The response of rocky shore ecosystems to increased nutrient availability was examined in eight land-based mesocosms designed for hard-bottom littoral communities built at Marine Research Station Solbergstrand (Norway). The average seawater volume in each basin was 9 m3 with an average water residence time of about 2 h. A tidal regime resembling that in the fjord was maintained in the basins, and waves were generated regularly. NH4NO3 and H3PO4, at a constant molar NP ratio of 16:1, was added into 6 basins at concentrations 1, 2, 4, 8, 16, 32 M DIN above the background DIN concentration during 1 1/2 years. Two mesocosms were kept as control treatment. Marine communities were introduced into the basins two years prior to the start of nutrient dosage. The effects of nutrient enrichment were few and only marginal during the first year of nutrient addition, while some effects became more obvious during the second year. The growth rate of the periphyton and fast-growing macroalgae communities was stimulated by nutrient enrichment, while the response was less evident among the perennial fucoids. The structure of the macroalgal communities, however, did not change during 16 months' measurements. In contrast, growth on artificial rock substrates during the same period of time revealed intensive growth of the fast-growing Ulva lactuca in high-dosed basins compared with low-dosed and control basins, which were dominated by the fucoid Fucus serratus. The fauna communities exhibited only a minor response to nutrient treatment. The common periwinkle Littorina littorea, however, appeared with increased abundance in the high-dosed basins. The total system metabolism tended to increase slightly, but not significantly, with increased nutrient loading.  相似文献

Osmotic shrinkage of Ehrlich ascites tumor cells (EATC) elicited translocation of myosin II from the cytosol to the cortical region, and swelling elicits concentration of myosin II in the Golgi region. Rho kinase and p38 both appeared to be involved in shrinkage-induced myosin II reorganization. In contrast, the previously reported shrinkage-induced actin polymerization [Pedersen et al. (1999) Exp. Cell Res. 252, 63-74] was independent of Rho kinase, p38, myosin light chain kinase (MLCK), and protein kinase C (PKC), which thus do not exert their effects on the shrinkage-activated transporters via effects on F-actin. The subsequent F-actin depolymerization, however, appeared MLCK- and PKC-dependent, and the initial swelling-induced F-actin depolymerization was MLCK-dependent; both effects were apparently secondary to kinase-mediated effects on cell volume changes. NHE1 in EATC is activated both by osmotic shrinkage and by the serine/threonine phosphatase inhibitor Calyculin A (CL-A). Both stimuli caused Rho kinase-dependent myosin II relocation to the cortical cytoplasm, but in contrast to the shrinkage-induced F-actin polymerization, CL-A treatment elicited a slight F-actin depolymerization. Moreover, Rho kinase inhibition did not significantly affect NHE1 activation, neither by shrinkage nor by CL-A. Implications for the possible interrelationship between changes in F-actin and myosin II, protein phosphorylation, and cell volume regulation are discussed.  相似文献

In a reaction medium mixture of 9:11 t-BuOH and pyridine (v/v) the effect of fatty acid chain length (C-4-C-12) on C. antarctica lipase B (Novozym 435, EC 3.1.1.3) catalysed esterification was studied. alpha and beta maltose 6'-O-acyl esters in an anomeric molar ratio of 1.0:1.1 were synthesised independently of the chain length, but the initial specific reaction rate increased with decreasing chain length of the acyl donor. The product yield followed the same trend with a lauryl ester yield of 1.1% (mol/mol) and a butyl ester yield of 27.6% (mol/mol) after 24 h of reaction. With sucrose as the acyl acceptor the 6'-O-acyl and 6-O-acyl monoesters were formed with fatty acids of chain length C-4 and C-10 while the 6',6-O-acyl diester was formed only with butanoic acid (C-4:0) as acyl donor. The 6'-O-acyl and 6-O-acyl monoesters and the 6',6-O-acyl diester of butanoic acid were produced in a molar ratio of 1.0:0.5:0.2 and with decanoic acid (C-10:0) the 6'-O-acyl and 6-O-acyl monoesters were formed in the ratio of 1.0:0.3. The highest initial reaction rate and yield were obtained with the shortest chain length of the acyl donor. Initial reaction rates and ester yields were affected by the solubility of the disaccharide, with higher reaction rates and yields with maltose than with sucrose, while no formation of esters were observed with either cellobiose or lactose as acyl acceptors.  相似文献

