The experiment assessed the variability of in seven clones of willow plants of high biomass production (Salix smithiana S-218, Salix smithiana S-150, Salix viminalis S-519, Salix alba S-464, Salix ’Pyramidalis’ S-141, Salix dasyclados S-406, Salix rubens S-391). They were planted in a pots for three vegetation periods in three soils differing in the total content of risk elements. Comparing the calculated relative decrease of total metal contents in soils, the phytoextraction potential of willows was obtained for cadmium (Cd) and zinc (Zn), moderately contaminated Cambisol and uncontaminated Chernozem, where aboveground biomass removed about 30% Cd and 5% Zn of the total element content, respectively. The clones showed variability in removing Cd and Zn, depending on soil type and contamination level: S. smithiana (S-150) and S. rubens (S-391) demonstrated the highest phytoextraction effect for Cd and Zn. For lead (Pb) and arsenic (As), the ability to accumulate the aboveground biomass of willows was found to be negligible in both soils. The results confirmed that willow plants show promising results for several elements, mainly for mobile ones like cadmium and zinc in moderate levels of contamination. The differences in accumulation among the clones seemed to be affected more by the properties of clones, not by the soil element concentrations or soil properties. However, confirmation and verification of the results in field conditions as well as more detailed investigation of the mechanisms of cadmium uptake in rhizosphere of willow plants will be determined by further research.  相似文献

Cryptococcus neoformans was grown in 96-well microtiter plates sealed by foil which is less than 0.01 % permeable to oxygen. On day 14 of the cultivation, we observed peculiar clusters of small droplike daughter cells arranged around ≤4 % of mother cells. The fact that most of the other cells had died indicates that few cells had been able to survive hypoxic conditions and escape the cell-cycle arrest. However, their daughters were unable to separate from them and to continue their proliferation under such conditions.  相似文献

There has been a considerable expansion of Moliniacaerulea after the cessation of cutting management of Nardus stricta sub-alpine grasslands in the Giant Mts. (Krkonoše/Karkonosze/Riesengebirge, the Czech Republic) in the last 50 years. The aim of this study was to investigate whether the reestablishment of traditional management (one cut per year in late July) could reverse Molinia encroachment and help to restore the original Nardus grassland.  相似文献

Question: Is there any effect of cutting frequency and liming on P and K availability in the soil, biomass production and plant species composition after cessation of fertilizer application? Location: Eifel Mountains, SW Germany. Methods: The long‐term Grassland Extensification and Nutrient Depletion Experiment was established on a fertilized and mown pasture (Lolio‐Cynosuretum) in 1993. Treatments were: (1) two cuts per year without liming, (2) two cuts with liming, (3) four cuts without liming, (4) four cuts with liming and (5) continued intensive mowing as the control. Results: From 1993 to 2006, the plant available P concentration in the soil decreased substantially, whereas K concentration decreased only slightly. Biomass production decreased from 7 to 5 t DM ha?1. These trends were affected by cessation of NPK fertilizer application but not by cutting frequency or liming. In 2007, substantial differences in species composition between the control and the two‐cut and four‐cut treatments were recorded, whereas liming had no effect. Higher species richness was recorded in cut treatments compared to the control, but no effects of cutting frequency or liming were observed. Ellenberg indicator values indicated that soil nutrients influenced changes in species composition only marginally. Conclusions: The decrease in productivity and available soil P and K in favor of species richness was not achieved to any greater extent by four cuts than by two cuts, or by lime application. Although species richness slightly increased, we conclude that the restoration of low productive grasslands cannot be achieved by cutting management.  相似文献

The influence of a subconvulsant dose of kainic acid (KA) on the activity of neurons was studied in the sensorimotor cortical area of urethane-anesthetized rats. A total of 41 neurons was recorded, 38 of these in layer V (probably pyramidal cells). The activity of 18 neurons was recorded before as well as more than 30 min after KA administration (6 mg/kg i.p.). Nine out of these 18 neurons increased their firing rate significantly even 20 min after KA injection, whereas the remaining neurons did not change their activity. Altogether, the increase in the firing rate was significant. KA was found to enhance markedly the firing rate of a part of cortical neurons at very early stages of its action.  相似文献

