The effect of picrotoxin (at sub-convulsant doses, i.v.) on the background and invoked spike activity of cat dorsal horn internerons was investigated while thermal and mechanical influences were at work on the skin receptor areas. Male albino rats were exposed to daily emotional painful stress (EPS) for 4 weeks. The arterial blood pressure of the stressed animals increased and the dynamics of the heart rate changed after functional loading (hypokinesis during one or two hours) as compared with the control group. The increase of the heart weight and activation of cytochrome oxidase activity in the cerebral cortex and hippocampus of rats exposed to EPS were also demonstrated. The use of 20% ethyl alcohol instead of drinking water during EPS partially prevented vegetative disturbances and activation of hippocampal cytochrome oxidase and fully prevented the heart hypertrophy and activation of the enzyme in the cortex. Alcoholization resulted in the increased weight of the spleen and brown adipose tissue and thymus involution. A possible mechanism of the antistress action of alcohol linked with normalization of intensified lipid peroxidation under stress is discussed. 相似文献

Activities of glycolytic enzymes in the aorta were investigated in female Wistar rats. There were two groups of rats; one served as the control (sedentary rats), while the other group was forced to run on a treadmill for 10 weeks. In the control animals, the activities of hexokinase, phosphofructokinase and aldolase were relatively lower than those of the other glycolytic enzymes (phosphoglucose isomerase, lactate dehydrogenase and pyruvate kinase). After exercise, the activity of phosphofructokinase increased by 15%, whereas the other enzymatic activities were much the same as in the controls. Within the limits of the experiments, the increased percentage of phosphofructokinase was statistically significant (p less than 0.05). Since phosphofructokinase is a putative rate limiting enzyme, this enzymatic activation may indicate that glycolytic activity in the rat aorta is enhanced during and after running exercise. 相似文献

Blockade of the spread depression [SD] in chemically or electrically stimulated areas of the cerebral cortex was analyzed in a series of experiments performed in 40 curarized, locally anaesthetised rats. Longlasting stimulation of the cerebral cortex (0.5 to 1.7 mA, 6 Hz, 0.1 ms) elicited recurrent episodes of enhanced evoked potentials propagating to remote cortical areas [Co], the caudate nucleus [Cd], the hippocampus [Hi] and the thalamus [Th] and was accompanied by marked slow potential shifts (4-6 mV amplitude, 2-3 min duration, at 2-5 min intervals]. The projected discharge interfered with SD initiation and propagation in all the examined structures. The SD blockade was most pronounced during the episodes and almost absent in the intervals between them. The block was manifested by reduced amplitude and duration of the slow potential of SD. Pentobarbital [20 mg/kg] suppressed the recurrent discharges and eliminated the corresponding SD blockade. Recurrent excitability changes induced by Cd and Th stimulation elicited similar effects but the threshold was higher in Cd and Th than in Co. SD was less effectively blocked by the projected discharge than by stimulation of the same structure, particularly in the vicinity of the recording electrodes, where the blockade could be observed even under pentobarbital. The SD blockade outlasted stimulation by a considerably longer period of time in Th (about 10 min) than in the Cd (about 3 min). The onset of stimulation and the projected episodes sometimes elicited SD waves but SD blockade prevailed with continued stimulation. The present findings support the hypothesis that excessive neural activity increases the potassium clearance and thus prevents the autoregenerative accumulation of potassium ions, mediating SD propagation. 相似文献