Pit1 is the human receptor for gibbon ape leukemia virus (GALV) and feline leukemia virus subgroup B (FeLV-B), while the related human protein Pit2 is a receptor for amphotropic murine leukemia virus (A-MuLV). The A-MuLV-related isolate 10A1 can utilize both Pit1 and Pit2 as receptors. A stretch of amino acids named region A was identified in Pit1 (residues 550 to 558 in loop 4) as critical for GALV and FeLV-B receptor function. We have here investigated the role of region A in A-MuLV and 10A1 entry. Insertion of a single amino acid in region A of mouse Pit1 resulted in a functional A-MuLV receptor, showing that region A plays a role in A-MuLV infection. Moreover, the downregulation of 10A1 receptor function by changes in region A of human Pit1 indicates that this region is also involved in 10A1 entry. Therefore, region A seems to play a role in infection by all viruses utilizing Pit1 and/or Pit2 as receptors.  相似文献

Abstract: Many cases of autosomal dominant inherited forms of early-onset Alzheimer's disease are caused by mutations in the genes encoding presenilin-1 (PS-1; chromosome 14) and presenilin-2 (PS-2; chromosome 1). PSs are expressed in neurons throughout the brain wherein they appear to be localized primarily to the endoplasmic reticulum (ER) of cell bodies and dendrities. PS-1 and PS-2 show high homology and are predicted to have eight transmembrane domains with the C terminus, N terminus, and a loop domain all on the cytosolic side of the membrane; an enzymatic cleavage of PSs occurs at a site near the loop domain. The normal function of PSs is unknown, but data suggest roles in membrane trafficking, amyloid precursor protein processing, and regulation of ER calcium homeostasis. Homology of PSs to the C. elegans gene sel-12 , which is involved in Notch signaling, and phenotypic similarities of PS-1 and Notch knockout mice suggest a developmental role for PSs in the nervous system. When expressed in cultured cells and transgenic mice, mutant PSs promote increased production of a long form of amyloid β-peptide (Aβ1-42) that may possess enhanced amyloidogenic and neurotoxic properties. PS mutations sensitize cultured neural cells to apoptosis induced by trophic factor withdrawal, metabolic insults, and amyloid β-peptide. The mechanism responsible for the proapoptotic action of mutant PSs may involve perturbed calcium release from ER stores and increased levels of oxidative stress. Recent studies of apoptosis in many different cell types suggest that ER calcium signaling can modulate apoptosis. The evolving picture of PS roles in neuronal plasticity and Alzheimer's disease is bringing to the forefront the ER, an organelle increasingly recognized as a key regulator of neuronal plasticity and survival.  相似文献

The accuracy ofthe acoustic reflections method for the evaluation of human nasalairway geometry is determined by the physical limitations of thetechnique and also by the in vivo deviations from the assumptions ofthe technique. The present study 1)examines the sound loss caused by nonrigidity of the nasal mucosa andviscous loss caused by complex geometry and its influence on theestimation of the acoustic area-distance function;2) examines the optimal relation between sampling frequency and low-pass filtering, and 3) evaluates advantages of breathingHe-O2 during the measurements onaccuracy. Measurements made in eight plastic models, withcavities exactly identical to the "living" nasal cavities,revealed only minor effects of nonrigidity of the nasal mucosa. Thiswas confirmed by an electrical analog model, based on laser vibrometryadmittance measurements of the nasal mucosa, which indicated that theerror in the acoustic measurements caused by wall motion isinsignificant. The complex geometry of the nasal cavity per se (i.e.,departure from circular) showed no significant effects on themeasurements. Low-pass filtering of the signal is necessary to cut offcross modes arising in the nasal cavity. Computer simulations andmeasurements in models showed that the sampling frequency should beapproximately four times the low-pass filtering frequency (i.e., twicethe Nyquist frequency) to avoid influence on the result. No advantagewas found for the the use of He-O2vs. air in the nasal cavity.

