In a previous communication we described the preparation of the alpha/beta-chain complex of HLA-D membrane antigens from a homozygous lymphoblastoid B cell line, H2LCL (HLA-A3,3; B7,7; Dw2,2; DR2,2; MT1,1; DC1,1; MB1,1). In this paper we present the separation of the alpha- and beta-chains in quantitative scale using hydroxylapatite chromatography with a pH 7.15 phosphate buffer, containing sodium dodecyl sulfate. An alternative isolation procedure is electrophoresis, taking advantage of the different isoelectric points of the alpha- and beta-chains. The relative molecular masses, the isoelectric points and the N-terminal sequences are discussed and compared with the results of other investigators. Remarkable is the polymorphism of both the chains, especially the beta-chains. The importance of the previously described three step purification procedure for the preparation of these antigens for sequence studies has been pointed out.  相似文献

Bovine serum albumin is generally employed as a substrate depot for the delivery of acyl units to lipid metabolizing enzymes in vitro. Here we test the possibility that albumin alters the availability of substrate to mitochondrial carnitine palmitoyltransferase I and thereby alters its apparent kinetics. Binding competition with palmitoyl-CoA indicates that albumin has 5-6 high affinity sites which avidly bind the substrate, while isolated mitochondria compete favorably for substrate only as the albumin sites become saturated. In contrast to albumin, artificial phospholipid vesicles bind palmitoyl-CoA uniformly. Palmitoyl-CoA distribution between vesicles and mitochondrial membranes appears simply to be a function of the relative size of the two lipid compartments. Both albumin and artificial vesicles reduce the effective concentration of substrate available to the enzyme and in this way reduce apparent affinity. Direct measurement of mitochondrially bound substrate removes this effect and brings the results into agreement with an affinity constant of 6-7 nmol/mg. Changes in gross mitochondrial structure, as indicated by decreased optical density and increased nonpelleting protein, do not begin occurring until levels of mitochondrially bound palmitoyl-CoA are 15 times greater than this. The highly sigmoidal activity profile of carnitine palmitoyltransferase with respect to palmitoyl-CoA (apparent Hill coefficient = 3.0 +/- 0.3) is lost when vesicles are substituted for albumin, suggesting that albumin binding sites contribute to the sigmoidal kinetics in the range of palmitoyl-CoA studied.  相似文献

Into a hole drilled in a block of dry ice, a metal microtome object disk is placed to cool. A drop of water is placed on the disk, and the specimen to be cut is fixed in place. By setting the dry ice in a well-insulated box, the specimen is thoroughly frozen. The disk is then clamped in the microtome, and chips of dry ice are wedged between the metal disk and the object clamp of the microtome. Zusammenfassung Die Anwendung der eben besprochenen, voneinander völlig verschiedenen Methoden zur Aufklärung der Struktur der zytoplasmatischen Membran führte zu dem Schluß, daß sowohl die Zellmembran selbst als auch die intrazellulären Membranen aller Zellen, seien es Pflanzenzellen, tierische Zellen oder Mikroorganismen, einen einheitlichen Bauplan aufweisen. In Abb. 13 werden in einer schematischen Zeichnung die Beziehungen zwischen den einzelnen Membranabschnitten in der Zelle illustriert. Es ligen heute Hinweise für einen kontinuierlichen Übergang zwischen der äußeren Zellmembran und dem Membransystem des endoplasmatischen Retikulums vor. Es scheint, daß der Zellkern über das kanalartige System des endoplasmatischen Retikulums direkt mit dem Außenmedium in Verbindung stehen kann. Die benutzten Methoden können jedoch — abgesehen von der elektrischen Methode — ihrer Natur nach keine Information über die Funktion der Membran liefern. Es wird die Aufgabe der Zukunft sein, die Erkenntnisse über die filmartige Natur der zytoplasmatischen Membran zusammen mit den Kenntnissen über den Stofftransport in einem umfassenden Modell darzustellen.  相似文献

