To test the influence of past vicariant events on population genetic structure of the sooty tern Sterna fuscata , we examined sequence variation in the mitochondrial control region of individuals from the Indo-Pacific and Atlantic Oceans. Our analyses indicate a rapid population expansion at a global scale during the last 100 000 years, consistent with global recolonisation during the interstade following the Pleistocene glacial maxima (125 000–175 000 years bp). We estimate islands of the Great Barrier Reef and Coral Sea were colonised no more than 16 000 years ago, most likely in association with the appearance of new breeding habitat following the final Pleistocene glacial retreat (19 000–22 000 years bp). Our results suggest that ice sheets linked to major glacial events not only impact genetic structuring in temperate seabirds, but that sea level changes in the tropics associated with these same events have also significantly impacted contemporary genetic structuring in tropical seabird species.  相似文献

The periplasmic hydrogenase containing equivalent amounts of nickel and selenium plus non-heme iron [NiFeSe) hydrogenase) has been purified from cells of the sulfate reducing bacterium Desulfovibrio baculatus (DSM 1748) grown on a lactate/sulfate medium containing natural Se isotopes and the nuclear isotope, 77Se. Both the 77Se-enriched and unenriched hydrogenases were shown to be free of other hydrogenases and characterized with regard to their Se contents. EPR studies of the reduced nickel signal generated by redox titrations of the enriched and unenriched (NiFeSe) hydrogenases demonstrated that the gx = 2.23 and gy = 2.17 resonances are appreciably broadened by the spin of the 77Se nucleus (I = 1/2). This observation demonstrates unambiguously that the unpaired electron is shared by the Ni and Se atoms and that Se serves as a ligand to the nickel redox center of the (NiFeSe) hydrogenase.  相似文献

Existing methods for detection of food-borne pathogens and their toxins are frequently time-consuming, require specialized equipment, and involve lengthy culture procedures and/or animal testing and are thus unsuitable for a rapid response to an emergency public health situation. A series of simple and rapid affinity immunochromatography column (AICC) assays were developed to detect Clostridium botulinum neurotoxin types A, B, E, and F and Escherichia coli O157 in food matrices. Specifically, for milk, grape juice with peach juice, and bottled water, the detection limit for the botulinum neurotoxin type A complex was 0.5 ng. Use of this method with a 10-ml sample would therefore result in a detection limit of 50 pg ml−l. Thus, this assay is approximately 2 orders of magnitude more sensitive than a comparable lateral-flow assay. For botulinum neurotoxin complex types B, E, and F, the minimum detection limit was 5 ng to 50 ng. Sensitive detection of E. coli O157 was achieved, and the detection limit was 500 cells. The AICC test was also shown to be specific, rapid, and user friendly. This test takes only 15 to 30 min to complete without any specialized equipment and thus is suitable for use in the field. It has the potential to replace existing methods for presumptive detection of botulinum neurotoxin types A, B, E, and F and E. coli O157 in contaminated matrices without a requirement for preenrichment.The majority of conventional methods used for detection and identification of pathogenic microorganisms, viruses, and/or their toxins lack the speed and sensitivity necessary for use in the field (they typically are not completed in a single day) and also require specialized equipment (20). Rapid methods, including antibody-based and nucleic acid-based assays, have revolutionized the methodology for detection of microbial pathogens and their toxins in foods (16). However, while most antibody-based and nucleic acid-based assays are rapid, specialized equipment is often required, and specific enrichment is needed to achieve the necessary sensitivity. This means that the analysis time can still be several days (16). Lateral-flow assays (LFAs) and column flow assays are tests that have considerable merit in terms of rapidity and ease of use in the field without specialized equipment (4, 5, 8, 19, 34).Two contrasting agents were used as detection targets in this study: (i) a potent microbial toxin (Clostridium botulinum neurotoxin), including type A, B, E, and F neurotoxins; and (ii) an infectious pathogen, Escherichia coli O157. These two targets present different problems for detection; the first target is a protein toxin, and the second target is intact bacterial cells. The botulinum neurotoxin is the most potent toxin known, and as little as 30 to 100 ng has the potential to be fatal to humans (28). It is responsible for botulism, a severe neuroparalytic disease that affects humans and also animals and birds (28). There are seven antigenically distinct botulinum neurotoxins (types A to G), and a number of subtypes have also been described (9, 11, 15, 28, 36). Botulism in humans is associated principally with neurotoxin types A, B, E, and F (27, 29). Since the botulinum neurotoxins are the toxic agents and they can be produced by six physiologically distinct clostridia (28), considerable emphasis has been placed on detection of the neurotoxins rather than the bacteria. The “gold standard” method for detecting botulinum neurotoxins is the mouse bioassay due to its high levels of sensitivity and specificity. However, this technique is also problematic (33). It typically requires 24 to 48 h to yield results, is expensive, and is becoming less favored because of its use of animals (4). The alternative tests include enzyme-linked immunosorbent assays (ELISAs), lateral-flow assays (LFAs), a chemiluminescent slot blot immunoassay, surface plasmon resonance (SPR), the assay with a large immunosorbent surface area (ALISSA) test, and quantum dot immunoassays (4, 5, 7, 22, 43, 46). Lateral-flow assays are available and are convenient for toxin testing as they are easy to perform and rapid (<30 min) and no additional equipment is required. However, their poor sensitivity has limited their use (23).E. coli O157 produces a cytotoxin (verotoxin), and an E. coli O157 infection can lead to severe bloody diarrhea, kidney failure, brain damage, and death. Enumeration, identification, and control of this pathogen are challenging due to the low infectious dose necessary to cause disease, which is between 2 and 2,000 ingested cells (41). Sources of E. coli O157 infection include ground beef and unpasteurized milk and apple juice (1), raw milk (6), and spinach and lettuce (42). Isolation of E. coli O157:H7 from water, food, and environmental samples is laborious. Culture is difficult due to the large competing microflora that either overgrows or mimics the non-sorbitol-fermenting organism E. coli O157:H7 (12). According to Tokarskyy and Marshall (41), the largest group of rapid test kits commercially available for testing for the presence of E. coli O157 in food includes immunological methods, such as latex agglutination, reverse passive latex agglutination, immunodiffusion, ELISA, immunomagnetic separation (IMS), and immunoprecipitation. The other methods that have been developed include a dipstick test device (2), a lateral-flow immunoassay (8), real-time PCR (39), and an enzyme-linked immunomagnetic chemiluminescent assay (17). However, in many cases these tests require preenrichment or have limited sensitivity.The objective of the work described here was to develop a rapid sensitive diagnostic test for detection of botulinum neurotoxins A, B, E, and F and E. coli O157 that can be used without preenrichment.  相似文献

