AimsThe objective of this work was to evaluate the acute toxicity of long-circulating and pH-sensitive liposomes containing cisplatin (SpHL-CDDP), after their intraperitoneal administration in male and female mice.Main methodsAfter single administration of free CDDP (5,10,and 20 mg/kg) or SpHL-CDDP (7,12,30,45 and 80 mg/kg), the body weight was recorded and the LD50 was calculated. Blood samples were collected for biochemical and hematological analysis. Kidneys, liver, spleen and bone marrow were removed to histopathological examination.Key findingsMice treated with high doses of free CDDP showed a greater loss of body weight and more delayed recovery time than those treated with SpHL-CDDP. The LD50 values for SpHL-CDDP treatment for male and female mice groups were 2.7 and 3.2 fold higher, respectively, than that obtained for free CDDP. The red and white blood cells counts and quantification of hemoglobin and hematocrit presented no change upon administration of SpHL-CDDP treatment. Free CDDP treatment, however, did lead to an appearance of mild anemia and a reduction in total white blood cell counts. As regards nephrotoxicity, it was observed that free CDDP treatment caused pronounced alterations in the blood urea and creatinine levels of mice. In contrast, these parameters were slightly altered only after SpHL-CDDP treatment at a dose of 30 mg/kg. Microscopic analysis of kidneys from mice treated with SpHL-CDDP showed no morphological alteration. Concerning hepatotoxicity, no histopathological alteration was observed after both treatments.SignificanceThese findings reveal that SpHL-CDDP can eliminate CDDP-induced toxicity and is thus a promising candidate for intraperitoneal chemotherapy.  相似文献

The sludge produced in sewage treatment plants can contain toxic substances. Among these, the genotoxic substances are of great concern. The present paper aimed at evaluating the genotoxicity of treated sludge samples collected at four different sewage treatment plants (STP) located in the State of S?o Paulo, Brazil using the Trad-MN assay. Another objective of the study was to compare the responses of the Clone #4430 with the Tradescantia pallida. Sludge samples mixed with reference soil in concentrations of 10, 25 and 50% (v/v) were tested in experiments with 3 months exposure in the field. Negative and positive controls (arsenic trioxide) were also tested with both plants. In Clone #4430 two sludge samples induced genotoxicity while in T. pallida three were positive, although no clear dose-response were observed for both plants. Results with the negative and positive controls suggest that T. pallida presented similar results when compared to the Clone #4430. The protocol using plants chronically exposed to sludge mixed with soil seems to be a promising tool to assess the genotoxicity of sludge although time consuming.  相似文献

Caffeine-induced Ca2+ transients (CICTs) in rabbit nodose ganglion neurons (NGNs) are produced by two distinct mechanisms: release from intracellular stores via ryanodine receptors and Ca2+ influx across the plasma membrane, due to activation of an unknown receptor. In isolated rat NGNs, we used single-cell microfluorimetry to measure changes in intracellular Ca2+ and to test whether TRPV1 receptors underlie the Ca2+ influx pathway. Caffeine (10 mM) evoked CICTs in all NGNs tested (n = 47) averaging 365 ± 32 nM. CICTs were partially dependent upon a Ca2+ influx pathway that ranged between 33% and 98% of the total Ca2+ transient. Application of two selective TRPV1 antagonists significantly attenuated CICTs. The peak average amplitudes of CICTs in Ca2+-free Locke solution and Ca2+-free Locke solution with IRTX or with BCTC were not significantly different from one another (n = 5 and 7, respectively). These observations suggest that caffeine can induce Ca2+ influx by activating TRPV1 channels.  相似文献

A thermophilic spore-forming bacterium (strain AMP) was isolated from a thermophilic methanogenic bioreactor that was fed with cobalt-deprived synthetic medium containing methanol as substrate. 16S rRNA gene analysis revealed that strain AMP was closely related to the acetogenic bacterium Moorella thermoacetica DSM 521T (98.3% sequence similarity). DNA–DNA hybridization showed 75.2 ± 4.7% similarity to M. thermoacetica DSM 521T, suggesting that strain AMP is a M. thermoacetica strain. Strain AMP has a unique one-carbon metabolism compared to other Moorella species. In media without cobalt growth of strain AMP on methanol was only sustained in coculture with a hydrogen-consuming methanogen, while in media with cobalt it grew acetogenically in the absence of the methanogen. Addition of thiosulfate led to sulfide formation and less acetate formation. Growth of strain AMP with CO resulted in the formation of hydrogen as the main product, while other CO-utilizing Moorella strains produce acetate as product. Formate supported growth only in the presence of thiosulfate or in coculture with the methanogen. Strain AMP did not grow with H2/CO2, unlike M. thermoacetica (DSM 521T). The lack of growth with H2/CO2 likely is due to the absence of cytochrome b in strain AMP.  相似文献