1 alpha-Hydroxyvitamin D-3 25-hydroxylase activity was measured in subcellular fractions of rat and human liver. The formation of 1,25-dihydroxyvitamin D-3 was determined by high pressure liquid chromatography. In rat liver 1 alpha-hydroxyvitamin D-3 25-hydroxylase activities were found in the purified nuclei, the heavy mitochondrial fraction and the microsomal fraction. The enrichment of 25-hydroxylase activity was highest in the heavy mitochondrial fraction. With this fraction a minimum amount (about 0.5 mg) of protein was required before formation of 1,25-dihydroxyvitamin D-3 could be detected. Above this amount the reaction was linear with amount of protein up to at least 2 mg/ml. The reaction was also linear with time up to 60 min. An apparent Km value of 2 X 10(-5) M was found. The mitochondrial 25-hydroxylase was stimulated by addition of cytosolic protein or bovine serum albumin. The degree of stimulation was dependent on the amount of mitochondrial protein present in the incubation mixture. Maximal stimulation was seen with 0.2 mg/ml of either protein in the presence of 0.5 mg mitochondrial protein. The stimulating effect remained after heating the protein for 5 min at 100 degrees C. The cytosolic protein did not stimulate a reconstituted mitochondrial 1 alpha-hydroxyvitamin D-3 25-hydroxylase. The mitochondrial vitamin D-3 25-hydroxylase was inhibited both by cytosolic protein and by bovine serum albumin. Human liver revealed only one 1 alpha-hydroxyvitamin D-3 25-hydroxylase activity located to the heavy mitochondrial fraction. The results are in agreement with previous studies on the localization of vitamin D-3 25-hydroxylase in rat and human liver. The difference in localization of the 25-hydroxylase between rat and human liver implies that studies on the regulation of the microsomal 25-hydroxylase in rat liver may not be relevant to the situation in human liver. 相似文献

Abstract: Fossils of dyrosaurid crocodyliforms are limited in South America, with only three previously diagnosed taxa including the short‐snouted Cerrejonisuchus improcerus from the Paleocene Cerrejón Formation of north‐eastern Colombia. Here we describe a second dyrosaurid from the Cerrejón Formation, Acherontisuchus guajiraensis gen. et sp. nov., based on three partial mandibles, maxillary fragments, teeth, and referred postcrania. The mandible has a reduced seventh alveolus and laterally depressed retroarticular process, both diagnostic characteristics of Dyrosauridae. Acherontisuchus guajiraensis is distinct among known dyrosaurids in having a unique combination of craniomandibular characteristics, and postcranial morphology that suggests it may have occupied a more placid, fluvial habitat than most known Old‐World dyrosaurids. Results from a cladistic analysis of Dyrosauridae, using 82 primarily cranial and mandibular characters, support an unresolved relationship between A. guajiraensis and a combination of New‐ and Old‐World dyrosaurids including Hyposaurus rogersii, Congosaurus bequaerti, Atlantosuchus coupatezi, Guarinisuchus munizi, Rhabdognathus keiniensis and Rhabdognathus aslerensis. Our results are consistent with an African origin for Dyrosauridae with multiple dispersals into the New World during the Late Cretaceous and a transition from marine habitats in ancestral taxa to more fluvial habitats in more derived taxa. 相似文献

Understanding the nature of organic matter is a necessary first step in assessing contaminant bioavailability and allowing
water supply managers to optimise the treatment train in the aim of providing safe and inexpensive drinking water. This study
provides further insight into the composition, structure and functional groups of dissolved organic matter (DOM) (both hydrophobic
and hydrophilic) from urban aquatic systems by means of various analytical techniques (DAX-8/XAD-4 fractionation, elemental
analysis, UV and FTIR spectroscopies, 13C and 15N isotopic analysis, size exclusion chromatography and Pyrolysis-GC-MS). The analytical range chosen for this study constitutes
a powerful tool in the characterisation of DOM in urban water. The inclusion of information from one technique to the next
might not only serve as a support to each one, but also as a complement. The DOM fraction from treated effluent and, more
generally, DOM from urban water (i.e. receiving treated effluent) display a strong hydrophilic characteristic [i.e. low humic
substance (HS) content, low SUVA], along with a high distribution in molecular weights observed by SEC and low average molecular
weight. Due to the origin of this DOM, proteinaceous structures constitute the main compounds, as observed by FTIR and Py-GC-MS.
Such characteristics (i.e. heterogeneity, low average molecular weight and diverse functional groups, which make up a total
of N) could explain that DOM from treated effluent displayed a strong reactive potential metals pollutants as previously demonstrated. 相似文献
