Adenylyl and guanylyl cyclases synthesize second messenger molecules by intramolecular esterification of purine nucleotides, i.e., cAMP from ATP and cGMP from GTP, respectively. Despite their sequence homology, both families of mammalian cyclases show remarkably different regulatory patterns. In an attempt to define the functional domains in adenylyl cyclase responsible for their isotypic-common activation by Galphas or forskolin, dimeric chimeras were constructed from soluble guanylyl cyclase alpha1 subunit and the C-terminal halves of adenylyl cyclases type I, II, or V. The cyclase-hybrid generated cAMP and was inhibited by P-site ligands. The data establish structural equivalence and the ability of functional complement at the catalytic sites in both cyclases. Detailed enzymatic characterization of the chimeric cyclase revealed a crucial role of the N-terminal adenylyl cyclase half for stimulatory actions, and a major importance of the C-terminal part for nucleotide specificity.  相似文献

Hypopigmentation is a characteristic of several diseases associated with vesicle traffic defects, like the Hermansky–Pudlak, Chediak–Higashi, and Griscelli syndromes. Hypopigmentation is also a characteristic of the zebrafish mutant vps18hi2499A, which is affected in the gene vps18, a component of the homotypic fusion and protein sorting complex that is involved in tethering during vesicular traffic. Vps18, as part of this complex, participates in the formation of early endosomes, late endosomes, and lysosomes. Here, we show that Vps18 is also involved in the formation of melanosomes. In the zebrafish mutant vps18hi2499A the retroviral insertion located at exon 4 of vps18, leads to the formation of two abnormal splicing variants lacking the coding sequence for the clathrin repeat and the RING finger conserved domains. A deficiency of Vps18 in zebrafish larvae results in hepatomegaly and skin hypopigmentation. We also observed a drastic reduction in the number of melanosomes in the eye's retinal pigmented epithelium along with the accumulation of immature melanosomes. A significant reduction in the vps18hi2499A larvae visual system capacity was found using the optokinetic response assay. We propose that the insertional mutant vps18hi2499A can be used as a model for studying hypopigmentation diseases in which vesicle traffic problems exist.  相似文献

Certain cellular processes take the form of activity pulses that can be interpreted in terms of noise-driven excitable dynamics. Here we present an overview of different gene circuit architectures that exhibit excitable pulses of protein expression, when subject to molecular noise. Different types of excitable dynamics can occur depending on the bifurcation structure leading to the specific excitable phase-space topology. The bifurcation structure is not, however, linked to a particular circuit architecture. Thus a given gene circuit design can sustain different classes of excitable dynamics depending on the system parameters.  相似文献

We report a novel application for the operator-repressor titration (ORT) plasmid maintenance system. The ability of ORT to maintain a plasmid during production of DNA has been demonstrated previously. In this study, we have used the ORT system to maintain a plasmid during high cell density cultivation and expression of a recombinant protein. No evidence of plasmid loss was seen during protein expression at high cell densities. In addition, the quantity of protein produced using this system was similar to traditional plasmid maintenance systems.  相似文献

European starlings ( Sturnus vulgaris ) introduced to North America compete with native cavity-nesting birds for nest sites. I examined whether natural selection could favour a strategy of delayed nesting in a population of northern flickers ( Colaptes auratus ), a native woodpecker, to reduce overlap in breeding phenology with starling competitors. I developed a mathematical model based on reproductive parameters for a population of flickers from central British Columbia, Canada. On average, 7% of flicker nests each year were usurped by starlings; daily probabilities of takeover declined through the season but were relatively low (1–2%). Flickers laid between 3 and 13 eggs and there was a negative correlation between clutch size and date. The probability of renesting after nest failure also declined during the season, but renesting intervals (2–19 days) were not associated with female age, or stage of nest loss. The model suggested that costs of delaying reproduction would outweigh benefits of an early start except when the probability of nest usurpation is very high (>75%) early in the season and declines rapidly through the summer. Thus, early laying dates should be favoured especially in northern latitudes where breeding seasons are short.  相似文献