The N-feruloylserotonin containing fraction was isolated from seeds of Leuzea carthamoides (Willd.) DC by solvent extraction followed by column chromatography on silica gel or on Sephadex LH-20. Nuclear magnetic resonance spectroscopic analysis of the isolated fraction showed the presence of four structurally related compounds. These compounds were identified as four isomers of N-feruloylserotonin: N-(Z)-feruloylserotonin, N-(Z)-isoferuloylserotonin, N-(E)-feruloylserotonin and N-(E)-isoferuloylserotonin. They were analyzed by HPLC on Separon SGX C18, Separon SGX and Separon SGX phenyl, using various mobile phases. Separon SGX phenyl phase was found the most efficient for a rapid analysis and for the final separation of the N-feruloylserotonin isomers.  相似文献

This study evaluates the potential of flow cytometry for chromosome sorting in durum wheat (Triticum turgidum Desf. var. durum, 2n = 4x = 28). Histograms of fluorescence intensity (flow karyotypes) obtained after the analysis of DAPI-stained chromosomes consisted of three peaks. Of these, one represented chromosome 3B, a small peak corresponded to chromosomes 1A and 6A, and a large peak represented the remaining 11 chromosomes. Chromosomes sorted onto microscope slides were identified after fluorescence in situ hybridization (FISH) with probes for GAA microsatellite, pSc119.2, and Afa repeats. Genomic distribution of these sequences was determined for the first time in durum wheat and a molecular karyotype has been developed for this crop. Flow karyotyping in double-ditelosomic lines of durum wheat revealed that the lines facilitated sorting of any arm of the wheat A- and B-genome chromosomes. Compared to hexaploid wheat, flow karyotype of durum wheat is less complex. This property results in better discrimination of telosomes and high purities in sorted fractions, ranging from 90 to 98%. We have demonstrated that large insert libraries can be created from DNA purified using flow cytometry. This study considerably expands the potential of flow cytogenetics for use in wheat genomics and opens the possibility of sequencing the genome of this important crop one chromosome arm at a time.  相似文献

The role of T lymphocyte subpopulations in the protection against intraperitoneal (i.p.) and peroral Encephalitozoon cuniculi infections was compared in adoptive-transfer experiments using severe combined immunodeficient mice. Whereas CD8+ T cell-depleted, but not CD4+ T cell-depleted, BALB/c splenocytes failed to protect the mice against i.p. infection, only SCID mice reconstituted with both CD4+ T lymphocyte- and CD8+ T lymphocyte-depleted splenocytes succumbed to peroral infection. The results indicate that whereas CD8+ T cells are critical for the protection against an i.p. E. cuniculi infection, both CD4+ and CD8+ T lymphocyte subpopulations play a substantive protective role in a peroral infection, i.e., natural route of infection.  相似文献

NKR-P1 protein is an important activating receptor at the surface of the rat natural killer cells. GlcNAc and chitooligomers were identified as strong activation ligands in vitro and in vivo. Their clustering brings about increase of their affinity to the NKR-P1 by 3–6 orders. Here we describe novel methodology for preparation of neoglycoproteins based on BSA carying the chitooligomers (n = 2–5). Further on we developed novel methodology of the coupling of glycosylamines via aromatic-SCN activated linker both to protein or synthetic cores. Inhibition studies of chitooligomer glycoconjugates with the NKR-P1 receptor show that our neoglycoproteins are very strong ligands with high binding affinity (–log IC50 = 13–15). In analogy with our previous observations with GlcNAc clustered on protein or PAMAM backbones the synthetic chitooligomer clusters should provide considerably better ligands in the in vivo antitumor treatment.  相似文献

Activity was recorded intracellularly from the bodies of 87 reticulospinal neurones in the cat's gigantocellular nucleus, whose axons had a conduction velocity of 18-148 m.s-1. Slow-conducting neurones (18-45 m.s-1, 23%) were characterized by a wider action potential, higher input resistance (3.8-7.0 M omega) and a lower rheobase (1.0-1.7 nA). They were also very sensitive to changes in membrane polarity and generated regular rhythmic activity. Fast-conducting neurons (45-148 m.s-1) were characterized by a short action potential, low input resistance (0.7-2.9 M omega) and a higher rheobase (1.5-5.2 nA). When depolarizing current pulses were applied, they generated responses with action potentials with a high frequency, especially in the initial phase of depolarization, but their thresholds for the initiation of activity and steady firing were higher than in the case of slow neurones. Slow reticulospinal neurones always responded to stimulation of the spinal funiculi (mainly the dorsal funiculus) by a characteristic large postsynaptic potential on which large numbers of spike potentials were superimposed and which did not occur in fast neurones. The differences observed in membrane properties and in the character of generation of action potentials draw attention to the phasic character of fast, and the tonic character of slow, reticulospinal neurones.  相似文献
