Genetic constitution in the intertidal gastropod Nucella lapillus (L.) influences shell shape, growth rate and physiology. Clinal variation in these traits along a 5 km stretch of coastline in south Devon can be related to environmental variation in temperature and desiccation stress. We have examined mtDNA variation along this shore to investigate whether the cline represents primary or secondary contact. Two distinct mtDNA haplotypes were found which exhibit coincident step clines with karyotypic, allozymic and phenotypic variation and covary with the environmental pressures of temperature and desiccation. These results are interpreted in the context of the wider scale distribution of genetic and phenotypic variation in N. lapillus. It is suggested that the shore studied may represent one of a number of regions of secondary contact within a mosaic hybrid zone in N. lapillus , where coadapted phenotypic variation correlates with habitat and the position of the clines represents an environmental transition. 相似文献

Human alpha-galactosidase A (alpha-Gal A) is the lysosomal glycohydrolase
that cleaves the terminal alpha-galactosyl moieties of various
glycoconjugates. Overexpression of the enzyme in Chinese hamster ovary
(CHO) cells results in high intracellular enzyme accumulation and the
selective secretion of active enzyme. Structural analysis of the N -linked
oligosaccharides of the intracellular and secreted glycoforms revealed that
the secreted enzyme's oligosaccharides were remarkably heterogeneous,
having high mannose (63%), complex (30%), and hybrid (5%) structures. The
major high mannose oligosaccharides were Man5-7GlcNAc2 species.
Approximately 40% of the high mannose and 30% of the hybrid
oligosaccharides had phosphate monoester groups. The complex
oligosaccharides were mono-, bi- , 2,4-tri-, 2,6-tri- and tetraantennary
with or without core-region fucose, many of which had incomplete outer
chains. Approximately 30% of the complex oligosaccharides were mono- or
disialylated. Sialic acids were mostly N -acetylneuraminic acid and
occurred exclusively in alpha2, 3-linkage. In contrast, the intracellular
enzyme had only small amounts of complex chains (7.7%) and had
predominantly high mannose oligosaccharides (92%), mostly Man5GlcNAc2 and
smaller species, of which only 3% were phosphorylated. The complex
oligosaccharides were fucosylated and had the same antennary structures as
the secreted enzyme. Although most had mature outer chains, none were
sialylated. Thus, the overexpression of human alpha-Gal A in CHO cells
resulted in different oligosaccharide structures on the secreted and
intracellular glycoforms, the highly heterogeneous secreted forms
presumably due to the high level expression and impaired glycosylation in
the trans- Golgi network, and the predominately Man5-7GlcNAc2 cellular
glycoforms resulting from carbohydrate trimming in the lysosome.

Microtubules influence morphogenesis by forming distinct geometrical arrays in the cell cortex, which in turn affect the deposition of cellulose microfibrils. Although many chemical and physical factors affect microtubule orientation, it is unclear how cortical microtubules in elongating cells maintain their ordered transverse arrays and how they reorganize into new geometries. To visualize these reorientations in living cells, we constructed a microtubule reporter gene by fusing the microtubule binding domain of the mammalian microtubule-associated protein 4 (MAP4) gene with the green fluorescent protein (GFP) gene, and transient expression of the recombinant protein in epidermal cells of fava bean was induced. The reporter protein decorates microtubules in vivo and binds to microtubules in vitro. Confocal microscopy and time-course analysis of labeled cortical arrays along the outer epidermal wall revealed the lengthening, shortening, and movement of microtubules; localized microtubule reorientations; and global microtubule reorganizations. The global microtubule orientation in some cells fluctuates about the transverse axis and may be a result of a cyclic self-correcting mechanism to maintain a net transverse orientation during cellular elongation. 相似文献

MOTIVATION: Tandem repeats are associated with disease genes, play an important role in evolution and are important in genomic organization and function. Although much research has been done on short perfect patterns of repeats, there has been less focus on imperfect repeats. Thus, there is an acute need for a tandem repeats database that provides reliable and up to date information on both perfect and imperfect tandem repeats in the human genome and relates these to disease genes. RESULTS: This paper presents a web-accessible relational tandem repeats database that relates tandem repeats to gene locations and disease genes of the human genome. In contrast to other available databases, this database identifies both perfect and imperfect repeats of 1-2000 bp unit lengths. The utility of this database has been illustrated by analysing these repeats for their distribution and frequencies across chromosomes and genomic locations and between protein-coding and non-coding regions. The applicability of this database to identify diseases associated with previously uncharacterized tandem repeats is demonstrated. 相似文献

