This study evaluated the effects of flavone eupafolin (6-methoxy 5,7,3',4'-tetrahydroxyflavone), extracted from dry leaves of Eupatorium litoralle. Eupafolin (25-200microM) promoted inhibition of the respiratory rate in state 3, in the presence of glutamate or succinate. During succinate oxidation, it was found that only state 4 respiratory rate was stimulated approximately 30% by eupafolin (100microM) and ADP/O ratio and RCC were reduced with all doses. When glutamate was used as substrate, RCC was similarly reduced. Eupafolin caused a reduction of enzymatic activities between complexes I and III of the respiratory chain. Cytochrome c oxidase and ATPase activities were not affected. Using voltammetry cyclic analysis, eupafolin give rise to irreversible oxidation with an anodic peak potential at +0.08V (SHE). We also observed that eupafolin can undergo oxidation catalyzed by EDTA-Fe, promoting cytochrome c reduction in the presence of NADH, resulting in the production of the superoxide radical and hydrogen peroxide. All together, the results could explain the cytotoxic effects observed previously with the eupafolin.  相似文献

The electrochemical H+ gradient generated in tonoplast vesicles isolated from maize seeds was found to be able to drive the reversal of the catalytic cycle of both vacuolar H+-pumps (Façanha and de Meis, 1998). Here we describe the reversibility of the vacuolar V-type H+-ATPase (V-ATPase) even in the absence of the H+ gradient in a water-Me2SO co-solvent mixture, resulting in net synthesis of [γ-32P]ATP from [32P]Pi and ADP. The water-Me2SO (5 to 20 %) media promoted inhibition of both PPi hydrolysis and synthesis reactions whereas it slightly affected the ATP hydrolysis and clearly stimulated the ATP synthesis, which was unaffected by uncoupling agents (FCCP, Triton X-100 or NH4+). This effect of Me2SO on the ATP⇔32P exchange reaction seems to be related to a decrease of the apparent Km of the V-ATPase for Pi. The results are in accordance to the concept that the energetics of ATP synthesis catalysis depends on the solvation energies interacting in the enzyme microenvironment. In this study, we present the first data about putative source populations of the vagrant Subantarctic fur seal, Arctocephalus tropicalis, found on the Brazilian coast, through the comparison of their mitochondrial DNA control sequences to exclusive haplotypes from the main breeding colonies of the species. The results indicated that, despite the majority of the vagrant individuals are from Gough Island (the closest breeding site to the Brazilian coast), they also come from other reproductive colonies, such as Crozet Island, a distance around 16,500 km from the Brazilian coast. Furthermore, the molecular data identified three possible management units: (1) Gough, (2) Amsterdam, and (3) Marion, Macquarie and Crozet. This significant genetic subdivision must be taken into account in any future management plan for the species conservation, including rehabilitation and even reintroduction of vagrant fur seals. Oxygen is not only one of life's essential elements but also a source of protein damage, mutagenesis, and ageing. Many proteome adaptations have been proposed to tackle such stresses and we assessed them using comparative genomics in a phylogenetic context. First, we find that aerobiosis is a trait with important phylogenetic inertia but that oxygen content in proteins is not. Instead, oxygen content is close to the expected values given the nucleotide composition. Accordingly, we find no evidence of oxygen being a scarce resource for protein synthesis even among anaerobes. Second, we searched for counterselection of amino acids more prone to oxidation among aerobes. Only cysteine follows the expected trend, whereas tryptophan follows the inverse one. When analyzing composition in the context of protein structures and residue accessibility, we find that all oxidable residues are avoided at the surface of proteins. Yet, there is no difference between aerobes and anaerobes in this respect, and the effect might be explained by the hydrophobicity of these residues. Third, we revisited the hypothesis that atmospheric enrichment in molecular oxygen led to the development of the communication capabilities of eukaryotes. With a larger data set and adequate controls, we confirm the trend of longer oxygen-rich outer domains in transmembrane proteins of eukaryotes. Yet, we find no significant association between oxygen concentration in the environment and this trait within prokaryotes, suggesting that this difference is clade specific and independent of oxygen availability. We find that genes involved in cellular responses to oxygen are much more frequent among aerobes, and we suggest that they erase most expected differences in terms of proteome composition between organisms facing high and low oxygen concentrations.  相似文献

We compare physicians and laypeople within and across cultures, focusing on similarities and differences across samples, to determine whether cultural differences or lay-professional differences have a greater effect on explanatory models of the common cold. Data on explanatory models for the common cold were collected from physicians and laypeople in South Texas and Guadalajara, Mexico. Structured interview materials were developed on the basis of open-ended interviews with samples of lay informants at each locale. A structured questionnaire was used to collect information from each sample on causes, symptoms, and treatments for the common cold. Consensus analysis was used to estimate the cultural beliefs for each sample. Instead of systematic differences between samples based on nationality or level of professional training, all four samples largely shared a single-explanatory model of the common cold, with some differences on subthemes, such as the role of hot and cold forces in the etiology of the common cold. An evaluation of our findings indicates that, although there has been conjecture about whether cultural or lay-professional differences are of greater importance in understanding variation in explanatory models of disease and illness, systematic data collected on community and professional beliefs indicate that such differences may be a function of the specific illness. Further generalizations about lay-professional differences need to be based on detailed data for a variety of illnesses, to discern patterns that may be present. Finally, a systematic approach indicates that agreement across individual explanatory models is sufficient to allow for a community-level explanatory model of the common cold.  相似文献

