A Nicotiana tabacum L. SR-1 leaf protoplast system was used to study the effects of dehydroascorbate and glutathione on cellular development. We found that dehydroascorbate is readily taken up by protoplasts and internally reduced to ascorbate. Concomitantly, cell division was inhibited and cell expansion stimulated. In this respect, dehydroascorbate counteracted auxin-mediated leaf protoplast development. In contrast to the effects of dehydroascorbate, glutathione-induced cell dedifferentiation, and this effect is similar to that mediated by high auxin concentrations. We conclude that dehydroascorbate and glutathione affect the auxin-mediated regulation of cellular development. Therefore, the biological role of these compounds extends beyond stress tolerance and defense.  相似文献

Abstract Common features in the induction of pores by various agents are as follows: induction is stochastic and progressive; damage by different agents is often synergistic and limited. The prevention of membrane damage is affected by trivalent and divalent cations, by low pH, by low ionic strength and by high osmotic pressure. The inhibitory role of protons and divalent cations is considered in greater detail: pore-forming agents can be classified into two groups: channels across planar lipid bilayers induced by the first group display voltage-sensitive, reversible inhibition by divalent cations; channels of the second group show voltage-insensitive, irreversible inhibition by divalent cations. A search for the ligands to which divalent cations and protons bind has proved elusive. Comparison with the phenomenon of 'surface conductance' through narrow apertures, that is manifest in the absence of any pore-forming agent, may prove fruitful.  相似文献

A total of 50 patients suffering from acute and chronic bacterial pneumonia were treated with sodium carbenicillin. The bacteriological analysis of the sputum showed that Str. pneumoniae predominated in the monoculture or its association with other bacteria. Connection of the results of the bacteriological analysis with the clinical efficiency of the therapy was studied. When carbenicillin was administered intramuscularly in a dose of 1 gm, its therepeutic effect was maintained in the blood for 4 to 6 hours. A satisfactory clinical effect was registered in 41 out of 50 patients treated with carbenicillin in a daily dose 4--6 gm for 8--10 days. No toxic effect of carbenicillin on the parenchimatous organs and peripheral blood was noted.  相似文献

  1. Many aquatic organisms can consume parasite larvae, thus hampering parasite transmission; however, information about feeding on them in the presence of an alternative prey remains scarce. When having a food choice, predators may decrease parasite consumption, therefore, it is important to assess the role of parasites in the diet of predators in natural communities with different types of prey available. Our study aims to test whether common freshwater cyclopoids feed on trematode free-living stages (cercariae) when an alternative food source is present.
  2. We experimentally studied ingestion rates of cyclopoids Macrocyclops distinctus fed with cercariae of trematode Diplostomum pseudospathaceum, a common and harmful parasite of freshwater fishes, and ciliates Paramecium caudatum (an alternative prey, known as suitable food for copepods). First, the feeding response of cyclopoids to different densities of each prey was studied. Then, feeding selectivity in the mixtures of cercariae and ciliates was tested.

Feeding rates of cyclopoids increased with prey densities (both ciliates and cercariae) but almost stopped growing at high prey densities, which indicated saturation (Holling type II functional response). In most cases, cyclopoids consumed cercariae at higher rates than ciliates. Maximum ingestion rates estimated from the obtained curves were 37 cercariae ind−1 hr−1 and 17 ciliate ind−1 hr−1.
  4. When exposed to prey mixtures, cyclopoids fed on cercariae selectively. When cercariae were offered to cyclopoids at concentrations exceeding the saturation level, the ingestion of ciliates remained constantly low at all ciliate densities. In contrast, the ingestion of cercariae increased with rising cercariae densities even when ciliates were presented ad libitum, decreasing only at very high prey densities. Possible reasons of such feeding preferences are discussed.
  5. Our study demonstrated that cyclopoids may prefer to feed on cercariae when there is an alternative food choice and can ingest cercariae at high rates. These experimental results could be extended to natural communities, suggesting that cyclopoids can reduce the transmission of parasites and contribute to the incorporation of parasite production in food webs of lentic ecosystems.

Dictyostelium myosin has been examined under conditions that reveal intramolecular and intermolecular interactions that may be important in the process of assembly and its regulation. Rotary shadowed myosin molecules exhibit primarily two configurations under these conditions: straight parallel dimers and folded monomers. All of the monomers bend in a specific region of the 1860-A-long tail that is 1200 A from the head-tail junction. Molecules in parallel dimers are staggered by 140 A, which is a periodicity in the packing of myosin molecules originally observed in native thick filaments of muscle. The most common region for interaction in the dimers is a segment of the tail about 200-A-long, extending from 900 to 1100 A from the head-tail junction. Parallel dimers form tetramers by way of antiparallel interactions in their tail regions with overlaps in multiples of 140 A. The folded configuration of the myosin molecules is promoted by phosphorylation of the heavy chain by Dictyostelium myosin heavy chain kinase. It appears that the bent monomers are excluded from filaments formed upon addition of salt while the dimeric molecules assemble. These results may provide the structural basis for primary steps in myosin filament assembly and its regulation by heavy chain phosphorylation.  相似文献

The proteins of the cuticle of adult Ascaris lumbricoides suum were characterized with respect to heterogeneity, glycosylation, and susceptibility to collagenase. Pepsin digestion of intact cuticles was used to determine the extent of stable triple-helical structures of the cuticular components. With sodium dodecyl sulfate-poly-acrylamide gel electrophoresis, it was shown that treatment of purified cuticles with beta-mercaptoethanol released three components (99, 90, and 68 kDa) which comprise 95% of the total solubilized material. The remaining fraction consists of at least four components (16, 28, 154, and 173 kDa). Periodic acid-Schiff staining showed that the only glycoprotein was the 173-kDa component. All cuticular components, except the 173-kDa protein, were degraded by bacterial collagenase. Pepsin digestion of intact cuticles for 24 hr at 4 C produced, after reduction, a 95-kDa fragment; by 96 hr, four fragments (95, 90, 83, and 77 kDa) were evident. When the 96-hr pepsin digest was treated with fresh pepsin, the 77-kDa fragment became the major constituent. With agarose gel electrophoresis, analysis of non-reduced, pepsin-released material revealed intact aggregates that were greater than 2 X 10(3) kDa. The enzyme digestion studies indicate that, with the exception of the 173-kDa component, each cuticular protein contains collagenous domains and that, within the cuticle, the longest contiguous collagen chain in a triple-helical conformation has a uniform molecular size of 77 kDa.  相似文献
