Control of emerging infectious diseases often hinges on identifying a pathogen reservoir, the source of disease transmission. The potential to function as a pathogen reservoir can be influenced by host lifespan, geographic provenance and phylogeny. Yet, no study has identified factors that causally determine the reservoir potential of diverse host species. We propose the host physiological phenotype hypothesis, which predicts that hosts with short‐lived, poorly defended, nutrient rich and high metabolism tissue have greater values for three epidemiological parameters that determine reservoir potential: host susceptibility to infection, competence to infect vectors and ability to support vector populations. We experimentally tested these predictions using a generalist vectored virus and six wild grass species. Host physiological phenotype explained why hosts differed in all three epidemiological parameters while host lifespan, provenance and phylogeny could not explain host competence. Thus, a single, general axis describing variation in host physiological phenotype may explain reservoir potential.  相似文献

Fast-growing, aerobic, heterotrophic bacteria from the root surface of young sugar beet plants were inventoried. Isolation of the most abundant bacteria from the root surface of each of 1,100 plants between the second and tenth leaf stage yielded 5,600 isolates. These plants originated from different fields in Belgium and Spain. All isolates were characterized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis of total cellular proteins. Comparison of protein fingerprints allowed us to inventory the bacteria of individual plants of different fields or leaf stages and to analyze the composition and variability of the rhizobacterial population of young sugar beet plants. Each field harbored a specific population of bacteria which showed a highly hierarchic structure. A small number of bacteria occurring frequently at high densities dominated in each field. The major bacteria were identified as Pseudomonas fluorescens, Xanthomonas maltophilia, Pseudomonas paucimobilis, and Phyllobacterium sp. The former three species showed a high genetic variability as they were represented by different protein fingerprint types on the same or different fields or leaf stages. Twinspan analysis and relative abundance plots showed that the structure and composition of the bacterial populations varied strongly over time. Pseudomonads were typically early colonizers which were later replaced by X. maltophilia or Phyllobacterium sp.  相似文献

Although formants (vocal tract resonances) can often be observed in avian vocalizations, and several bird species have been shown to perceive formants in human speech sounds, no studies have examined formant perception in birds' own species-specific calls. We used playbacks of computer-synthesized crane calls in a modified habituation—dishabituation paradigm to test for formant perception in whooping cranes ( Grus americana ). After habituating birds to recordings of natural contact calls, we played a synthesized replica of one of the habituating stimuli as a control to ensure that the synthesizer worked adequately; birds dishabituated in only one of 13 cases. Then, we played the same call with its formant frequencies shifted. The birds dishabituated to the formant-shifted calls in 10 out of 12 playbacks. These data suggest that cranes perceive and attend to changes in formant frequencies in their own species-specific vocalizations, and are consistent with the hypothesis that formants can provide acoustic cues to individuality and body size.  相似文献

The classification system of diatoms (Bacillariophyta) is based almost entirely on the organization and structure of their silica cell walls. We have discovered Miocene freshwater fossils that contain valves of two different orders within the same organism. Within frustules of the genus Ellerbeckia Crawford, a genus of the order Melosirales, are contained frustules of the genus Actinocyclus Ehrenberg, a member of the Coscinodiscales. Based on the abundance of each morphotype, they appear to be able to reproduce themselves. The ability of a single diatom taxon to express this degree of morphological variability is unprecedented. These results suggest species concepts in diatoms may be in need of review and challenge the basis of the classification system for this widely distributed and ecologically important group of organisms.  相似文献

Reclamation of highly disturbed lands typically includes establishing fast‐growing, non‐native plants to achieve rapid ground cover for erosion control. Establishing native plant communities could achieve ecosystem functions beyond soil erosion, such as providing wildlife habitat. Pipelines, or other disturbed corridors through a landscape, present unique challenges for establishing native plant communities given the heterogeneity of soil environments and invasive plant propagule pressure. We created two structural equation models to address multiple related hypotheses about the influence of soil pH on plant community composition (current diversity and vegetative cover of the original restoration seed mix and background flora, and invasive plant density during mix establishment and current density) of a highly disturbed landscape corridor restored with native species. To test our hypotheses we conducted a plant survey on a gas pipeline crossing two state forests in the north‐central Appalachians that had been seeded with a native‐based mixture 8 years prior. Low soil pH was a strong predictor of density of the invasive annual plant, Microstegium vimineum, and had resulted in lower species diversity and cover of the seeded mix. Overall, our data provide evidence that native‐based grass and forb mixtures can establish and persist on a wide range of soil environments and thrive in competition with invasive plants in moderately acidic to neutral soils. Advancing knowledge on restoration methods using native species is essential to improving restoration practice norms to incorporate multifunctional ecological goals.  相似文献