The complex of alpha and beta chains of HLA-D membrane antigens has been isolated from a lymphoblastoid homozygous B cell line, H2LCL (HLA-A3,3; B7,7; Dw2,2; DR2,2; MT1,1; DC1,1; MB1,1), by an exclusively chemical two-step procedure and characterized by electrophoresis as well as isoelectric focusing in polyacrylamide gel. Cells were gained using long term cultivation in large scale, the crude membrane by differential centrifugation. The proteins of the crude membrane were then solubilized in NP-40, pH 5.0. The first purification step for HLA-D antigens consisted in an ion-exchange chromatography on carboxymethyl cellulose using the solubilization buffer. By this procedure the complex of proteins with relative molecular masses of Mr approximately 34 000 and Mr approximately 29 000 was in a high percentage not bound to the carboxymethyl cellulose. The bound fraction contained the HLA-A, -B and -C antigens and a component with Mr approximately 31 000 corresponding to the well known Ii-fraction. The bound proteins could be recovered from the column by a sodium chloride gradient. The proteins not bound to the carboxymethyl cellulose were precipitated with acetone, dissolved, dialysed against SDS buffer, pH 7.2 and then submitted to the second purification step, the Sephacryl S-300 chromatography. By this procedure the corresponding complex could be further separated from higher and lower molecular proteins. The complex was used as the starting material for the separation of alpha and beta chains. Amino-acid sequences established of the isolated chains have already been communicated.  相似文献

Differentiated epithelial cells are an important source of infectious EBV virions in human saliva, and latent Epstein-Barr virus (EBV) infection is strongly associated with the epithelial cell tumor, nasopharyngeal carcinoma (NPC). However, it has been difficult to model how EBV contributes to NPC, since EBV has not been shown to enhance proliferation of epithelial cells in monolayer culture in vitro and is not stably maintained in epithelial cells without antibiotic selection. In addition, although there are two major types of EBV (type 1 (T1) and type 2 (T2)), it is currently unknown whether T1 and T2 EBV behave differently in epithelial cells. Here we inserted a G418 resistance gene into the T2 EBV strain, AG876, allowing us to compare the phenotypes of T1 Akata virus versus T2 AG876 virus in a telomerase-immortalized normal oral keratinocyte cell line (NOKs) using a variety of different methods, including RNA-seq analysis, proliferation assays, immunoblot analyses, and air-liquid interface culture. We show that both T1 Akata virus infection and T2 AG876 virus infection of NOKs induce cellular proliferation, and inhibit spontaneous differentiation, in comparison to the uninfected cells when cells are grown without supplemental growth factors in monolayer culture. T1 EBV and T2 EBV also have a similar ability to induce epithelial-to-mesenchymal (EMT) transition and activate canonical and non-canonical NF-κB signaling in infected NOKs. In contrast to our recent results in EBV-infected lymphoblastoid cells (in which T2 EBV infection is much more lytic than T1 EBV infection), we find that NOKs infected with T1 and T2 EBV respond similarly to lytic inducing agents such as TPA treatment or differentiation. These results suggest that T1 and T2 EBV have similar phenotypes in infected epithelial cells, with both EBV types enhancing cellular proliferation and inhibiting differentiation when growth factors are limiting.  相似文献

Mean activity coefficients of different potassium salts KX (X = F-, Cl-, Br-, I-, NO3-, SCN-) have been measured in concentrated isoionic bovine serum albumin (BSA) solutions, by use of the EMF method with ion-exchange membrane electrodes. These solutions may be regarded as simple model systems for the cytoplasm of living cells as far as the influence of the macromolecular component on the activity coefficients of the salts is concerned. Two series of measurements have been carried out: (a) varying the amount of salt from 0.01 to 0.5 molal and maintaining the BSA concentration constant at 20 wt% and (b) varying the protein concentration up to 25 wt% and keeping the salt concentration constant at 0.1 molal. For all salts studied, the mean activity coefficients in the protein-salt solutions increase as the salt concentration rises, when the protein concentration is maintained constant. In the series of measurements (b) the activity coefficients of all salts change linearly with the protein concentration. Marked qualitative differences, however, were observed depending on the anion species, which could be interpreted in terms of specific ion binding of X- to the protein molecule. By taking into account BSA-bound 'non-solvent' water, the results were analyzed in terms of numbers of anions bound per BSA molecule. Comparison with the results of Scatchard, obtained at low protein concentrations, showed only a very small electrostatic effect of the BSA-(X-)v polyions on the activity coefficient of the salts at higher protein and salt concentrations.  相似文献

The author described a case of this disease connected with a growing blindness and a progressive renal disfunction in a male black lemur at Tierpark Berlin.
