The aim of this work was to determine whether laminin (Ln), an extracellular matrix protein, induces the intracellular events that may be involved in producing the acrosome reaction in human sperm. To this end, we evaluated the effect of Ln on tyrosine phosphorylation, intracellular calcium concentration, proteasome activity, and phosphorylation in human sperm. Aliquots of highly motile sperm selected with a Percoll gradient, were incubated with different concentrations of Ln (0-20 μg/ml) for different periods (0-18 h). The percentage of viable acrosome-reacted sperm was evaluated using fluorescein isothiocyanate-labeled Pisum sativum agglutinin and Hoechst 33258 DNA dye. Tyrosine phosphorylation was evaluated by Western blot analysis. The chymotrypsin-like activity of the proteasome was evaluated with a fluorogenic peptide, and intracellular calcium concentration was measured with fura-2. The results indicate that Ln stimulated the acrosome reaction of human sperm in a dose-dependent manner. This increase was drastically inhibited in the presence of herbimycin A, SU6656, and epoxomicin. In addition, Ln increased proteasome activity and phosphorylation; both events were inhibited by herbimycin A and SU6656. Finally, Ln induced an increase in intracellular calcium concentration, which was inhibited by SU6656 and epoxomicin. These results suggest that Ln is able to induce the acrosome reaction. This effect may be mediated by Src kinase and the proteasome, with the consequent induction of a calcium influx.  相似文献

Andean Patagonian lakes are ultraoligotrophic, highly transparent water bodies with very low densities of organisms at the different levels of their trophic food webs. Little is known about spatial distribution and habitat use by fish in these lakes. Hydroacoustic and active sampling techniques were used to study the distribution, composition, and displacement of organisms in an Andean lake throughout the diel cycle on two different moon phase days. Sound scattering layers (SSLs) were found both in the near-shore and pelagic habitats at different times of the day. These SSLs, formed by galaxiid larvae and adults, underwent displacement during the time periods of dawn and dusk, giving rise to the redistribution of organisms in the different habitats. The organisms show high sensitivity to light intensity, displaying different behaviors depending on moon phases. Extensive diel vertical migrations show that the deep pelagic habitats provide diurnal refuge for native galaxiids in Andean Patagonian lakes. Due to the high densities of galaxiid larvae and adults, it is likely that diel migrations generate an active flow of energy and matter between habitats, which could have a profound influence on whole lake dynamics.  相似文献

Mutations in the SLC4A11 protein, reported as a sodium-coup-led borate transporter of the human plasma membrane, are responsible for three corneal dystrophies (CD): congenital hereditary endothelial dystrophy type 2, Harboyan syndrome, and late-onset Fuch's CD. To develop a rational basis to understand these diseases, whose point mutations are found throughout the SLC4A11 sequence, we analyzed the protein biochemically. Hydropathy analysis and an existing topology model for SLC4A1 (AE1), a bicarbonate transporter with the lowest evolutionary sequence divergence from SLC4A11, formed the basis to propose an SLC4A11 topology model. Immunofluorescence studies revealed the cytosolic orientation of N- and C-termini of SLC4A11. Limited trypsinolysis of SLC4A11 partially mapped the folding of the membrane and cytoplasmic domains of the protein. The binding of SLC4A11 to a stilbenedisulfonate inhibitor resin (SITS-Affi-Gel) was prevented by preincubation with H(2)DIDS, with a significantly higher half-maximal effective concentration than AE1. We conclude that stilbenedisulfonates interact with SLC4A11 but with a lower affinity than other SLC4 proteins. Disease-causing mutants divided into two classes on the basis of the half-maximal [H(2)DIDS] required for resin displacement and the fraction of protein binding H(2)DIDS, likely representing mildly misfolded and grossly misfolded proteins. Disease-causing SLC4A11 mutants are retained in the endoplasmic reticulum of HEK 293 cells. This phenotype could be partially rescued in some cases by growing the cells at 30 °C.  相似文献

Crop simulation models can be used to estimate impact of current and future climates on crop yields and food security, but require long‐term historical daily weather data to obtain robust simulations. In many regions where crops are grown, daily weather data are not available. Alternatively, gridded weather databases (GWD) with complete terrestrial coverage are available, typically derived from: (i) global circulation computer models; (ii) interpolated weather station data; or (iii) remotely sensed surface data from satellites. The present study's objective is to evaluate capacity of GWDs to simulate crop yield potential (Yp) or water‐limited yield potential (Yw), which can serve as benchmarks to assess impact of climate change scenarios on crop productivity and land use change. Three GWDs (CRU, NCEP/DOE, and NASA POWER data) were evaluated for their ability to simulate Yp and Yw of rice in China, USA maize, and wheat in Germany. Simulations of Yp and Yw based on recorded daily data from well‐maintained weather stations were taken as the control weather data (CWD). Agreement between simulations of Yp or Yw based on CWD and those based on GWD was poor with the latter having strong bias and large root mean square errors (RMSEs) that were 26–72% of absolute mean yield across locations and years. In contrast, simulated Yp or Yw using observed daily weather data from stations in the NOAA database combined with solar radiation from the NASA‐POWER database were in much better agreement with Yp and Yw simulated with CWD (i.e. little bias and an RMSE of 12–19% of the absolute mean). We conclude that results from studies that rely on GWD to simulate agricultural productivity in current and future climates are highly uncertain. An alternative approach would impose a climate scenario on location‐specific observed daily weather databases combined with an appropriate upscaling method.  相似文献

Root hypoxia produced by flooding or over-irrigation limits stone fruit tree development, particularly in orchards established on soils with restricted drainage. To overcome this problem, stone fruit trees are usually grafted on rootstocks (species or hybrid of the Prunus L. genus) with different degrees of tolerance to root hypoxia. However, the molecular base of such variability is largely unknown. In Arabidopsis thaliana (Heynh.), as well as in a number of crops and tree species, hemoglobin (Hb)-like genes stand out among hypoxia-related genes, but no such studies have been done with the Prunus species used as rootstocks. In this study, we analyzed the expression pattern of class 1 non-symbiotic Hb-like (nsHb) and class 3 truncated Hb-like (trHb) genes in Prunus rootstock roots with different responses to this stress. We observed that the putative Prunus nsHb and trHb genes were induced by root hypoxia in all analyzed Prunus genotypes, independently of their tolerance to hypoxia. However, Prunus nsHb and trHb genes had higher expression levels in roots of tolerant rootstocks. Prunus nsHb and trHb genes were also regulated by other abiotic stresses, such as salt stress and low temperatures. Our results suggest that changes in nsHb and trHb expressions could be part of the adaptive mechanisms that have evolved in the Prunus species to survive under hypoxia or other types of environmental stress that commonly challenge stone fruit tree orchards.  相似文献
