We have addressed the control of longitudinal axon pathfinding in the developing hindbrain, including the caudal projections of reticular and raphe neurons. To test potential sources of guidance signals, we assessed axon outgrowth from embryonic rat hindbrain explants cultured in collagen gels at a distance from explants of midbrain-hindbrain boundary (isthmus), caudal hindbrain, or cervical spinal cord. Our results showed that the isthmus inhibited caudally directed axon outgrowth by 80% relative to controls, whereas rostrally directed axon outgrowth was unaffected. Moreover, caudal hindbrain or cervical spinal cord explants did not inhibit caudal axons. Immunohistochemistry for reticular and raphe neuronal markers indicated that the caudal, but not the rostral projections of these neuronal subpopulations were inhibited by isthmic explants. Companion studies in chick embryos showed that, when the hindbrain was surgically separated from the isthmus, caudal reticulospinal axon projections failed to form and that descending pioneer axons of the medial longitudinal fasciculus (MLF) play an important role in the caudal reticulospinal projection. Taken together, these results suggest that diffusible chemorepellent or nonpermissive signals from the isthmus and substrate-anchored signals on the pioneer MLF axons are involved in the caudal direction of reticulospinal projections and might influence other longitudinal axon projections in the brainstem. 相似文献

Oxidative stress designates the state of imbalance between reactive oxygen species (ROS) production and antioxidant levels. In a healthy placenta, there is an increase in ROS production, due to formation of new tissues and inherent metabolism, but this is balanced by higher levels of antioxidants. However, this balance is lost in some situations, with a consequent increase in oxidative stress levels. Oxidative stress has been implicated in several placental disorders and pregnancy pathologies. Species diversity within the genus Osmundea in the Macaronesian region was explored by conducting a comprehensive sampling in the Azores, the Canary, and the Madeira archipelagos. Toward identification, all specimens were first observed alive to verify the absence of corps en cerise, a diagnostic character for the genus and morphometric data were measured (thallus length and width, first‐order branches length and width, branchlets length and width, cortical cell length and width in surface view, cortical cell length and width in transverse section). Specimens were sequenced for COI‐5P (39 specimens) and three species delimitation methods (Generalized Mixed Yule Coalescent, Automatic Barcode Gap Discovery method, and Poisson Tree Processes) were used to assess the threshold between infra‐ and interspecific relationships. Subsequently, one or several sequences of plastid‐encoded large subunit of RuBisCO (21 specimens) per delimited species were generated to assess the phylogenetic relationships among Macaronesian Osmundea. Moreover, for each delineated species, vegetative and reproductive anatomy was thoroughly documented and, when possible, specimens were either assigned to existing taxa or described as novel species. This integrative approach has provided data for (i) the presence of O. oederi, O. pinnatifida, and O. truncata in Macaronesia; (ii) the proposal of two novel species, O. prudhommevanreinei sp. nov. and O. silvae sp. nov.; and (iii) evidence of an additional species referred as “Osmundea sp.1,” which is a sister taxon of O. hybrida. 相似文献

Prions are the infectious agents responsible for prion diseases, which appear to be composed exclusively by the misfolded prion protein (PrP(Sc)). Disease is transmitted by the autocatalytic propagation of PrP(Sc) misfolding at the expense of the normal prion protein. The biggest challenge of the prion hypothesis has been to explain the molecular mechanism by which prions can exist as different strains, producing diseases with distinguishable characteristics. Here, we show that PrP(Sc) generated in vitro by protein misfolding cyclic amplification from five different mouse prion strains maintains the strain-specific properties. Inoculation of wild-type mice with in vitro-generated PrP(Sc) caused a disease with indistinguishable incubation times as well as neuropathological and biochemical characteristics as the parental strains. Biochemical features were also maintained upon replication of four human prion strains. These results provide additional support for the prion hypothesis and indicate that strain characteristics can be faithfully propagated in the absence of living cells, suggesting that strain variation is dependent on PrP(Sc) properties. 相似文献

In the present study we analyzed medicinal and edible plant utilization in Cuyin Manzano, a small rural population located
near the Andean forests of Argentina. We also studied where and when plant knowledge was learned, who the principal transmitters
were, and how people were taught. The participants were interviewed individually and at random, by means of a semi-structured
questionnaire. Interviews were carried out in 16 families in order to examine the present use of wild plants. The inhabitants
of Cuyin Manzano cited 87 plants: 63 medicinal and 24 edible species. They mentioned on average 31 ± 10 species per person.
Similar patterns of plant use were found in young and old people alike, irrespective of gender. Learning about useful plants
took place at an early age as a result of family tradition. This local knowledge is acquired and taught “by doing,” and is
mostly transmitted vertically through family dissemination. Wild plant learning implies the acquisition of plants' physical

