The inhibition of aromatase, the enzyme responsible for converting androgens to estrogens, is therapeutically useful for the endocrine treatment of hormone-dependent breast cancer. Research by our laboratory has focused on developing competitive and irreversible steroidal aromatase inhibitors, with an emphasis on synthesis and biochemistry of 7α-substituted androstenediones. Numerous 7α-thiosubstituted androst-4-ene-3,17-diones are potent competitive inhibitors, and several 1,4-diene analogs, such as 7α-(4′-aminophenylthio)-androsta-1,4-diene-3,17-dione (7α-APTADD), have demonstrated effective enzyme-activated irreversible inhibition of aromatase in microsomal enzyme assays. One focus of current research is to examine the effectiveness and biochemical pharmacology of 7α-APTADD in vivo. In the hormone-dependent 7,12-dimethylbenz(a)anthracene (DMBA)-induced rat mammary carcinoma model system, 7α-APTADD at a 50 mg/kg/day dose caused an initial decrease in mean tumor volume during the first week, and tumor volume remained unchanged throughout the remaining 5-week treatment period. This agent lowers serum estradiol levels and inhibits ovarian aromatase activity. A second research area has focused on the synthesis of more metabolically stable inhibitors by replacing the thioether linkage at the 7α position with a carbon-carbon linkage. Several 7α-arylaliphatic androst-4-ene-3,17-diones were synthesized by 1,6-conjugate additions of appropriate organocuprates to a protected androst-4,6-diene or by 1,4-conjugate additions to a seco-A-ring steroid intermediate. These compounds were all potent inhibitors of aromatase with apparent Kis ranging between 13 and 19 nM. Extension of the research on these 7α-arylaliphatic androgens includes the introduction of a C1---C2 double bond in the A-ring to provide enzyme-activated irreversible inhibitors. The desired 7α-arylaliphatic androsta-1,4-diene-3,17-diones were obtained from their corresponding 7α-arylaliphatic androst-4-ene-3,17-diones by oxidation with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ). These inhibitors demonstrated enzyme-mediated inactivation of aromatase with apparent kinacts ranging from 4.4 × 10−4 to 1.90 x 10−3 s−1. The best inactivator of the series was 7α-phenpropylandrosta-1,4-diene-3,17-dione, which exhibited a T1/2 of 6.08 min. Aromatase inhibition was also observed in MCF-7 human mammary carcinoma cell cultures and in JAr human choriocarcinoma cell cultures, exhibiting IC50 values of 64-328 nM. The 7α-arylaliphatic androgens thus demonstrate potent inhibition of aromatase in both microsomal incubations and in choriocarcinoma cell lines expressing aromatase enzymatic activity. Additionally, the results from these studies provide further evidence for the presence of a hydrophobic binding pocket existing near the 7α-position of the steroid in the active site of aromatase. The size of the 7α-substituent influences optimal binding of steroidal inhibitors to the active site and affects the extent of enzyme-mediated inactivation observed with androsta-1,4-diene-3,17-dione analogs.  相似文献

The Escherichia coli -galactosidase gene is frequently used as a reporter gene in transgenic studies because its activity can be easily detected at the cellular level. Here we report a procedure for monitoring -galactosidase activity directly in tissue sections, which involves the use of a mixture of ethanol and poly-ethylene-glycol as a fixative (Kryofix) and a special paraffin characterized by a lower fusion point of 42 °C. After embedding and cutting, the sections are stained by the chromogenic substrate 5-bromo-4-chloro-3-indoyl--d galactopyranoside (X-Gal). This procedure allows both the retention of a high level of -galactosidase activity and the preservation of good tissue morphology. Furthermore, it can be combined with immunohistochemical methods to detect other cellular components without compromising reporter gene detection  相似文献

