Dependence of behavioral responses form during shake-off conditional reflex on laterality of the neocortex

Pattern of discharges and interhemispheric interaction of parietal neurons (somatosensory presentation of the ear) were compared in interstimulus intervals before active shake-off and passive freezing reactions at stimuli after elaboration defensive avoid reflex. Before passive reactions in contrast to active right-side influences on left-side neurons were reinforced with delay to 100 ms, it result in appearence of asymmetry in interhemispheric interaction with right-side dominance. Before passive reactions intensity of delta-frequencies was increased in impulsation of single neurons and in interaction of cells pair. On this evidence the pattern of interhemispheric asymmetry before stimuli can be one of a number of factors, determinant active or passive form of behavioral reactions and also reflective of activation level of neocortex.     

Peculiarities of functional activity of circulating phagocytes in patients with different forms of drug-resistant pulmonary tuberculosis

Functional activity of circulating phagocytes (macrophages and neutrophils — Ns) was studied in 30 patients with infiltrative and 30 patients with fibro-cavernous pulmonary tuberculosis (IPT and FCPT, respectively), characterized by similar biological properties of mycobacterium tuberculosis (MTB), dissemination of the process, and manifestations of intoxication. Differences of the functional activity of both types of cells depending on the PT form were found: a more significant increase in the oxygen-depending activity in FCPT while bactericide potential estimated with the zymosan induced NST-test was more pronounced in IPT patients. These data correlate with the blood levels of neopterin and elastase, the markers of the mononuclear and neutrophil activity, respectively. Involvement of adenosine deaminase (ADA) and neopterin in realization of intracellular oxygen-dependent processes was demonstrated. Results of the multivariate analysis of the whole set of the studied phagocyte parameters, reflecting their different roles in this pathological process demonstrated a prevailing role of mononuclears in newly diagnosed IPT and neutrophils in the chronic progressive process.     

Volatiles from four Astragalus species: phenological changes and their chemotaxonomical application

This paper shows the changes of the volatile compounds from four Astragalus species at three phenological stages: leaf development, flowering and fructification, which might be connected with the plant defense. After GC/MS analyses of Astragalus glycyphyllos L., A. hamosus L., A. cicer L. and A. spruneri Boiss., different groups of volatile compounds were found: hydrocarbons, alcohols, aldehydes and ketones, esters, terpenes, chlorinated compounds, etc. Identified volatiles were used for a cluster analysis in order to make chemotaxonomic conclusions for these evolutionary different species.     

Activity of rabbit's neocortical and hippocampal neurons in orienting-exploratory behavior and freezing

By plotting autocorrelation histograms, the character of impulsation of neurons in bilateral derivations from cortical visual and somatosensory areas and hippocampal field CA1 was studied in rabbits during free behavior under the influences of emotional stimuli. During active orienting-exploratory reaction of rabbits, most cortical and hippocampal neurons manifested bursting discharges and theta-frequency oscillation (predominantly 4-5 Hz in cortex and 4-5, 6-7 in hyppocampus). During freezing, as compared with active locomotor reaction, the number of neurons with equiprobabilistic discharges increased; in impulsation of neurons with periodicity, the intensity of delta-frequency oscillation increased (predominantly 2-4 Hz) but theta-frequency oscillation decreased. The number of neurons with delta-frequency oscillation during freezing was greater then in calmly sitting rabbits.     

Overproduction of single-chain antibodies against human IFN-alpha2B in Escherichia coli and their isolation in biologically active form

The gene encoding mouse single chain antibody (ScFv) against human interferon alpha2b (IFN-alpha2b) was cloned into the plasmid vector under the control of promoter from phage T7 and the recombinant protein was expressed in Escherichia coli as inclusion bodies. After the isolation of inclusion bodies the desired protein containing affinity tail "6His tag" was solubilized and purified under denaturing conditions by immobilized-metal affinity chromatography. The soluble and purified ScFv was obtained by "on column" refolding and the recovery of biological activity were demonstrated. The higher levels of ScFv production for intracellular expression system in comparison with ScFv obtained by secretion were shown. The advantages of described refolding method are simplicity and high efficacy, moreover, refolding using a chromatographic process represents the manufacturable approach because it is easily automated using commercially available materials and preparative chromatography systems and also can be combined with simultaneous purification.     

Influence of emotional stimuli on behavior and respiration of rabbits with various locomotor activity in the "open field"

Different behavioral reactivity of rabbit groups differentiated by locomotor activity in the "open field" was revealed during exposure to emotional stimuli (rustle, loud sound, pressuring on back of the neck, vibroacoustic tactile stimulation of an ear). In passive rabbits, the active locomotor reactions were induced harder and freezing was obtained easier than in active animals. During exposure to sound stimuli, passive rabbits increased their locomotion more rarely than active animals, pressing on back of the neck produced longer freezing, a threshold of defensive ear shaking in response to a vibroacoustic stimulus in passive animals was highest. Training to mild immobilization increased the threshold of defensive responses in active rabbits and animals of the intermediate type. Changes in respiratory parameters were correlated with behavioral reactions to emotional stimuli. The duration of exhalation and respiratory cycle increased during freezing and increased during enhanced locomotion. The duration of inhalation decreased in response to emotional stimuli irrespective of a behavioral reaction. The respiratory reactions to emotional stimuli differed in rabbits of different groups. The respiratory rate more frequently changed in passive rabbits than in animals of other groups. Passive animals reacted mainly by exhalation, active rabbits and animals from the intermediate group predominantly responded by inhalation.     

Loss and recovery of androgen receptor protein expression in the adult rat testis following androgen withdrawal by ethane dimethanesulfonate

Androgens are especially important for the maintenance of spermatogenesis in adulthood and the experimental withdrawal of testosterone (T) production by ethane dimenthanesulfonate (EDS) is a valuable tool for studying androgen-dependent events of spermatogenesis. The aim of the present study was to investigate the specific changes in immunoexpression of androgen receptor (AR) in the testis in relation to degeneration and regeneration of Leydig cell (LC) population and seminiferous epithelium. Immunohistochemistry for AR and 3beta-hydroxysteroid dehydrogenase (3beta-HSD) as well as TUNEL assay for apoptosis were performed on testicular sections of control and EDS-treated rats. Serum LH and T levels were measured by RIA. Our results revealed a total loss of AR immunoexpression from the nuclei of Sertoli (SCs), LCs and peritubular cells during the first week after EDS administration and that coincided with severe drop in T levels. Two weeks after EDS administration, the AR expression was recovered in these cells but normal stage-specificity in SCs was replaced by uniform intensity of AR immunostaining at all the stages of the spermatogenic cycle. The stage-specific pattern of androgen expression in SCs with a maximum at stages VII-VIII appeared 5 weeks after treatment. LC immunoreactivity for 3beta-HSD at different time points after EDS administration correlated with values of T concentration. The maximal germ cell apoptosis on day 7 was followed by total loss of elongated spermatids 2 weeks after EDS treatment. Regeneration of seminiferous epithelium 3 weeks after EDS administration and onwards occurred in tandem with the development of new LC population indicated by the appearance of 3beta-HSD-positive cells and gradual increase in T production. The specific changes in AR after EDS including their loss and recovery in Sertoli cells paralleled with degenerative and regenerative events in Leydig and germ cell populations, confirming close functional relationship between Sertoli, Leydig and germ cells.