Hendra virus (HeV) and Nipah virus (NiV) are closely related emerging viruses comprising the Henipavirus genus of the Paramyxovirinae, which are distinguished by their ability to cause fatal disease in both animal and human hosts. These viruses infect cells by a pH-independent membrane fusion event mediated by their attachment (G) and fusion (F) glycoproteins. Previously, we reported on HeV- and NiV-mediated fusion activities and detailed their host-cell tropism characteristics. These studies also suggested that a common cell surface receptor, which could be destroyed by protease, was utilized by both viruses. To further characterize the G glycoprotein and its unknown receptor, soluble forms of HeV G (sG) were constructed by replacing its cytoplasmic tail and transmembrane domains with an immunoglobulin kappa leader sequence coupled to either an S-peptide tag (sG(S-tag)) or myc-epitope tag (sG(myc-tag)) to facilitate purification and detection. Expression of sG was verified in cell lysates and culture supernatants by specific affinity precipitation. Analysis of sG by size exclusion chromatography and sucrose gradient centrifugation demonstrated tetrameric, dimeric, and monomeric species, with the majority of the sG released as a disulfide-linked dimer. Immunofluorescence staining revealed that sG specifically bound to HeV and NiV infection-permissive cells but not to a nonpermissive HeLa cell line clone, suggesting that it binds to virus receptor on host cells. Preincubation of host cells with sG resulted in dose-dependent inhibition of both HeV and NiV cell fusion as well as infection by live virus. Taken together, these data indicate that sG retains important native structural features, and we further demonstrate that administration of sG to rabbits can elicit a potent cross-reactive neutralizing antibody response against infectious HeV and NiV. This HeV sG glycoprotein will be exceedingly useful for structural studies, receptor identification strategies, and vaccine development goals for these important emerging viral agents. 相似文献

T-cell receptors (TCRs) are created by a stochastic gene rearrangement process during thymocyte development, generating thymocytes bearing useful, as well as unwanted, specificities. Within the latter group, autoreactive thymocytes arise which are subsequently eliminated via a thymocyte-specific apoptotic mechanism, termed negative selection. The molecular basis of this deletion is unknown. Here, we show that TCR triggering by peptide/MHC ligands activates a caspase in double-positive (DP) CD4+ CD8+ thymocytes, resulting in their death. Inhibition of this enzymatic activity prevents antigen-induced death of DP thymocytes in fetal thymic organ culture (FTOC) from TCR transgenic mice as well as apoptosis induced by anti-CD3epsilon monoclonal antibody and corticosteroids in FTOC of normal C57BL/6 mice. Existing DNA sequence data on the Sry gene, the mammalian sex- determining
locus in the Y chromosome, were analyzed for primates, rodents, and bovids.
In all three taxonomic groups, the terminal sequences evolved faster than
the HMG (high mobility group) boxes, and this applies both to synonymous
(Ks) and nonsynonymous (Ka) nucleotide substitutions. Similar intragenic
correlation between synonymous and nonsynonymous substitution rates was not
found either in other mammalian genes that contain a conservative box (Sox,
Msx) or in the MADS-box genes of plants. The rate of nonsynonymous
substitutions exceeds significantly that of synonymous substitutions in the
terminal Sry sequences of apes. We did not find good support for the
hypothesis that the high evolutionary rate of Sry would be associated with
a promiscuous mating system.

A radiotelemetry apparatus is described for sensing and transmitting electromyograms (EMGs) from free-swimming fish. EMGs are recorded from the epaxial muscles of adult rainbow trout during periods in spontaneous (= routine) activity, and forced-swim, respirometers. When such EMG records are integrated, subjected to spectral analysis, and computer-averaged, the EMG values (in μV) are highly correlated with the fish oxygen consumption during the activity periods. However, there is a marked difference between the regression slopes for oxygen v . EMG value for the data from the spontaneous, and forced-swim, respirometers; the former slope is the steeper. The probable explanation of this phenomenon is that whereas in forced swims the epaxial myomeres are responsible for most of the activity of the fish, in spontaneous activity other muscle systems (e.g. of the lateral, dorsal and ventral fins) come to account for a greater relative proportion of body movement. The difference in slope, although great, is evidently a regular phenomenon. The shift from one regression to the other occurs at a fairly precise epaxial EMG value ( c . 5 μV). This suggests that the laboratory calibration of EMG value in terms of oxygen consumption can be utilized in the wild so that EMG records from free-swimming fish, fitted with telemetry packages can be used to deduce oxygen consumption attributable to activity. It also appears that such records can be used as a guide to the type of activity of the fish, i.e. desultory movements or free cruising. 相似文献