Rhodolith beds are built by the aggregation of free living marine benthic coralline algae. Herein, we described phytobenthic communities associated with subtidal rhodolith beds in northeastern Brazil and tested the hypothesis that depth affects their structure. We compared macroalgal assemblages from depths of 10, 15 and 20 m. The genus Lithothamnion was dominant in these beds. Rhodolith density was similar at different depths, but volume decreases as depth increases. Sixty-seven species of fleshy algae were collected. The red algal order Ceramiales was dominant. A distinct community corresponds to each sampled depth. The shallower depth presented higher values for biomass, number of species, Shannon-Wiener diversity, and Pielou's evenness. When depth and water transparency increased, the number of species and the abundance of macroalgae decreased.  相似文献

Elatol was isolated as the major compound from the red alga Laurencia microcladia Kütz. collected in Southern Brazil. This is the first report of elatol in this species. We also investigated the herbivore behaviour of the black sea urchin Echinometra lucunter (Linnaeus, 1758) towards L. microcladia, Ulva fasciata Delile and Gracilaria domingensis (Kütz.) Sond. ex Dickie through live algal multiple-choice feeding assay, as well as artificial feeding assay. The sea urchins ate the crude algae L. microcladia and pellets containing the powdered algae, extract and all tested concentrations of elatol, suggesting that this seaweed and its main compound are palatable for E. lucunter.  相似文献

The Bignoniaceae possess sensitive bilobate stigmas that close after stimulation. We determined factors involved in stigma closure and reopening in four species of Bignoniaceae from the National Parks of Catimbau, Northeastern Brazil (Anemopaegma laeve, Arrabidaea limae, Jacaranda rugosa and Tabebuia impetiginosa). The study showed that any mechanical touch promoted the stigmatic closure. Only the deposition of viable con-specific pollen (self- and cross-pollen), however, maintained stigmas permanently closed. In Arrabidaea limae part of the stigmas reopened after self- and cross-pollination. After mechanical stimulation, deposition of pollen from other plant species, of dead con-specific pollen or of sand, the stigma lobes opened again after 38 to 68 min. The definitive closure may have a negative effect on the reproductive success of the involved plants, because the stigmas are permanently unavailable for pollen deposition. In self-incompatible species with mass-flowering blooming pattern, where pollinators promote high levels of geitonogamy, definitive stigma closure causes low fruit set.  相似文献

Bioremediation technologies are used in order to remove pollutants from the environment in a safe, economical and harmless way during the treatment of waste, especially with the use of techniques such as biodegradation. A lubricant and vegetable oil contaminated water sample was studied in order to evaluate the biodegradability of different types of oils, considering the relevance of the obtained data in the bioremediation procedures. The objective of this paper is to use respirometry technique as a biodegradation process data source, and then apply to the obtained data the experimental design of mathematical models to characterize and determinate how the different types of oils are capable of affecting the parameters in biodegradation kinetics. The kinetics was then evaluated through selected models with a reasonable fit to experimental data. The Bartha and Pramer respirometer is used as a method to accurately measure the CO2 formation in the organic compounds degradation by microorganisms. Therefore, the difference in biodegradation efficiency process is compared in the different groups of oils using mathematical models fitting the obtained data for the kinetics of biodegradation. The results demonstrated that used lubricant automotive oils are more susceptible to the biodegradation process, since their molecular structures had already been altered after use. In general, automotive lubricant oils shown better performance in biodegradation than vegetable oils. The models proposed for the obtained data in each of these assays demonstrated that vegetable oils biodegradation rate tends to decrease faster and end sooner than the automotive oils. Also, the modeling predicted that higher rates of biodegradation and total CO2 production are to be expected in automotive lubricant oils rather than vegetable oils.  相似文献