WNK protein kinases form a kinase subfamily expressed in multi-cellular organisms and the human genome encodes four distinct WNK genes. Human WNK2 has been recently identified as a cell growth regulator that modulates activation of the ERK1/2 protein kinase and is epigenetically silenced in gliomas. Here we provide mechanistic insight into how WNK2 affects ERK activation. We found that WNK2 depletion decreased RhoA activation and promoted GTP-loading of Rac1, leading to stimulation of the Rac1-effector PAK1, which is the kinase responsible for subsequent phosphorylation of MEK1 at serine 298, thereby increasing MEK affinity towards ERK1/2. We propose that WNK2 controls a RhoA-mediated cross-talk mechanism that regulates the efficiency with which MEK1 can activate ERK1/2 upon growth factor stimulation.  相似文献

The role of hydrological droughts in shaping meiofauna abundance through alterations in biofilm biomass and composition was investigated. In January 2005, continental Portugal was under a moderate to severe drought resulting from a 40% to 60% decrease in rainfall during the previous 12 months relative to the long-term average (1961–1990). Reservoir capacity was reduced by 30–50% relative to average values and the width of streams was reduced by 20–80% in the Zêzere River Basin (central Portugal). Algal biomass and algal class composition of biofilms was assessed through quantification of algal pigments in three reservoir and six river locations. During drought, habitat alterations are expected to be sharp in rivers while, in the absence of water quality deterioration, the habitat characteristics of reservoirs are expected to remain fairly unaffected. Chlorophylls and carotenoid pigments were extracted from biofilm samples and analysed using high performance liquid chromatography (HPLC). In the winter of 2003, during the period of average rainfall, biofilm biomass did not exceed 5 μg chlorophyll a cm−2 at any location. River biofilm biomass was roughly half of that measured in the reservoirs. In the winter of 2005 (drought), biofilm biomass increased by more than 5-fold in river locations and remained low or decreased in the reservoirs. Algal biofilms were either dominated by Bacillariophyceae or by Chlorophyceae regardless of the existence of drought. The relative contribution of Bacillariophyceae to total biofilm biomass was higher during the drought than under average hydrological conditions. The abundance of harpacticoids, cladocerans and ostracods was favoured by the drought only in the reservoirs where an increase in diatom proportion in biofilms was observed. The increase in the abundance of cyclopoid copepods, turbellarians, nematodes and chironomids in rivers during the drought could be explained by algal class composition and biomass of biofilms and environmental variables (organic matter sediment content, phosphorus availability content and sediment granulometry). The hydrological drought appears to regulate meiofauna abundance only in river locations, possibly through the promotion of the growth of biofilms and the availability of organic matter deposited in rivers during the drought. Plants grown at limited P supply can increase the activity of phosphatases in roots to hydrolyse organic-P compounds in the soil thus improving plant P acquisition, but little information is available about the role of these enzymes for internal plant metabolism at limited-P conditions. This work intended to measure the activities of acid phosphatases and phytases in nodules of common bean (Phaseolus vulgaris) genotypes at different levels of P supply. The experiment was carried out in a 5?×?5 factorial design with four replicates, comprising five bean genotypes and five P levels (20, 40, 80, 160 and 320 μmol P plant?1 week?1) in nutrient solution. Root seedlings were inoculated with Rhizobium tropici and plants were grown in 1-l bottles. Nodule samples were detached from 39-day-old plants and enzyme activities were determined in crude extracts. Plants were harvested at the stage of pod setting. Polynomial models fitted to data indicated maximal values at the level of 194 μmol P for shoot mass, at 206 μmol P for nodule mass and at 221 μmol P for shoot N. Whereas shoot mass was 1.7 times lower at 20 than at 160 μmol P, nodule mass was 7.5 times lower. Concentration of P in nodules increased from 40 to 320 μmol P but remained stable between 20 and 40 μmol P, suggesting a minimal threshold concentration of 3 mg P g?1 for nodule growth. Activities of phosphatases and phytases in nodules decreased strongly as P supply was raised from 20 to 80 μmol P, remaining almost stable at higher P levels. Phosphatase activity ranged from 1,154 to 406 nmol min?1 g?1 (nodule fresh mass) from 20 to 80 μmol P respectively, while the phytase activity ranged from 55 to 14 nmol min?1 g?1 from 20 to 80 μmol P. Bean genotypes differed in shoot and nodule mass at the levels of 80 and 160 μmol P, whilst they differed in nodule enzyme activities only at the lowest P level, the relationship between nodule enzyme activities and growth of different bean genotypes was not evident. It is concluded that bean plants at P-deficient conditions increase the activities of phosphatases and phytases in nodules. This may constitute an adaptive mechanism for N2-fixing legumes to tolerate P deficiency, by increasing the utilisation of the scarce P within the nodules.  相似文献
