Mutations at the apeA locus in Salmonella typhimurium lead to loss of a soluble enzyme (protease I) that hydrolyzes the chromogenic endoprotease substrate N-acetyl phenylalanine -naphthyl ester. We have isolated pseudorevertants of S. typhimurium apeA mutations that have regained the ability to hydrolyze this compound. These pseudorevertants contain mutations (apeR) that lead to overproduction of a membrane-bound esterase different from protease I. The apeR locus is phage P1 cotransducible with ilvC (83 map units) and is unlinked to apeA. Mutations at still another locus, apeE, lead to loss of the membrane-associated esterase. The apeE locus is P1 cotransducible with purE (12 map units). In an apeE-lacZ operon fusion strain, an apeR mutation increases the level of -galactosidase approximately 60-fold. We propose that apeR encodes a repressor of apeE. The evidence available suggests that the ApeE protein is not a protease. 相似文献

Fifty-eight enterotoxigenic Escherichia coli (ETEC) strains, isolated from children with and without diarrhea in Sao Paulo, were examined for the presence of colonization factor antigens (CFAs) and their ability to adhere to HeLa cells. Antisera to CFA/I, the coli surface (CS) antigens CS1CS3, CS2CS3, and CS2 of CFA/II, CFA/III, and CS5CS6 and CS6 of CFA/IV were used. CFAs were identified in 43% of the ETEC strains: 40% of the strains with CFAs harbored CFA/I, 24% carried CFA/II (CS1CS3), 24% carried CFA/IV (CS6), and 12% carried CFA/IV (CS5CS6). CFAs occurred mainly among ETEC strains producing only heat-stable (ST-I) enterotoxin and in strains also producing heat-labile toxin (LT-I). No ETEC strains tested expressed CFA/III. A marked change in serotypes of ST-I-producing strains was found in Sao Paulo between 1979 and 1990. Adherence to HeLa cells was detected in 14% of the ETEC strains. All of them had a diffuse adherence pattern and produced only ST-I, and 88% carried CS6 antigen. 相似文献

A new approach to insect control—using sodium trichloroacetate (NaTCA) to inhibit synthesis of the hydrophobic cuticular lipids that protect insects from dehydration—was tested on Triatoma infestans. In vivo and in vitro studies of incorporation of radioactive precursors showed diminished cuticular hydrocarbon synthesis after NaTCA treatment. Thin layer chromatography and scanning electron microscopy showed disruption of the cuticular lipid layer of NaTCA-treated insects, which also have increased mortality and altered molting cycles. Triatoma infestans eggs are shown to synthesize hydrocarbons. Radio-gas chromatography was used to demonstrate metabolism of [1-14C]propionate into precursor methyl-branched fatty acids and into methyl-branched hydrocarbons in T. infestans eggs. These reactions have not been demonstrated previously in insect eggs. An in vivo study showed that hydrocarbons are also transported to eggs by the hemolymph. Inhibition of hydrocarbon synthesis by sodium trichloroacetate (NaTCA) was correlated with reduced oviposition, reduced hatchability, and reduced insect survival. Summary As a prelude to complete structure calculations of both the oxidized and reduced forms of Escherchia coli thioredoxin (Mr 11 700), we have analyzed the NMR data obtained for the two proteins under identical conditions. The complete aliphatic 13C assignments for both oxidized and reduced thioredoxin are reported. Correlations previously noted between 13C chemical shifts and secondary structure are confirmed in this work, and significant differences are observed in the C and C shifts between cis- and trans-proline, consistent with previous work that identifies this as a simple and unambiguous method of identifying cis-proline residues in proteins. Reduction of the disulfide bond in the active-site Cys32-Gly-Pro-Cys35 sequence causes changes in the 1H, 15N and 13C chemical shifts of residues close to the active site, some of them quite far distant in the amino acid sequence. Coupling constants, both backbone and side chain, show some differences between the two proteins, and the NOE connectivities and chemical shifts are consistent with small changes in the positions of several side chains, including the two tryptophan rings (Trp28 and Trp31). These results show that, consistent with the biochemical behavior of thioredoxin, there are minimal differences in backbone configuration between the oxidized and reduced forms of the protein. 相似文献

