants. We propose that loss of virulence in the carboxin-resistant transformants
is owing to loss of respiratory fitness, which probably represents an important component of virulence in this fungus.

Mining the isotopic complexity of nitrous oxide: a review of challenges and opportunities 总被引:1,自引:0,他引:1
Nitrous oxide (N2O) is an important focus of international greenhouse gas accounting agreements and mitigation of emissions will likely depend on understanding the mechanisms of its formation and reduction. Consequently, applications of stable isotope techniques to understand N2O cycling are proliferating and recent advances in technology are enabling (1) increases in the frequency of isotope analyses and (2) analyses not previously possible. The two isotopes of N and 3 isotopes of O combine to form a total of 12 possible isotopic molecules of N2O. Consequently, this remarkably simple molecule contains a wealth of isotopic information in the form of bulk (δ15N, δ18O), position dependent (site preference), mass independent (Δ17O) and multiply-substituted or clumped isotope compositions. With recent developments in high-mass resolution double sector instruments all 12 isotopic molecules will likely be resolved in the near future. Advances in spectroscopic instruments hold the promise of substantial increases in sample throughput; however, spectroscopic analyses require corrections due to interferences from other gases and frequent and accurate calibration. Mass spectrometric approaches require mass overlap corrections that are not uniform between research groups and interlaboratory comparisons remain imprecise. The continued lack of attention to calibration by both funding agencies and investigators can only perpetuate disagreement between laboratories in reported isotope values for N2O that, in turn, will compromise global assessments of N2O sources and sinks based on isotope ratios.