The baculiform, rod-like morphotypes belong to several phylogenetic lineages within Desmidiaceae (Zygnematophyceae, Viridiplantae). Some, for example the genus Pleurotaenium, form independent lineages, but reductive evolution of complicated desmid cells toward baculiform morphology also occurred in individual lineages, for example Micrasterias. In this genus, the rod like Triploceras forms evolved from more complex ancestors. In this study, we tested for an independent position of the subtropical and tropical genus Triplastrum, previously separated from Triploceras on the basis of morphological data. In addition, monophyly of Pleurotaenium was also investigated with multiple isolates corresponding to seven species of this genus, including the morphologically dissimilar P. nodosum and P. ovatum. Finally, two isolates of Docidium baculum were also investigated. Molecular phylogenetic analysis of concatenated rbcL?+?coxIII sequence data implied that the baculiform taxa investigated were in three distantly related positions within Desmidiaceae. The genus Triplastrum proved to be unrelated to Triploceras, because it clustered in the “omniradiate” lineage of Desmidiaceae among morphologically dissimilar taxa. The genus Pleurotaenium was monophyletic, but P. ovatum was recovered in a weakly supported sister position to all the other members of the genus. The trnGucc phylogeny of Pleurotaenium taxa concurred with the rbcL?+?coxIII phylogram, and generally revealed the poor morphological concepts of some species in this genus. The most common taxa P. ehrenbergii and P. trabecula were resolved as polyphyletic because their strains were distributed among several strongly supported clades. However, strains of P. nodosum and P. archeri formed separate, well supported lineages within the genus. 相似文献

The gut microbiota plays an important yet incompletely understood role in the induction and propagation of ulcerative colitis (UC). Organism-level efforts to identify UC-associated microbes have revealed the importance of community structure, but less is known about the molecular effectors of disease. We performed 16S rRNA gene sequencing in parallel with label-free data-dependent LC-MS/MS proteomics to characterize the stool microbiomes of healthy (n = 8) and UC (n = 10) patients. Comparisons of taxonomic composition between techniques revealed major differences in community structure partially attributable to the additional detection of host, fungal, viral, and food peptides by metaproteomics. Differential expression analysis of metaproteomic data identified 176 significantly enriched protein groups between healthy and UC patients. Gene ontology analysis revealed several enriched functions with serine-type endopeptidase activity overrepresented in UC patients. Using a biotinylated fluorophosphonate probe and streptavidin-based enrichment, we show that serine endopeptidases are active in patient fecal samples and that additional putative serine hydrolases are detectable by this approach compared with unenriched profiling. Finally, as metaproteomic databases expand, they are expected to asymptotically approach completeness. Using ComPIL and de novo peptide sequencing, we estimate the size of the probable peptide space unidentified (“dark peptidome”) by our large database approach to establish a rough benchmark for database sufficiency. Despite high variability inherent in patient samples, our analysis yielded a catalog of differentially enriched proteins between healthy and UC fecal proteomes. This catalog provides a clinically relevant jumping-off point for further molecular-level studies aimed at identifying the microbial underpinnings of UC. 相似文献

The Shannon, Ireland’s largest river, is used for hydroelectricity generation since 1929. Subsequently, the Electricity Supply
Board assumed responsibility for management of its eel stocks, due to the impact of the hydro-dam on recruitment to the commercial
fishery. In order to negate a decline in juvenile recruitment resulting from the installation of hydroelectric facilities,
management was focused on stocking lakes with elvers and fingerling eels. These were trapped at the hydropower facilities
and in estuarine tributaries during their up-stream migrations. Due to the decline of natural recruitment in more recent times,
attempts have also been made to develop an estuarine glass eel fishery. Stock levels are then monitored through annual surveys
of the population trends of juvenile (glass eel, elver), growing phase (yellow eel) and downstream migrating pre-spawners
(silver eels). Survey results and fishery management programmes are reviewed in this article. In addition to the long-term
effects the hydroelectric facilities have had on the stock levels, there is also an annual effect on the migratory patterns
of downstream migratory silver eels. In the lower reaches of the river system flow rates are regulated by the hydroelectric
stations. We review previous work that had highlighted the importance of flow in determining the timing of the silver eels
migrations, and assess the relationship between flow and migration in more detail through the use of hydroacoustic and telemetric
studies. Current research on seaward migrating silver eel populations, suggests that spawner escapement rates can most effectively
be increased by trapping migrating eels at fishing weirs located up-stream of the power station and transporting them towards
the estuary.