Organic Lake is a shallow, marine-derived hypersaline lake in the Vestfold Hills, Antarctica that has the highest reported concentration of dimethylsulfide (DMS) in a natural body of water. To determine the composition and functional potential of the microbial community and learn about the unusual sulfur chemistry in Organic Lake, shotgun metagenomics was performed on size-fractionated samples collected along a depth profile. Eucaryal phytoflagellates were the main photosynthetic organisms. Bacteria were dominated by the globally distributed heterotrophic taxa Marinobacter, Roseovarius and Psychroflexus. The dominance of heterotrophic degradation, coupled with low fixation potential, indicates possible net carbon loss. However, abundant marker genes for aerobic anoxygenic phototrophy, sulfur oxidation, rhodopsins and CO oxidation were also linked to the dominant heterotrophic bacteria, and indicate the use of photo- and lithoheterotrophy as mechanisms for conserving organic carbon. Similarly, a high genetic potential for the recycling of nitrogen compounds likely functions to retain fixed nitrogen in the lake. Dimethylsulfoniopropionate (DMSP) lyase genes were abundant, indicating that DMSP is a significant carbon and energy source. Unlike marine environments, DMSP demethylases were less abundant, indicating that DMSP cleavage is the likely source of high DMS concentration. DMSP cleavage, carbon mixotrophy (photoheterotrophy and lithoheterotrophy) and nitrogen remineralization by dominant Organic Lake bacteria are potentially important adaptations to nutrient constraints. In particular, carbon mixotrophy relieves the extent of carbon oxidation for energy production, allowing more carbon to be used for biosynthetic processes. The study sheds light on how the microbial community has adapted to this unique Antarctic lake environment.  相似文献