Seasonality is known to influence ant activity in many tropical rain forests in the world such as South America and Africa. We surveyed ant fauna in the leaf litter in the locality of Minko'o. The work aimed to evaluate the effect of seasonal variation on the diversity and composition of litter ants. Ants were sampled from November 2015 to June 2017, using four sampling methods: visual capture, bait, pitfall trap and extraction. Species richness, Shannon diversity index and analysis of similarities were used to characterise diversity of ant communities between seasons. We collected 306 ant species, shared out between 56 genera and 11 subfamilies. Subfamilies Myrmicinae, Ponerinae, Dolichoderinae, Formicinae, Dorylinae, Cerapachyinae and Pseudomyrmecinae occurred in all the seasons. Species richness was highest in major dry season with 243 species followed by minor rainy season with 188, major rainy season with 177 species and finally minor dry season that recorded the lowest with 155 species. Kruskal–Wallis test showed that ant species richness did not differ between seasons (p > 0.05). Species diversity index indicated that diversity was the highest during minor dry season (H′ = 4.24), followed by the major dry season (H′ = 4.23), minor rainy season (H′ = 4.21) and lowest during major rainy season (H′ = 4.06). Eight most frequents ants have been recorded: Axinidris sp.1, Camponotus flavomarginatus, Monomorium guineense, Myrmicaria opaciventris, Odontomachus troglodytes, Carebara perpusilla, Paltothyreus tarsatus and Pheidole megacephala. Assessment of the seasonal effect on diversity reveals that dry season is richer and more diverse than rainy season and the season significantly influence the diversity of litter ants.  相似文献

Cellular genomes suffer extensive damage from exogenous agents and reactive oxygen species formed during normal metabolism. The MutT homologs (MutT/MTH) remove oxidized nucleotide precursors so that they cannot be incorporated into DNA during replication. Among many repair pathways, the base excision repair (BER) pathway is the most important cellular protection mechanism responding to oxidative DNA damage. The 8-oxoG glycosylases (Fpg or MutM/OGG) and the MutY homologs (MutY/MYH) glycosylases along with MutT/MTH protect cells from the mutagenic effects of 8-oxoG, the most stable and deleterious product known caused by oxidative damage to DNA. The key enzymes in the BER process are DNA glycosylases, which remove different damaged bases by cleavage of the N-glycosylic bonds between the bases and the deoxyribose moieties of the nucleotide residues. Biochemical and structural studies have demonstrated the substrate recognition and reaction mechanism of BER enzymes. Cocrystal structures of strated the substrate recognition and reaction mechanism of BER enzymes. Cocrystal structures of several glycosylases show that the substrate base flips out of the sharply bent DNA helix and the minor groove is widened to be accessed by the glycosylases. To complete the repair after glycosylase action, the apurinic/apyrimidinic (AP) site is further processed by an incision step, DNA synthesis, an excision step, and DNA ligation through two alternative pathways. The short-patch BER (1-nucleotide patch size) and long-patch BER (2–6-nucleotide patch size) pathways need AP endonuclease to generate a 3′ hydroxyl group but require different sets of enzymes for DNA synthesis and ligation. Protein-protein interactions have been reported among the enzymes involved in BER. It is possible that the successive players in the repair pathway are assembled in a complex to perform concerted actions. The BER pathways are proposed to protect cells and organisms from mutagenesis and carcinogenesis.  相似文献

Phenotypically distinct clinical isolates of Mycobacterium tuberculosis are capable of altering the balance that exists between the pathogen and human host and ultimately the outcome of infection. This study has identified two M. tuberculosis strains (i.e. HN885 and HN1554) among a bank of clinical isolates with a striking defect in phagocytosis by primary human macrophages when compared with strain Erdman, a commonly used laboratory strain for studies of pathogenesis. Mass spectrometry in conjunction with NMR studies unequivocally confirmed that both HN885 and HN1554 contain truncated and more branched forms of mannose-capped lipoarabinomannan (ManLAM) with a marked reduction of their linear arabinan (corresponding mainly to the inner Araf-alpha(1-->5)-Araf unit) and mannan (with fewer 6-Manp residues and more substitutions in the linear Manp-alpha(1-->6)-Manp unit) domains. The truncation in the ManLAM molecules produced by strains HN885 and HN1554 led to a significant reduction in their surface availability. In addition, there was a marked reduction of higher order phosphatidyl-myo-inositol mannosides and the presence of dimycocerosates, triglycerides, and phenolic glycolipid in their cell envelope. Less exposed ManLAM and reduced higher order phosphatidyl-myo-inositol mannosides in strains HN885 and HN1554 resulted in their low association with the macrophage mannose receptor. Despite reduced phagocytosis, ingested bacilli replicated at a fast rate following serum opsonization. Our results provide evidence that the clinical spectrum of tuberculosis may be dictated not only by the host but also by the amounts and ratios of surface exposed mycobacterial adherence factors defined by strain genotype.  相似文献

Infection of hepatocytes by Plasmodium falciparum sporozoites requires the host tetraspanin CD81. CD81 is also predicted to be a coreceptor, along with scavenger receptor BI (SR-BI), for hepatitis C virus. Using SR-BI-knockout, SR-BI-hypomorphic and SR-BI-transgenic primary hepatocytes, as well as specific SR-BI-blocking antibodies, we demonstrate that SR-BI significantly boosts hepatocyte permissiveness to P. falciparum, P. yoelii, and P. berghei entry and promotes parasite development. We show that SR-BI, but not the low-density lipoprotein receptor, acts as a major cholesterol provider that enhances Plasmodium infection. SR-BI regulates the organization of CD81 at the plasma membrane, mediating an arrangement that is highly permissive to penetration by sporozoites. Concomitantly, SR-BI upregulates the expression of the liver fatty-acid carrier L-FABP, a protein implicated in Plasmodium liver-stage maturation. These findings establish the mechanistic basis of the CD81-dependent Plasmodium sporozoite invasion pathway.  相似文献