The influence of growth irradiance on the non-steady-state relationship between photosynthesis and tissue carbon (C) and nitrogen (N) pools in Chaetomorpha linum (Muller) Kutzing in response to abrupt changes in external nitrogen (N) availability was determined in laboratory experiments. For a given thallus N content, algae acclimated to low irradiance consistently had a higher rate of light-saturated photosynthesis (Pmax normalized to dry weight) than algae acclimated to saturating irradiance; for both treatments, Pmax was correlated to thallus N. Both Pmax and the photosynthetic efficiency (αdw) were correlated in C. linum grown at either saturating or limiting irradiance over the range of experimental conditions, indicating that variations in electron transport were coupled to variations in C-fixation capacity despite the large range of tissue N content from 1.1% to 4.8%. Optimizing both α and Pmax and thereby acclimating to an intermediate light level may be a general characteristic of thin-structured opportunistic algae that confers a competitive advantage in estuarine environments in which both light and nutrient conditions are highly variable. Nitrogen-saturated algae had the same photosynthesis–irradiance relationship regardless of light level. When deprived of an external N supply, photosynthetic rates did not change in C. linum acclimated to low irradiance despite a two-fold decrease in tissue N content, suggesting that the active pools of chlorophyll and Rubisco remained constant. Both α and Pmax decreased immediately and continuously in algae acclimated to high irradiance on removal of the N supply even though tissue N content was relatively high during most of the N-starvation period, indicating a diversion of energy and reductant away from C fixation to support high growth rates. Carbon and nitrogen assimilation were equally balanced in algae in both light treatments throughout the N-saturation and -depletion phases, except when protein synthesis was limited by the depletion of internal N reserves in severely N-starved high-light algae and excess C accumulated as starch stores. This suggests that the ability for short-term adjustment of internal allocation to acquire N andC in almost constant proportions may be especially beneficial to macroalgae living in environments characterized by high variability in light levels and nutrient supply.  相似文献

Reaction of L-ascorbic acid with hydrogen bromide in acetic acid gave 6-bromo-6-deoxy-L-ascorbic acid, which was converted into 5,6-dideoxy-D-glycero-hex-2,3-enono-1,4-lactone. Hexonic acids or their lactones also gave bromo compounds on treatment with HBrAcOH. From D-galactono-1,4-lactone a 6-bromo derivative was obtained. Calcium D-gluconate yielded 2,6-dibromo-2,6-dideoxy-D-mannono-1,4-lactone, whereas D-mannono-1,4-lactone gave 2,6-dibromo-2,6-dideoxy-D-glucono-1,4-lactone.  相似文献

Comparative culture studies on isolates from Lanzarote (Canary Islands) and from Danish waters of Giraudia sphacelarioides show that temperature plays the key role to determine its geographical distribution. Experiments show that the upper lethal temperature is the same for both isolates 26.5—-31.1°C for microthalli, and formation of macrothalli is inhibited between 23.2 and 26.5°C. The lower lethal temperature for both micro- and macrothalli in the Lanzarote isolate is between 10.8° C and 7.6°C, while the Danish isolate survives 54°C. The microthallus is thus the more persistent stage in relation to temperature, while the macrothalli occur only when more suitable conditions exist in nature. In the North Atlantic Ocean G. sphacelarioides is distributed over approximately 30° latitudes with severe winter temperatures especially in Danish waters. This distribution can only be explained by ecotypic differentiation. In Danish waters salinity decreases from c. 30 ‰ in the northern Kattegat to below 8 ‰ in the inner parts and therefore becomes an important factor for the immigration of species into the Baltic Sea. Combined salinity (30–20–10–5 ‰) and temperature (4 to 30°C) experiments show that the Lanzarote isolate only grows at 20 ‰ in an attenuated temperature interval and at 10 ‰ only fertile microthalli are observed, however, with limited liberation of swarmers. The Danish isolate grows and reproduces also at salinities from 20–5 ‰ both as microand macrothalli, but in a still more attenuated temperature interval, and the morphology of the macrothalli becomes increasingly aberrant vs. decreasing salinity. This is in agreement with the fact that G. sphacelarioides is reported from the northern part of Kattegat with salinity from 28—-25 ‰, and it is only recorded a few times in the southern part of Kattegat and limited by the 10 ‰ isohaline.  相似文献
