Streptococcus pneumoniae is a common cause of septicemia in the immunocompetent host. To establish infection, S. pneumoniae has to overcome host innate immune responses, one component of which is the complement system. Using isogenic bacterial mutant strains and complement-deficient immune naive mice, we show that the S. pneumoniae virulence factor pneumolysin prevents complement deposition on S. pneumoniae, mainly through effects on the classical pathway. In addition, using a double pspA-/ply- mutant strain we demonstrate that pneumolysin and the S. pneumoniae surface protein PspA act in concert to affect both classical and alternative complement pathway activity. As a result, the virulence of the pspA-/ply- strain in models of both systemic and pulmonary infection is greatly attenuated in wild-type mice but not complement deficient mice. The sensitivity of the pspA-/ply- strain to complement was exploited to demonstrate that although early innate immunity to S. pneumoniae during pulmonary infection is partially complement-dependent, the main effect of complement is to prevent spread of S. pneumoniae from the lungs to the blood. These data suggest that inhibition of complement deposition on S. pneumoniae by pneumolysin and PspA is essential for S. pneumoniae to successfully cause septicemia. Targeting mechanisms of complement inhibition could be an effective therapeutic strategy for patients with septicemia due to S. pneumoniae or other bacterial pathogens. 相似文献

Pneumolysin (PLY) is a major virulence factor released by Streptococcus pneumoniae and has been implicated in the pathogenesis of pneumococcal pneumonia. In this study, we evaluated the contribution of newly recruited neutrophils and monocytes and resident alveolar macrophages to the pathogenesis of PLY-induced lung injury. Mice received either adhesion-blocking Abs to inhibit alveolar leukocyte trafficking or liposomal clodronate to deplete alveolar macrophages before intratracheal application of native PLY or its noncytotoxic derivative PdB. We found that treatment with PLY but not PdB resulted in increased lung vascular permeability. In addition, PLY also induced a decrease in the resident alveolar macrophage population, and the recruitment of peripheral blood neutrophils and monocytes into the alveolar space. Blockade of PLY-induced alveolar leukocyte trafficking by pretreatment of mice with anti-CD18 plus anti-CD49d Abs or depletion of circulating neutrophils did not attenuate the increase in lung permeability observed in response to intratracheal PLY. In addition, depletion of resident alveolar macrophages with clodronated liposomes did not reduce alveolar injury developing in response to PLY. PLY-induced lung injury was associated with only a small increase in bronchoalveolar lavage concentrations of cytokines. These data indicate that PLY-induced lung injury results from direct pneumotoxic effects on the alveolar-capillary barrier and is independent of both resident and recruited phagocytic cells. 相似文献

The immune defense against FMDV has been correlated to the antibody mediated component. However, there are occasions when some animals with high virus neutralising (VN) antibody are not protected following challenge and some with low neutralising antibody which do not succumb to disease. The importance of cell mediated immunity in clinical protection is less clear and so we investigated the source and production of interferon-gamma (IFN-γ) in re-stimulated whole blood of FMDV immunized cattle and its correlation to vaccine induced protection and FMDV persistence. We were able to show a positive correlation between IFN-γ response and vaccine induced protection as well as reduction of long term persistence of FMD virus. When combining this IFN-γ response in re-stimulated blood with virus neutralizing antibody titer in serum on the day of challenge, a better correlation of vaccine-induced protection with IFN-γ and VN antibody was predicted. Our investigations also showed that CD4+ T-cells are the major proliferating phenotype and IFN-γ producing cells. 相似文献

Bioluminescence-based bacterial biosensors are often reported as reliable and efficient tools for risk assessment and environmental monitoring. However, there are few data comparing the metabolism of genetically engineered strains to the corresponding wild type. A pollutant-degrading bacterium capable of mineralising 2,4-dichlorophenoxyacetic acid (2,4-D), Burkholderia sp. strain RASC c2, was genetically engineered to produce light constitutively and tested for assessing the main causes of biodegradation constraint affected by growth rates, toxicity, bioavailability and metal speciation in complex environments. This research focuses on such aspects by characterizing two pollutant-degrading isolates, the wild type and the genetically engineered biosensor (lux-marked). Degradation and growth rates of both isolates were assessed with different concentrations of 2,4-D as the sole carbon source. Kinetic rates were affected by initial concentration of substrate and isolates showed distinct growth rates at different 2,4-D concentrations. Toxic effects of zinc and copper were also comparatively assessed using a dehydrogenase assay and light output. The isolates were sensitive to both metals and at similar EC(50) values. Therefore, bioluminescence response of the lux-marked isolate accurately reflected the toxic response of the parental organism towards zinc and copper, making it an ideal test-organism for assessing toxicity in the context of pollutant mineralization. 相似文献

The effects of topographic position on water regime in a semideciduous tropical forest on Barro Colorado Island in Panamá were assessed by measuring soil matric potential using the filter paper technique and by using measured soil water release characteristics to convert a long-term (20 years) gravimetric water content data-set to matric potential. These were also compared against predictions from a simple water balance model. Soil matric potentials on slope sites were significantly higher than on plateau sites throughout the measurement interval and slopes experienced a shorter duration of drought during the annual dry-season. Measured values of matric potential agreed with those predicted from converting the gravimetric measurements using water release characteristics. Annual duration of drought predicted by the simple water balance model agreed with values determined from the converted long term water content data-set and was able to predict the annual duration of drought on plateau sites. On slope sites, the water balance systematically and significantly overestimated the duration of drought obtained from the water content data-set, suggesting that slope sites were supplied with water from upslope. Predictions of annual drought duration from sites with higher annual rainfall than Barro Colorado Island (BCI), suggest that while plateau sites on BCI experience a water regime consistent with annual rainfall, slopes experience a water regime more similar to that of forests with much higher rainfall. We conclude that such large variations in water regime over small spatial scales may play a role in maintaining high species richness through providing opportunities for niche specialisation and by buffering slopes against possible climate change. 相似文献

Foot-and-mouth disease (FMD) is an extremely infectious viral infection of cloven-hoofed animals which is highly challenging to control and can give rise to national animal health crises, especially if there is a lack of pre-existing immunity due to the emergence of new strains or following incursions into disease-free regions. The 2001 FMD epidemic in the UK was on a scale that initially overwhelmed the national veterinary services and was eventually controlled by livestock lockdown and slaughter on an unprecedented scale. In 2020, the rapid emergence of COVID-19 has led to a human pandemic unparalleled in living memory. The enormous logistics of multi-agency control efforts for COVID-19 are reminiscent of the 2001 FMD epidemic in the UK, as are the use of movement restrictions, not normally a feature of human disease control. The UK experience is internationally relevant as few countries have experienced national epidemic crises for both diseases. In this review, we reflect on the experiences and lessons learnt from UK and international responses to FMD and COVID-19 with respect to their management, including the challenge of preclinical viral transmission, threat awareness, early detection, different interpretations of scientific information, lockdown, biosecurity behaviour change, shortage of testing capacity and the choices for eradication versus living with infection. A major lesson is that the similarity of issues and critical resources needed to manage large-scale outbreaks demonstrates that there is benefit to a ‘One Health’ approach to preparedness, with potential for greater cooperation in planning and the consideration of shared critical resources. 相似文献
