Nitric oxide (NO) is a ubiquitous, cell-permeable intercellular messenger. The current concept assumes that NO diffuses freely through the plasma membrane into the cytoplasm of a target cell, where it activates its cytosolic receptor enzyme, soluble guanylyl cyclase (sGC). Recent evidence, however, suggests that cellular membranes are not only the predominant site of calcium-dependent NO synthesis, but also the site of its distribution and binding. Here we extend this concept to NO signalling to show that active sGC is partially associated with the plasma membrane in a state of enhanced NO sensitivity. After cellular activation, sGC further translocates to the membrane fraction in human platelets and associates with the NO-synthase-containing caveolar fraction in rat lung endothelial cells, in a manner that is dependent on the concentration of intracellular calcium. Our data suggest that the entire NO signalling pathway is more spatially confined than previously assumed and that sGC dynamically translocates to the plasma membrane, where it is sensitized to NO.  相似文献

Monomeric forms of E. coli glyceraldehyde-3-phosphate dehydrogenase have been prepared using two different experimental approaches: (1) covalent immobilization of a tetramer on a solid support via a single subunit with subsequent dissociation of non-covalently bound subunits in the presence of urea, and (2) entrapment of monomeric species into reversed micelles of Aerosol OT in octane. Isolated monomers were shown to be catalytically active, exhibiting K M values close to the parameters characteristic of the tetrameric forms. Like tetramers, isolated monomers did not use NADP7 as a coenzyme.  相似文献

The first η2-olefinic monocarbon metallacarbone closo-2-(Ph3P)-1-N,2-[μ-(η2-CH2CH=Ch2)]-1-N-(σ-CH2CH=CH2)-2,1- RhCB10H10 has been prepared by the reaction of the dimeric anion {[Ph3PRhB10H10CNH2]2-μ-H}[PPN]+ with allyl bromide and characterized by a combination of spectroscopic methods and a single-crystal X-ray diffraction study. The variable temperature 1H and 13C NMR studies revealed the fluxional behavior of the η2-olefinic complex in CD2Cl2 solution which is associated with the allyl side-chain exchange process.  相似文献

Trichofumins A-D were isolated from cultures of Trichoderma sp. HKI 0276 as new 11 and 13mer peptaibols. Similar to 15mer peptaibols they promote morphogenesis of the fungus Phoma destructiva and cause hypothermia in mice as a characteristic of neuroleptic activity. Membrane measurements using a synthetic BLM model showed that A, B, C and D increased membrane permeability for cations in a similar manner as was shown for larger peptaibols but with comparably lower efficiency.  相似文献

The present work reports on the investigation of a mixed binary chiral stationary phase (CSP) prepared by simultaneous attachment of permethylated‐β‐cyclodextrin ( D selector) and resorcinarene with pendant l ‐ or d ‐valine diamide groups ( L′ and D′ selectors, respectively) to a polysiloxane matrix via platinum‐catalyzed hydrosilylation. The gas‐chromatographic investigation of a number of racemates on the four different CSPs ( D, D′, DD′, and DL′ ) showed that the enantioselectivity of the individual chiral selectors was retained in the mixed binary CSPs. As a consequence, hydrocarbons, underivatized alcohols, ketones, and almost all proteinogenic amino acid derivatives could be separated simultaneously on each of the mixed CSPs. Matched and mismatched cases of enantioseparation on the mixed binary CSPs were observed but turned out to be of minor importance for enantiomeric separation. In general, more racemates were separated with α ≥ 1.02 on the mixed phases as compared to the single phases. In order to analyze the influence of the presence of the diamide selector on the enantioselectivity of the cyclodextrin selector, a mixed ternary CSP containing the selector D and a racemic mixture of the selectors D′ and L′ [ D ( D′L′ )] was prepared and investigated. A pollen record from the Taishizhuang site (40°21.5′N, 115°49.5′E) located in the transitional forest-steppe zone near the present-day limit of the summer monsoon is used to reconstruct vegetation and climate. Quantitative biome reconstruction suggests that between ca. 5700 and 4400 cal. years B.P. temperate deciduous forest dominated the vegetation cover around the Taishizhuang site. After that time the landscape became more open and the scores of the steppe biome were always higher than those of the temperate deciduous forest except for two oscillations dated to ca. 4000 cal. years B.P. and ca. 3500 cal. years B.P. However, ca. 3400-2100 cal. years B.P. the common vegetation became steppe and the landscape was more open in comparison with the previous time interval. The results of the pollen-based precipitation reconstruction suggest that annual precipitation was ca. 550-750 mm (ca. 100-300 mm higher than present) during the mid-Holocene ‘forest phase’, and ca. 450-650 mm during the following ‘forest-steppe phase’. From ca. 3400 cal. years B.P. during the ‘steppe phase’ annual precipitation was similar to modern values (ca. 300-500 mm). Archaeological records from 100 sites prove the habitation of northeastern China during the prehistoric and early historic periods from ca. 8200 cal. years B.P., but do not provide evidence of the use of wood resources intensive enough to influence the regional vegetation development and to leave traces in the pollen assemblages. Both archaeological and palaeoenvironmental data support the conclusion that changes in pollen composition in northeastern China between 5700 and 2100 cal. years B.P. reflect natural variations in precipitation and not major deforestation caused by humans.  相似文献
