The spatio‐temporal dynamics of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), was investigated to evaluate the effect of the landscape elements and host plants on pest distribution, in an agricultural landscape of 500 ha located in Central Italy. Two farms (farm 1 and farm 2) are located in the experimental area, composing mixed fruit orchards and surrounded by hedgerows, small woodlots, private gardens and cereal fields. Ceratitis capitata population fluctuation was monitored, from 2006 to 2008, using traps baited with trimedlure. Geostatistical methods such as Inverse distance squared weighted were used to obtain distributional maps of adults, mainly males. Results showed that the adult Mediterranean fruit flies were primarily distributed inside farm 1, with the maximum density found in the months of September and October. Away from the principal host plants, particularly in cereal fields, the number of trapped individuals was always low or zero. In both farms, flies were caught sequentially in traps located on host plants (i.e. peach, apple, pear, oriental persimmon and prickly pear) at varying times of maturation, especially when fruits remained on the trees. Distributional maps provided evidence that allowed to identify habitats in which the fly developed early in the season (mixed peach orchards) and afterwards during the periodic flights. 相似文献

Malignant mesothelioma (MM) is an aggressive asbestos-related cancer of the serous membranes. Despite intensive treatment regimens, MM is still a fatal disease, mainly due to the intrinsic resistance to current therapies and the lack of predictive markers and new valuable molecular targets. Protein arginine methyltransferase 5 (PRMT5) inhibition has recently emerged as a potential therapy against methylthioadenosine phosphorylase (MTAP)-deficient cancers, in which the accumulation of the substrate 5'-methylthioadenosine (MTA) inhibits PRMT5 activity, thus sensitizing the cells to further PRMT5 inhibition. Considering that the MTAP gene is frequently codeleted with the adjacent cyclin-dependent kinase inhibitor 2A (CDKN2A) locus in MM, we assessed whether PRMT5 could represent a therapeutic target also for this cancer type. We evaluated PRMT5 expression, the MTAP status and MTA content in normal mesothelial and MM cell lines. We found that both administration of exogenous MTA and stable PRMT5 knock-down, by short hairpin RNAs (shRNAs), selectively reduced the growth of MTAP-deleted MM cells. We also observed that PRMT5 knock-down in MTAP-deficient MM cells reduced the expression of E2F1 target genes involved in cell cycle progression and of factors implicated in epithelial-to-mesenchymal transition. In this paper, we introduce a Bayesian statistical model for the analysis of functional data observed at several time points. Examples of such data include the Michigan growth study where we wish to characterize the shape changes of human mandible profiles. The form of the mandible is often used by clinicians as an aid in predicting the mandibular growth. However, whereas many studies have demonstrated the changes in size that may occur during the period of pubertal growth spurt, shape changes have been less well investigated. Considering a group of subjects presenting normal occlusion, in this paper we thus describe a Bayesian functional ANOVA model that provides information about where and when the shape changes of the mandible occur during different stages of development. The model is developed by defining the notion of predictive process models for Gaussian process (GP) distributions used as priors over the random functional effects. We show that the predictive approach is computationally appealing and that it is useful to analyze multivariate functional data with unequally spaced observations that differ among subjects and times. Graphical posterior summaries show that our model is able to provide a biological interpretation of the morphometric findings and that they comprehensively describe the shape changes of the human mandible profiles. Compared with classical cephalometric analysis, this paper represents a significant methodological advance for the study of mandibular shape changes in two dimensions. 相似文献

c-FLIP (cellular FLICE-like inhibitory protein) protein is mostly known as an apoptosis modulator. However, increasing data underline that c-FLIP plays multiple roles in cellular homoeostasis, influencing differently the same pathways depending on its expression level and isoform predominance. Few and controversial data are available regarding c-FLIP function in autophagy. Here we show that autophagic flux is less effective in c-FLIP−/− than in WT MEFs (mouse embryonic fibroblasts). Indeed, we show that the absence of c-FLIP compromises the expression levels of pivotal factors in the generation of autophagosomes. In line with the role of c-FLIP as a scaffold protein, we found that c-FLIPL interacts with Beclin-1 (BECN1: coiled-coil, moesin-like BCL2-interacting protein), which is required for autophagosome nucleation. By a combination of bioinformatics tools and biochemistry assays, we demonstrate that c-FLIPL interaction with Beclin-1 is important to prevent Beclin-1 ubiquitination and degradation through the proteasomal pathway. Taken together, our data describe a novel molecular mechanism through which c-FLIPL positively regulates autophagy, by enhancing Beclin-1 protein stability.Subject terms: Biochemistry, Autophagy 相似文献

