ac) was maintained 1.4°C above pre-exposure levels for 70 min by manipulating the work rate. On days 1 and 6, T
ac and local sweat rates (m˙
sw: eight sites) were monitored. The pre-exposure, resting T
ac and the T
ac sweat threshold decreased from day 1 to day 6 [36.83 (0.05)°C vs 36.62 (0.05)°C, and 36.90 (0.05)°C vs 36.75 (0.05)°C, respectively;
both P<0.05]. However, the sweat-onset time, sweat sensitivity (Δm˙
sw/ΔT
ac) and established m˙
sw were unaltered (P > 0.05). There was also no evidence of a post-acclimation redistribution in established m˙
sw between the eight skin regions, though both the sweat sensitivity and established m˙
sw for the forehead and hand were significantly greater than at the remaining sites (P<0.05). It is concluded that the 5-day heat acclimation regimen provided only a minimal stimulus for sudomotor adaptation.

We have investigated properties relevant to quantitative imaging in living cells of five green fluorescent protein (GFP) variants that have been used extensively or are potentially useful. We measured the extinction coefficients, quantum yields, pH effects, photobleaching effects, and temperature-dependent chromophore formation of wtGFP, alphaGFP (F99S/M153T/V163A), S65T, EGFP (F64L/S65T), and a blue-shifted variant, EBFP (F64L/S65T/Y66H/Y145F). Absorbance and fluorescence spectroscopy showed little difference between the extinction coefficients and quantum yields of wtGFP and alphaGFP. In contrast, S65T and EGFP extinction coefficients made them both approximately 6-fold brighter than wtGFP when excited at 488 nm, and EBFP absorbed more strongly than the wtGFP when excited in the near-UV wavelength region, although it had a much lower quantum efficiency. When excited at 488 nm, the GFPs were all more resistant to photobleaching than fluorescein. However, the wtGFP and alphaGFP photobleaching patterns showed initial increases in fluorescence emission caused by photoconversion of the protein chromophore. The wtGFP fluorescence decreased more quickly when excited at 395 nm than 488 nm, but it was still more photostable than the EBFP when excited at this wavelength. The wtGFP and alphaGFP were quite stable over a broad pH range, but fluorescence of the other variants decreased rapidly below pH 7. When expressed in bacteria, chromophore formation in wtGFP and S65T was found to be less efficient at 37 degrees C than at 28 degrees C, but the other three variants showed little differences between 37 degrees C and 28 degrees C. In conclusion, no single GFP variant is ideal for every application, but each one offers advantages and disadvantages for quantitative imaging in living cells. 相似文献

We have designed protein molecules based on an -helical coiled-coil structure. These proteins can be tailored to complement nutritionally unbalanced seed meals. In particular, these proteins may contain up to 43% mol/mol of the essential amino acid lysine. Genes encoding such proteins were constructed using synthetic oligonucleotides and the protein stability was tested for in vivo by expression in an Escherichia coli model system. A protein containing 31% lysine and 20% methionine (CP 3-5) was expressed in transgenic tobacco seeds utilizing the seed specific bean phaseolin and soybean -conglycinin promoters. Both promoters provided a level of expression in the mature transgenic tobacco seeds which resulted in a significant increase in the total lysine content of the seeds. Several of these transgenic lines were analyzed for three generations to determine the stability of gene expression. Plants transformed with the soybean -conglycinin promoter/CP 3-5 gene consistently expressed the high-lysine phenotype through three generations. However, expression of the high-lysine phenotype in plants transformed with the bean phaseolin/CP 3-5 was variable. This is the first report of a significant increase in seed lysine content due to the seed-specific expression of a de novo protein sequence. 相似文献

Heterotrophic flagellates, centrohelid heliozoa and filose amoebae were recorded from cultures derived from water collected
at marine and freshwater sites in the Antarctic. Marine samples were collected in the vicinity of Prydz Bay, southeast Antarctica
and freshwater samples from Sombre Lake on Signy Island and Crooked and Druzhby Lakes in the Vestfold Hills. Thirty-five species
were identified. One new species, Kiitoksia kaloista (Protista incertae sedis), is described. The other species have been previously reported from other geographic locations, providing no evidence for
endemism in protozoan species found in Antarctica.

OBJECTIVE: To determine whether a computerised decision support system for initiation and control of oral anticoagulant treatment improves quality of anticoagulant control achieved by trainee doctors. DESIGN: Randomised controlled trial. SETTING: District general hospital in North London. SUBJECTS: 148 inpatients requiring start of warfarin treatment. INTERVENTIONS: Management by trainee doctors (to achieve therapeutic range of international normalised ratio of 2 to 3) with indirect assistance from computerised decision support system (intervention group) or without such assistance (control group). MAIN OUTCOME MEASURES: Median time to therapeutic range, stable dose, and first pseudoevent (excessive international normalised ratio after therapeutic range has been reached) and person time spent in the therapeutic range. RESULTS: 72 patients were randomised to the intervention group and 76 to control group. Median time to reach international normalised ratio of > or = 2 was not significantly different in the two groups (3 days). Median time to achieve a stable dose was significantly lower in intervention group than in controls (7 days v 9 days, P = 0.01) without excessive overtreatment or undertreatment with anticoagulant. Patients in intervention group spent greater proportion of time in therapeutic range, both as inpatients (59% v 52%) and outpatients (64% v 51%). CONCLUSION: The computerised decision support system was safe and effective and improved the quality of initiation and control of warfarin treatment by trainee doctors. 相似文献

The statistical treatment of condition used in many previous studies has been to determine a predicted weight for a fish, and to calculate a ratio of actual to predicted weight. The ratios obtained, known as condition factor K , are then further analysed using analysis of variance or other techniques to relate changes in K to other independent variables. It is suggested that condition can be studied better by using a single model to analyse the response of fish weight to a number of factors simultaneously. This approach affords benefit from the simplicity of using an integrated analysis and avoids problems with skewed distributions of ratios. An example using data on the Pacific sardine, Sardinops sagax (Jenyns), is given. Changes in condition of this fish due to monthly, yearly and temperature effects are calculated. 相似文献

Sterols from cultured Dictyocha fibula Ehrenberg and Olisthodiscus luteus Carter were extracted and identified for comparison with sterols from other Chromophycota species. Although orientation at C-24 was not absolutely determined, the sterols of Dictyocha, a silicoflagellale, appeared to consist of only 24-methyl-22-dehydrocholesterol and 24-methylenecholesterol, a sterol composition not known in any other alga. This is in keeping with the taxonomic isolation Dictyocha has among algae. On the other hand, Olisthodiscus luteus contained 24-ethylcholesterol, 24-ethylcholestanol, 24-methylcholesterol, and cholesterol. This composition is in accord with sterols of members of the Xanthophyceae and Raphidophyceae. 相似文献