Many aphid species have shown remarkable adaptability by invading new habitats and agricultural crops, although they are parthenogenetic and might be expected to show limited genetic variation. To determine if the mode of reproduction limits the level of genetic variation in adaptively important traits, we assess variation in 15 life history traits of the pea aphid, Acyrhosiphon pisum (Harris), for five populations sampled along a north-south transect in central North America, and for three traits for three populations from eastern Australia. The traits are developmental times and rates as affected by temperature, body weights as affected by temperature, fecundity, measures of migratory tendency, and photoperiodic responses. The most southerly population from North America is shown to be obligately parthenogenetic, as are the Australian populations, and the four more northerly North American populations are facultatively parthenogenetic with the number of parthenogenetic generations per year increasing from north to south. The broad-sense heritabilities of life history traits varied from 0.36 to 0.71 for nine quantitive traits based on a comparison of within-and between-lineage variances. Using these traits, 7–13 distinct genotypes (i.e. clones) were identified among each of the 18 lines sampled from the North American populations, but the number did not differ significantly among populations. The level of genetic variation differed from trait to trait. For 4 of 12 quantitative traits, the level of variation in the obligately parthenogenetic population from North America was lowest, but significantly lower than all the sexual populations for only 1 trait. The obligately parthenogenetic population had the highest level of genetic variation for two traits, and had intermediate levels for the others. The most northerly population, which was sexual and had relatively few parthenogenetic generations each year, had the lowest level of variation for 5 of 12 traits and the highest level of variation for 2 traits. There was no decline in variability from north to south correlated with the increase in the annual number of parthenogenetic generations. The Australian populations showed no less variation than the North American populations for two of three traits, although the pea aphid was introduced to Australia only 5 years prior to the study, whereas the aphid has been in North America for at least 100 years. The mode of reproduction has not had a substantial impact on the level of genetic variation in life history traits of the pea aphid, but there are population-specific factors that effect the level of variation in certain traits. 相似文献

Casein Kinase II Activity in the Postischemic Rat Brain Increases in Brain Regions Resistant to Ischemia and Decreases in Vulnerable Areas 总被引:1,自引:1,他引:0
Abstract: Casein kinase II (CKII) is a protein kinase acting in the intracellular cascade of reactions activated by growth factor receptors, and that has a profound influence on cell proliferation and survival. In this investigation, we studied the changes in the activity and levels of CKII in the rat brain exposed to 10. 15 and 20 min of transient forebrain ischemia followed by variable periods of reperfusion. The cytosolic CKII activity decreased during reperfusion by ∼ 30 and ∼ 50% in the selectively vulnerable areas, striatum and the CA1 region of the hippocampus, respectively. In the resistant CA3 region of hippocampus and neocortex, the activity increased by ∼ 20 and ∼ 60%, respectively. The postischemic changes in CKII activity were dependent on the duration of the ischemic insult. The levels of CKII did not change after ischemia, suggesting that the enzyme is modulated by covalent modification or is interacting with an endogenous inhibitor/activator. Treatment of the cytosolic fraction from cortex of rats exposed to ischemia and 1 h of reperfusion with agarose-bound phosphatase decreased the activity of CKII to control levels, suggesting that CKII activation after ischemia involves a phosphorylation of the enzyme. The correlation between postischemic CKII activity and neuronal survival implies that preservation or activation of CKII activity may be important for neuronal survival after cerebral ischemia. 相似文献

Depression of Neuronal Protein Synthesis Initiation by Protein Tyrosine Kinase Inhibitors 总被引:2,自引:1,他引:1
Abstract— Growth factors stimulate cellular protein synthesis, but the intracellular signaling mechanisms that regulate initiation of mRNA translation in neurons have not been clarified. A rate-limiting step in the initiation of protein synthesis is the formation of the ternary complex among GTP, eukaryotic initiation factor 2 (elF-2), and the initiator tRNA. Here we report that genistein, a specific tyrosine kinase inhibitor, decreases tyrosine kinase activity and the content of phosphotyrosine proteins in cultured primary cortical neurons. Genistein inhibits protein synthesis by >80% in a dose-dependent manner (10–80 μg/ml) and concurrently decreases ternary complex formation by 60%. At the doses investigated, genistein depresses tyrosine kinase activity and concomitantly stimulates PKC activity. We propose that a protein tyrosine kinase participates in the initiation of protein synthesis in neurons, by affecting the activity of elF-2 directly or through a protein kinase cascade. 相似文献

Recent ethnobotanical exploration of henequen (Agave fourcroydes) in the Peninsula of Yucatan, Mexico, finds that inflorescence peduncles are used as emergency food and in the preparation of a fermented drink. Bromatological analysis and determination of total carbohydrates were made for the two length classes (ca. 3.30 m and ca. 0.60 m) which are consumed. The analysis of both the cultivated plant and its putative wild ancestor (Agave angustifolia) suggests that utilization of the inflorescence peduncles as food may have been involved in the initial stages of the history of its evolution under artificial selection, because the wild and the cultivated plants have similar palatability. The subsequent agricultural prevalence of annual crop species in the region was possibly responsible for the abandonment of henequen in the local diet. No significant differences are observed between the bromatological and total carbohydrate values of domesticated and wild plants. The preference for small inflorescence peduncles as a vegetable is a consequence of its significantly minor content of raw fiber and its larger content of total carbohydrates. As a fermented drink, longer peduncles are preferred because they provide more substrate material and because fiber can be eliminated by filtering. This agricultural byproduct, almost totally wasted, has potential value as a source of carbohydrates and raw fiber. 相似文献
