This overview summarizes the data for how epithelial cells sort and deliver proteins and lipids to the apical and basolateral cell surface domains. The basolateral pathway uses a Rab-SNARE mechanism for docking and fusion, while the apical route employs a different machinery. This latter mechanism is based on lipid microdomains, composed of clusters of sphingolipids and cholesterol, which function as rafts for apical delivery. The sphingolipid-cholesterol raft mechanism seems to be employed generally by mammalian cells to transport raft-associated proteins to their post-Golgi destinations. 相似文献

Social attachment is vital for human health and welfare. Recent experimental evidence in humans has identified the role of neuroactive hormones, especially the peptide oxytocin, in mediating trusting behaviors. Herein, we test if the endocrinological basis for trust between humans scales up to the country level. Trust pervades nearly every aspect of our daily lives, yet survey data on trust show substantial variation across countries. Using 31 measures of biological, social, and environmental factors associated with hormone levels for a sample of 41 countries, we find that two classes of factors are related to trust: consumption of plant-based estrogens (phytoestrogens), and the presence of environmental conditions that include measures of estrogen-like molecules. Our findings provide preliminary evidence that interpersonal trust at the country level may be related to the intake of neuroactive hormones. 相似文献

Few topics in palaeoanthropology have generated more recent debate than the nature and causes of the remarkable transformation in human behavioral patterns that marked the transition from the Middle to the Upper Paleolithic in Europe. 1 - 11 Those of us who have argued for an effective technological and cultural “revolution” at this point in the Paleolithic sequence have emphasized three main dimensions 1 , 2 , 9 , 11 - 14 : the wide range of different aspects of behavior that appear to have been affected (Fig. 1); the relative speed and abruptness with which most of these changes can be documented in the archeological records from the different regions of Europe; and the potentially profound social and cognitive implications of many of the innovations involved. Most striking of all in this context is the abrupt appearance and proliferation of various forms of perforated animal teeth, shells, beads, and other personal ornaments, and the even more dramatic eruption of remarkably varied and sophisticated forms of art, ranging from representations of male and female sex organs, through the highly stylized animal and combined animal‐human figures from southern Germany, to the striking wall paintings of the Chauvet Cave. 8 , 15 - 18 One might add to this the similar proliferation of more enigmatic but potentially equally significant abstract “notation” systems on bone and ivory artifacts. 19 To describe the Upper Paleolithic revolution in Europe as reflecting preeminently an explosion in explicitly symbolic behavior and expression is in no sense an exaggeration, as most prehistorians would now agree. The number of orientation, feeding and social calls emitted by pipistrelle batsPipistrellus pipistrellus Schreber, 1774 andP. pygmaeus Leach, 1825 was recorded throughout the night at eight different sites. Social calls were unaffected by weather variables,
whereas orientation calls and feeding buzzes were both significantly affected by cloud and temperature conditions. The number
of emissions of each call type was significantly different between sites, indicating that the bats used different sites for
different activities. Significant positive correlations between all three combinations of call types occurred only during

In Mytilus and Leucophaea the high-affinity binding site density is significantly lower in old animals than in young animals, whereas the low-affinity site density remains unchanged. In Mytilus the estimated met-enkephalin and met-enkephalin-Arg6-Phe7 levels are significantly higher in old than in young animals. In Leucophaea only the met-enkephalin level can be determined, and it is also higher in old animals. The decrease in the high-affinity binding site density and the corresponding increase in endogenous enkephalin levels suggest the existence of an opioid compensatory mechanism associated with the aging process. In Mytilus there is a demonstrated decrease with age in intraganglionic dopamine levels in response to applied opiates. In addition, the inhibition of dopamine-stimulated adenylate cyclase activity by opiates also decreases in older animals. In the albino mutant of an Okinawa strain of Locusta migratoria (L.) (Orthoptera: Acrididae), albinism is caused by the absence of the dark‐colour‐inducing neurohormone (DCIN), which is present in the corpora cardiaca (CC) of normally coloured phenotypes. This study tests whether the absence of DCIN is responsible for albinism in an albino mutant of another locust, Schistocerca gregaria (Forsk.) (Orthoptera: Acrididae). This seemed feasible because a single Mendelian unit controls albinism in both species. However, implantation of CC, or injection of an extract of CC, from albino donors of S. gregaria, induce dark coloration in crowded nymph recipients of the Okinawa albino mutant of L. migratoria, as effectively as do implanted CC, or injections of extract of CC, from normal phenotype donors of S. gregaria. Therefore, DCIN is present in the albino mutant of S. gregaria, and consequently, the albinism in this mutant is not caused by its absence. Implantation of CC, or injection of extracts of CC, from albino donors of S. gregaria to conspecific albino nymphs does not induce darkening. Only extremely high doses of synthetic DCIN injected into albino nymphs of S. gregaria are effective, inducing some darkening. The dose to induce such darkening in albino nymphs of S. gregaria is 50 nmol, ≈ 5 × 106 times higher than that (10 femtomol) needed to induce equivalent darkening in nymphs of the Okinawa albinos of L. migratoria. Expression of alternatively spliced mRNA variants at specific stages of development or in specific cells and tissues contributes to the functional diversity of the human genome. Aberrations in alternative splicing were found as a cause or a contributing factor to the development, progression, or maintenance of numerous diseases. The use of antisense oligonucleotides (ON) to modify aberrant expression patterns of alternatively spliced mRNAs is a novel means of potentially controlling such diseases. Oligonucleotides can be designed to repair genetic mutations, to modify genomic sequences in order to compensate for gene deletions, or to modify RNA processing in order to improve the effects of the underlying gene mutation. Steric block ON approach have proven to be effective in experimental model for various diseases. Here, we describe our experience in investigating two strategies for ON delivery: ON conjugation with basic peptides and lipid-based particulate system (lipoplex). Basic peptides or Cell Penetrating Peptides (CPP) such as the TAT-derived peptide appear to circumvent many problems associated with ON and drug delivery. This strategy may represent the next paradigm in our ability to modulate cell function and offers a unique avenue for the treatment of disease. Lipoplexes result from the intimate interaction of ON with cationic lipids leading to ON carrying particles able to be taken up by cells and to release ON in the cytoplasm. We have used as an experimental model the correction of a splicing alteration of the mutated β-globin intron causing thalassemia. Data on cell penetration and efficacy of correction of specific steric block ON delivered either by basic peptides or lipoplex are described. A comparison of the properties of both delivery systems is made respective to the use of this new class of therapeutic molecules. 相似文献
