Sorghum [Sorghum bicolor (L.) Moench] is a multipurpose grass cultivated in drylands due to its adaptation to drought. However the characteristics of sorghum-associated bacteria are not known in the Brazilian drylands. The aim of this study was to isolate and evaluate the plant growth promotion potential bacteria from field-grown sorghum under two irrigation and manure application levels in a Brazilian semi-arid reagion. Sorghum was irrigated with 3 or 1 mm day?1 and fertilized or not with liquid goat manure. Bacteria were obtained from surface-disinfected roots applying two nitrogen-free semi-solid media. The bacteria were evaluated for the presence of nifH gene, 16S rRNA sequences, calcium-phosphate solubilization, production of auxins and siderophores and for sorghum growth promotion. We obtained 20 out of 24 positive bacteria for nifH. The isolates were classified as in six different genera. All isolates produced auxins “in vitro”, six bacteria produced siderophores and three Enterobacteriaceae solubilized calcium-phosphate. At least ten bacteria resulted in the increased total N content in the sorghum shoots, comparable to fertilization with 50 mg N plant?1 week?1 and to inoculation with Azospirillum brasilense Ab-V5. Enterobacter sp. ESA 57 was the best sorghum plant-growth promoting bacteria isolated in this study.  相似文献

Some tropical Bignoniaceae form sporophytic apomictic polyploid complexes are similar to better studied temperate plants. Handroanthus ochraceus is a widely distributed Neotropical savanna tree with both monoembryonic/self-sterile, and polyembryonic/apomictic and self-fertile populations, but lacking chromosome number and morphological comparative studies. We tested if monoembryonic/non-apomictic and polyembryonic/apomictic populations differed in ploidy and morphological features, as a basis to understand evolution and biogeography of these plants. Chromosome number and embryo number per seed were investigated, and uni- and multivariate analyses of flower and leaf morphology were done for five populations of H. ochraceus. We found two pure monoembryonic diploid (2n?=?40), and one pure polyembryonic (62–94% of polyembryonic seeds) tetraploid population (2n?=?80). One of the diploid populations presented only one individual with 3.2% polyembryonic seeds and was considered a non-apomictic population. Another population showed predominantly polyembryonic (27–66% of polyembryonic seeds) tetraploid individuals, but one diploid individual with 2% of polyembryonic seeds, and was considered a mixed apomictic and non-apomictic population. Morphological analyses confirmed breeding system clusters, and that stigma width, as well as pollen grain area, was consistently larger in polyembryonic populations. Polyploid plants showed larger cells, as well as larger organs and other distinctive features, which will be useful to identify apomictic populations and to future taxonomic discussions. The species can be considered an agamic complex with apomixis associated with neopolyploidy. This trend is also found in other tropical sporophytic apomictics, contrasting with the usual reports linking diploidy or paleopolyploidy to this kind of apomictics.  相似文献

Some studies have reported the presence of leaf cork-warts in several families of plants; however, in Lauraceae, these structures have not been confirmed. Therefore, based on studies of leaf anatomy, we have herein documented the presence of leaf cork-warts in Mezilaurus species of Lauraceae. To accomplish this, we investigated fifteen species from all six genera of the Mezilaurus group, including Anaueria, Chlorocardium, Clinostemon, Mezilaurus, Sextonia, and Williamodendron. Analyses of leaf anatomy were performed through optical and scanning electron microscopy. Our results revealed cork-warts in six species of the genus Mezilaurus: M. crassiramea, M. navalium, M. revolutifolia, M. subcordata, M. synandra, and M. vanderwerffii. In the investigated species M. crassiramea and M. navalium, cork-warts originated from the stomatal complex, from epidermal cells covering oil cells located in mesophyll, and from epidermal cells around trichomes. Therefore, cork-warts could be considered an additional taxonomic character to delimit Mezilaurus species.  相似文献

