Cells that have lost the ability to grow in culture could be defined operationally as either alive or dead depending on the method used to determine cell viability. As a consequence, the interpretation of the state of 'nonculturable' cells is often ambiguous. Escherichia coli K12 cells inactivated by UV-irradiation with a low (UV1) and a high (UV2) dose were used as a model of nonculturable cells. Cells inactivated by the UV1 dose lost 'culturability' but they were not lysed and maintained the capacity to respond to nutrient addition by protein synthesis and cell wall synthesis. The cells also retained both a high level of glucose transport and the capacity for metabolizing glucose. Moreover, during glucose incorporation, UV1-treated cells showed the capacity to respond to aeration conditions modifying their metabolic flux through the Embden-Meyerhof and pentose-phosphate pathways. However, nonculturable cells obtained by irradiation with the high UV2 dose showed several levels of metabolic imbalance and retained only residual metabolic activities. Nonculturable cells obtained by irradiation with UV1 and UV2 doses were diagnosed as active and inactive (dying) cells, respectively.  相似文献

The effects of sodium propionate, acetate, lactate and citrate on cell proliferation, glucose and oxygen consumption, and ATP production in Listeria monocytogenes were investigated in growing and resting cells. Media pH was 6.7-6.8. Growth inhibition increased while glucose consumption continued in the presence of ≥ 1% propionate, ≥ 3% acetate and ≥ 5% lactate in broth during incubation at 35°C, indicating that glucose consumption was uncoupled from cell proliferation. Acetate and propionate were the most effective antilisterials, whereas citrate (5%) was only slightly inhibitory. Of the four salts, only lactate supported growth, oxygen consumption and ATP production. While concentrations of 1 and 5% propionate, acetate and citrate did not have an effect on oxygen consumption, they inhibited ATP production. ATP production in the presence of the four salts was consistently lower at pH 6.0 than at neutral pH. Lactate served as an alternative energy source for L. monocytogenes in the absence of glucose but became toxic to the organism in the presence of the carbohydrate.  相似文献

用Aedans标记肌动蛋白单体G-Actin上Cys374残基作为探针,研究了稀土离子Ce~(3+)与G-Actin的结合及引起的微构象变化。Ce~(3+)在低浓度(Ce~(3+)/Actin摩尔比<1)和Ca~(2+)竞争G-Actin上二价离子的高亲合位点。Ce~(3+)取代Ca~(2+)引起Aedans荧光强度增强与Mg~(2+)取代Ca~(2+)的结果相同。Ce~(3+)/Actin>l则导致Aedans荧光强度下降。说明Ce~(3+)在高低两种浓度条件下结合的位点及对Cvs374的微构象的影响不同。时间分辩测得的Aedans荧光寿命也支持这一结论。CD谱结果表明Ce~(3+)/Actin<0.4,Actin的二级结构增加,大于0.4又导致其失去。Ce~(3+)-Actin在有/无游离ATP时用聚合液诱导的聚合结果表明,无游离ATP时,极低浓度Ce~(3+)促进聚合,高浓度虽有促进但有所减弱;有游离ATP时,Ce~(3+)/Actin在实验范围内促进聚合。  相似文献

Methods used for the regeneration of cofactors in organic media are reviewed. Substratedriven regeneration methods include the use of a second substrate of the same enzyme and the use of a second enzyme and its substrate. The use of mediators in oxidoreductions is described and examples of photochemical and electrochemical regeneration methods are presented. The cellular pathway of sugar uptake in developing cotyledons of Vicia faba L. and Phaseolus vulgaris L. seed was evaluated using a physiological approach. The cotyledon interface with the seed coat is characterised by a specialised dermal cell complex. In the case of Vicia faba cotyledons, the epidermal component of the dermal cell complex is composed of transfer cells. Sucrose is the major sugar presented to the outer surface of both cotyledons and it is taken up from the apoplasm unaltered. Estimated sucrose concentrations within the apparent free space of Vicia and Phaseolus cotyledons were 105 and 113 mM respectively. Rates of in-vitro uptake of [14C]sucrose by cotyledon segments or by whole cotyledons following physical removal or porter inactivation of the outer cells demonstrated that, for both Vicia and Phaseolus cotyledons, the dermal cell complexes are the most intense sites of sucrose uptake. Accumulation of [14C]sucrose in the storage parenchyma of whole cotyledons was directly affected by experimental manipulation of uptake by the outer cell layers and plasmolytic disruption of the interconnecting plasmodesmata. These findings indicated that sucrose accumulated by the dermal cell complexes is transported symplasmically to the storage parenchyma. Overall, it is concluded that the dermal cell complexes of the developing legume embryo, irrespective of the presence or absence of wall ingrowths, are the major sites for the uptake of sucrose released from the maternal tissues to the seed apoplasm.