Summary As components of combined fermentation of both glucose and xylose to ethanol by separated or coculture processes, the effects of initial sugar concentrations on the fermentative performances ofPichia stipitis Y7124,Candida shehatae ATCC 22984,Saccharomyces cerevisiae CBS1200 andZymomonas mobilis ATCC10988 were investigated. Randomized peptide sequences displayed at the surface of filamentous phages are often used to select antibody ligands. The selected sequences are generally further used in the form of synthetic peptides; however, as such, their affinity for the selecting antibody is extremely variable and factors influencing this affinity have not been fully deciphered. We have used an f88.4 phage-displayed peptide library to identify ligands of mAb 11E12, an antibody reactive to human cardiac troponin I. A majority of the sequences thus selected showed a (T/A/I/L) EP(K/R/H) motif, homologous to the Y-TEPH motif identified by multiple peptide synthesis as the critical motif recognized by mAb 11E12 in the peptide epitope. A set of 15-mer synthetic peptides derived from the phage-selected sequences was used in BIACORE to characterize their interaction with mAb 11E12. Most peptides exhibited affinities in the 7-26 nM range. These affinities represented, however, only 1.9-7. 5% of the affinity of the 15-mer peptide epitope. In circular dichroism experiments, the peptide epitope showed a propensity to have some stabilized conformation, whereas a low-affinity peptide selected by phage-display did not. To try to decipher the molecular basis of this difference in affinity, new peptides were prepared by grafting the N- or the C-terminal sequence of the peptide epitope to the Y-TEPK motif of a low-affinity peptide selected by phage-display. These hybrid peptides showed marked increases both in affinity (as assessed using BIACORE) and in inhibitory potency (as assessed in competition ELISA), compared with the parent sequence. Thus, the sequences flanking the motif, although not containing critical residues, convey some determinants necessary for high affinity. The affinity of a given peptide strongly depends on its capacity to maintain the antigenically reactive structure it has on the phage, implying that it is impossible to predict whether high- or low-affinity peptides will be obtained from phage display.  相似文献

The structural and ultrastructural characteristics of adrenocorticotropic hormone cells in adult female goats in anoestrus, gestation and milk production were studied with an immunohistochemical method (peroxidase-antiperoxidase). Only one cellular type has been identified and is characterized by numerous secretory granules of different electron density and an average diameter of 275 nm. During pregnancy these cells increase in number and size, and there is a frequent presence of vacuoles. During lactation the number of size of the cells decreases but without reaching the state observed in anoestrus and the involution of the cytoplasmic vacuolizations which appear in pregnancy.  相似文献

Hummingbird-pollinated flowers are frequently subjected to nectar robbing. In this paper, I examine the impact of nectar robbing on plant reproductive success on a hummingbird-pollinated species. After studying the basic aspects of the floral morphology and reproduction of Macleania bullata (Ericaceae) in a tropical montane wet forest in southwest Colombia, I examined the percent of flowers robbed and the effect of nectar robbery on fruit set. The flowers of this species are typical for plants pollinated by long-bill hummingbirds. They are protandrous and open for four days. Fruit production requires a pollinator visit; fruit set following pollinator exclusion was zero. Fruit set following xenogamous pollen transfer (36.8%) differed significantly from that of population controls (11.9%) and of autogamous pollen transfer (6.3%). Nectar volume, sugar concentration and sugar production were measured at daily intervals from bud opening until the fading of flowers. Daily nectar production (both volume and amount of sugar) varied considerably with flower age. Sugar production peaked on the second day, coinciding with the male phase. The frequency of nectar robbing in the studied population was very high (75% of examined flowers) and was positively correlated with reduced fruit set. I discuss the probability of a relation between reduced fruit set on robbed flowers and an energetic investment. Robbing by non-pollinating visitors can suppose the plant to re-synthesize more nectar. The high incidence of nectar robbing impugns the advantage of specialization.  相似文献