The objective of this study was to determine cytotoxic activity, hemolytic activity, and to evaluate the ability of the essential oil from Cinnamodendron dinisii to induce DNA fragmentation of human lymphocytes. The essential oil was obtained by hydrodistillation. Cytotoxic activity was determined by the MTT method. Hemolytic activity was evaluated by spectrophotometric quantification of hemoglobin released by erythrocytes. Damage to lymphocyte DNA molecules was assessed by the Comet assay. The essential oil under study showed high cytotoxic activity on Vero cells (CC50 = 35.72 μg/mL) and induced hemolysis in both hematocrits, besides leading to the oxidation of hemoglobin released. The genotoxic activity of C. dinisii essential oil was also observed, which induced concentration‐dependent DNA fragmentation of human lymphocytes and, at 50 μL/mL, it was more active than the positive control. The essential oil from C. dinisii has a toxic action, suggesting a special attention in the application of this oil to health‐promoting activities; however, among its components, there are molecules with potential for future application in anticancer therapies.  相似文献

Breeding of competitive cultivars has long been fraught with difficulty owing to limited knowledge of the genetic basis of competitive ability. In this study, we examined the diversity of competitive ability in Asian rice and the genetic basis of this variation. Cultivated strains and wild perennial strains have higher competitive ability than wild annual strains. Quantitative trait locus (QTL) analysis of competitive ability for three weed species was conducted in the cross between cultivated and wild annual strains, and three QTLs for general competitive ability (GCA) were identified. GCA-QTLs conferred higher competitive ability by the cultivated rice alleles and were co-located with QTLs for plant architecture and root growth, detected in the same mapping population. Furthermore, a significant change in GCA was achieved by accumulation and epistatic interaction of three QTLs. Further studies on the genetic control of competitive ability would facilitate the breeding of competitive cultivars in rice.  相似文献

Our minds are continuously alternating between external attention (EA) and mind wandering (MW). An appropriate balance between EA and MW is important for promoting efficient perceptual processing, executive functioning, decision-making, auto-biographical memory, and creativity. There is evidence that EA processes are associated with increased activity in high-frequency EEG bands (e.g., SMR), contrasting with the dominance of low-frequency bands during MW (e.g., Theta). The aim of the present study was to test the effects of two distinct single session real-time EEG (rtEEG) protocols (SMR up-training/Theta down-training—SMR?Theta?; Theta up-training/SMR down-training—Theta?SMR?) on EA and MW processes. Thirty healthy volunteers were randomly assigned to one of two rtEEG training protocols (SMR?Theta?; Theta?SMR?). Before and after the rtEEG training, participants completed the attention network task (ANT) along with several MW measures. Both training protocols were effective in increasing SMR (SMR?Theta?) and theta (Theta?SMR?) amplitudes but not in decreasing the amplitude of down-trained bands. There were no significant effects of the rtEEG training in either EA or MW measures. However, there was a significant positive correlation between post-training SMR increases and the use of deliberate MW (rather than spontaneous) strategies. Additionally, for the Theta?SMR? protocol, increase in post-training Theta amplitude was significantly associated with a decreased efficiency in the orientation network.  相似文献

Water dynamics on germinating diaspores is normally used as a chronological marker in molecular biology and physiological studies focusing on germination. However, there is no protocol to analyze this process, which may undermine all results generated. In this context, we determined the sufficient sample size to study water dynamics on germinating diaspores by means of a modified maximum curvature method (MMCM). As a biological model, we used diaspores from agricultural species with different physiological qualities. These diaspores were put in contact with distilled water and were analyzed regarding mass, every hour, until embryo protrusion. Data collected in the last observation were used to measure the sufficient sample size. Here, we show that (i) MMCM is an adequate method to measure the variability of water dynamics on germinating diaspores; (ii) the b coefficient can be used to infer the homogeneity of water dynamics on diaspores of different species as a function of the increase in sample size; (iii) in general, to study this process in an easier, safer and standardized way, seven diaspores are needed and (iv) water dynamics on germinating diaspores estimated by weighted mass demonstrated to be a very stable process which depends more on the species than on the physiological quality. We also discussed why the sample size and water dynamics on germinating diaspores should be the first step for protocols on physiological and molecular aspects of the germination process.  相似文献