Eleven morphometric and one meristic character in 15 parthenogeneticArtemia populations have been studied by using discriminant andcluster analysis as well as scanning electron microscopy.Discriminant analysis revealed five main groups of morphologicalpatterns: (i) the coastal Chinese populations together with apopulation from Kazakhstan, (ii) the inland Chinese salt lakepopulations, (iii) the Greek populations, (iv) one African populationfrom Namibia and (v) a Chinese population from Xuyu (Jiangsuprovince). To understand the genetic regulation of vegetative to reproductive transition in higher plants, further characterization of the Arabidopsis mutant embryonic flower1, emf1, was conducted. Using three flowering symptoms, we showed that emf1 mutants could only grow reproductive and not rosette shoots under five different growth conditions. The mutant embryos did not produce the typical tunica–corpus shoot apical structures at the heart-, torpedo-, and mature stages. The divergent shoot apical development during mutant and wild-type embryogenesis indicated that the wild-type EMF1 gene was expressed in early embryogenesis. Mutations in the EMF1 gene affected the embryonic shoot apical development and caused the germinating embryo and regenerating callus to grow inflorescence, instead of rosette, shoots. Our results support the hypothesis that the EMF1 gene regulates the switch between vegetative and reproductive growth in Arabidopsis.  相似文献

Abstract An internal fragment representing approximately 85% of sod genes from seven Gram-positive bacteria was amplified by using degenerate primers in a polymerase chain reaction assay. The DNA sequences of sod polymerase chain reaction products from Clostridium perfringens, Enterococcus faecalis, Enterococcus faecium, Lactococcus lactis, Staphylococcus aureus, Streptococcus agalactiae, Streptococcus pneumoniae , and Streptococcus pyogenes were determined. Comparisons of their deduced amino acid sequences with those of the corresponding regions of the SOD proteins from Bacillus stearothermophilus, Listeria monocytogenes , and Streptococcus mutans revealed strong relatedness. Phylogenetic analysis of SOD peptides showed that members of the genera Streptococcus and those of the genera Enterococcus constitute two well-supported monophyletic groups. The method described in this study provides a means for easy recovery of sod genes and the construction of sod mutants of various Gram-positive pathogens.  相似文献

The influence of alternate bearing on nutrient utilization and total tree nutrient content was investigated in mature pistachio (Pistacia vera L. cv Kerman trees). Removal of N, P and Zn in fruit and abscised leaves of cropping (‘on’) trees averaged 5, 6, and 2 times, respectively, the removal in abscised leaflets of the non-fruiting, ‘off’ year trees. One hundred and thirty-five kg N, 131 kg K, 86 kg Ca, 39 kg Mg and 18 kg P per hectare were removed in fruits and abscised leaves in ‘on’ year trees. Tree nutrient contents and, presumably, the size of nutrient storage pools in dormant trees varied between ‘on’ and ‘off’ years. There was 22% and 14% more N and P, respectively, in dormant trees following ‘off’ than ‘on’ years. The greater N and P accumulation in ‘off’ year trees is depleted in support of the large fruit demand for N and P during ‘on’ years. In contrast to N and P, there was greater K and Ca accumulation in perennial tree parts during ‘on’ years than during ‘off’ years. The greater K accumulation in perennial tree parts and approximately 30% greater removal of K in annual organs during ‘on’ than ‘off’ years suggests that K uptake could be 4 times higher in ‘on’ year trees than in (non-cropping), ‘off’ year trees.  相似文献

Ergosterol, a membrane sterol found in fungi but not in plants, was used to estimate live mycelial biomass in ectomycorrhizae. Loblolly pine (Pinus taeda L.) seeds were sown in April 1993 and grown with standard nursery culture practices. Correlations between total seedling ergosterol and visual assessment of mycorrhizal colonization were high during July and August but low as ectomycorrhizal development continued into the growing season. Percentages of mycelial dry weight over lateral roots decreased from 9% in July to 2.5% in November because seedling lateral root dry weight accumulated faster than mycelial dry weight. Total ergosterol per seedling increased from July through February. As lateral root dry weight ceased to increase during winter months, ectomycorrhizal mycelia became the major carbohydrate sink of pine seedlings. No distinctive seasonal pattern of soil ergosterol content was observed. The impact of ectomycorrhizal fungi on plant carbohydrate source-sink dynamics can be quantitatively estimated with ergosterol analysis but not with conventional visual determination.  相似文献