Illegally paid blood donation was a risk factor for HIV acquisition exclusively in Henan and Hubei Provinces of China, and
not in Shanghai. Nucleotide sequences in the gag and env genes of HIV-1 were compared between isolates from Henan and Shanghai regions of China to test whether an expected higher
degree of a common source of infections from this unique blood donation transmission risk would be evident as decreased variation
among Henan isolates in an exploratory cross-sectional analysis. Among 38 isolates studied, 23 of 23 (100%) from Henan and
8 of 15 (54%) from Shanghai were subtype B. In addition, fewer sequence differences were found in gp41 of subtype B isolates
from Henan than from Shanghai isolates. Further studies with additional controls are therefore warranted to confirm the role
of the degree of a common source of infections in differences in HIV variation across populations.

The proteinase inhibitor WSCI, active in inhibiting bacterial subtilisin and a number of animal chymotrypsins, was purified from endosperm of exaploid wheat (Triticum aestivum, c.v. San Pastore) by ion exchange chromatography and its complete amino acid sequence was established by automated Edman degradation. WSCI consists of a single polypeptide chain of 72 amino acid residues, has a molecular mass of 8126.3 Da and a pl of 5.8. The inhibition constants (Ki) for Bacillus licheniformis subtilisin and bovine pancreatic alpha-chymotrypsin are 3.92 x 10(-9) M and 7.24 x 10(-9) M, respectively. The inhibitor contains one methionine and of tryptophan residue and has a high content of essential amino acids (41 over a total of 72 residues), but no cysteines. The primary structure of WSCI shows high similarity with barley subtilisin-chymotrypsin isoinhibitors of the Cl-2 type and with maize subtilisinchymotrypsin inhibitor MPI. Significant degrees of similarity were also found between sequences of WSCI and of other members of the potato inhibitor I family of the serine proteinase inhibitors. The wheat inhibitor WSCI has a single reactive site (the peptide bond between methionyl-48 and glutamyl-49 residues) as identified by affinity chromatography and sequence analysis. 相似文献

Background
Hypertension is the leading cause of death in developed countries and reduction of salt intake is recommended as a key preventive measure.Objective
To assess the dietary sodium and potassium intakes in a national sample of Italian children and adolescents and to examine their relationships with BMI and blood pressure (BP) in the framework of the MINISAL survey, a program supported by the Italian Ministry of Health.Population and Methods
The study population included 1424 healthy subjects (766 boys, 658 girls) aged 6-18 years (mean age: 10.1±2.9) who were consecutively recruited in participating National Health Service centers in 10 Italian regions. Electrolyte intake was estimated from 24 hour urine collections tested for completeness by the concomitant measurement of creatinine content. Anthropometric indices and BP were measured with standardized procedures.Results
The average estimated sodium intake was 129 mmol (7.4 g of salt) per day among boys and 117 mmol (6.7 g of salt) among girls. Ninety-three percent of the boys and 89% of the girls had a consumption higher than the recommended age-specific standard dietary target. The estimated average daily potassium intakes were 39 mmol (1.53 g) and 36 mmol (1.40 g), respectively, over 96% of the boys and 98% of the girls having a potassium intake lower than the recommended adequate intake. The mean sodium/potassium ratio was similar among boys and girls (3.5 and 3.4, respectively) and over 3-fold greater than the desirable level. Sodium intake was directly related to age, body mass and BP in the whole population.Conclusions
The Italian pediatric population is characterized by excessive sodium and deficient potassium intake. These data suggest that future campaigns should focus on children and adolescents as a major target in the framework of a population strategy of cardiovascular prevention. 相似文献70.