Most populations of migrant shorebirds around the world are in serious decline, suggesting that vital condition-dependent rates such as fecundity and annual survival are being affected globally. A striking example is the red knot (Calidris canutus rufa) population wintering in Tierra del Fuego, which undertakes marathon 30,000 km hemispheric migrations annually. In spring, migrant birds forage voraciously on horseshoe crab eggs in Delaware Bay in the eastern USA before departing to breed in Arctic polar deserts. From 1997 to 2002 an increasing proportion of knots failed to reach threshold departure masses of 180-200 g, possibly because of later arrival in the Bay and food shortage from concurrent over-harvesting of crabs. Reduced nutrient storage, especially in late-arriving birds, possibly combined with reduced sizes of intestine and liver during refuelling, had severe fitness consequences for adult survival and recruitment of young in 2000-2002. From 1997 to 2002 known survivors in Delaware Bay were heavier at initial capture than birds never seen again, annual survival of adults decreased by 37% between May 2000 and May 2001, and the number of second-year birds in wintering flocks declined by 47%. Population size in Tierra del Fuego declined alarmingly from 51,000 to 27,000 in 2000-2002, seriously threatening the viability of this subspecies. Demographic modelling predicts imminent endangerment and an increased risk of extinction of the subspecies without urgent risk-averse management.  相似文献

Unheated spores of nonproteolytic Clostridium botulinum were able to lead to growth in sterile deoxygenated turnip, spring green, helda bean, broccoli, or potato juice, although the probability of growth was low and the time to growth was longer than the time to growth in culture media. With all five vegetable juices tested, the probability of growth increased when spores were inoculated into the juice and then heated for 2 min in a water bath at 80°C. The probability of growth was greater in bean or broccoli juice than in culture media following 10 min of heat treatment in these media. Growth was prevented by heat treatment of spores in vegetable juices or culture media at 80°C for 100 min. We show for the first time that adding heat-treated vegetable juice to culture media can increase the number of heat-damaged spores of C. botulinum that can lead to colony formation.  相似文献

The hydrogenase (EC 1.2.2.1) of Desulfovibrio gigas is a complex enzyme containing one nickel center, one [3Fe-4S] and two [4Fe-4S] clusters. Redox intermediates of this enzyme were generated under hydrogen (the natural substrate) using a redox-titration technique and were studied by EPR and M?ssbauer spectroscopy. In the oxidized states, the two [4Fe-4S]2+ clusters exhibit a broad quadrupole doublet with parameters (apparent delta EQ = 1.10 mm/s and delta = 0.35 mm/s) typical for this type of cluster. Upon reduction, the two [4Fe-4S]1+ clusters are spectroscopically distinguishable, allowing the determination of their midpoint redox potentials. The cluster with higher midpoint potential (-290 +/- 20 mV) was labeled Fe-S center I and the other with lower potential (-340 +/- 20 mV), Fe-S center II. Both reduced clusters show atypical magnetic hyperfine coupling constants, suggesting structural differences from the clusters of bacterial ferredoxins. Also, an unusually broad EPR signal, labeled Fe-S signal B', extending from approximately 150 to approximately 450 mT was observed concomitantly with the reduction of the [4Fe-4S] clusters. The following two EPR signals observed at the weak-field region were tentatively attributed to the reduced [3Fe-4S] cluster: (i) a signal with crossover point at g approximately 12, labeled the g = 12 signal, and (ii) a broad signal at the very weak-field region (approximately 3 mT), labeled the Fe-S signal B. The midpoint redox potential associated with the appearance of the g = 12 signal was determined to be -70 +/- 10 mV. At potentials below -250 mV, the g = 12 signal began to decrease in intensity, and simultaneously, the Fe-S signal B appeared. The transformation of the g = 12 signal into the Fe-S signal B was found to parallel the reduction of the two [4Fe-4S] clusters indicating that the [3Fe-4S]o cluster is sensitive to the redox state of the [4Fe-4S] clusters. Detailed redox profiles for the previously reported Ni-signal C and the g = 2.21 signal were obtained in this study, and evidence was found to indicate that these two signals represent two different oxidation states of the enzyme. Finally, the mechanistic implications of our results are discussed.  相似文献

An increased rate of lipid synthesis in cancerous tissues has long been recognised as an important aspect of the rewired metabolism of transformed cells. However, the contribution of lipids to cellular transformation, tumour development and tumour progression, as well as their potential role in facilitating the spread of cancerous cells to secondary sites, are not yet fully understood. In this article, we review the recent findings that support the importance of lipid synthesis and metabolism in tumorigenesis. Specifically, we explore the role of aberrant lipid biosynthesis in cancer cell migration and invasion, and in the induction of tumour angiogenesis. These processes are crucial for the dissemination of tumour cells and formation of metastases, which constitute the main cause of cancer mortality.  相似文献
