The purpose of this paper is to examine the viewpoints of stakeholders in the management and conservation of farm animal biodiversity in Niger. The research applies Q methodology in order to reveal consensual and divergent discourses. After the development of the set of items on the topic of biodiversity (Q sample), the statements were sorted by the respondents through a 7-grade scale, from ?3 to +3. The analysis of Q-sort data with the qmethod package under the R software highlighted three distinct stakeholder viewpoints on the importance of biodiversity in agriculture and animal husbandry, the balance between progress and preservation and the effectiveness of different methods of conservation. The study shows an apparent consensus on the importance of biodiversity that is obviously a promoted topic in the country. Behind the consensus, different discourses are defined that all appear divided by the same dilemma between conservation and economic development. Understanding the different answers and weight attributed to each of the components of the dilemma will guide awareness-raising campaigns and help to pinpoint divergent interests among stakeholders. 相似文献

In Aedes mosquitoes, infections with arthropod-borne viruses (arboviruses) trigger or modulate the expression of various classes of viral and host-derived small RNAs, including small interfering RNAs (siRNAs), PIWI interacting RNAs (piRNAs), and microRNAs (miRNAs). Viral siRNAs are at the core of the antiviral RNA interference machinery, one of the key pathways that limit virus replication in invertebrates. Besides siRNAs, Aedes mosquitoes and cells derived from these insects produce arbovirus-derived piRNAs, the best studied examples being viruses from the Togaviridae or Bunyaviridae families. Host miRNAs modulate the expression of a large number of genes and their levels may change in response to viral infections. In addition, some viruses, mostly with a DNA genome, express their own miRNAs to regulate host and viral gene expression. Here, we perform a comprehensive analysis of both viral and host-derived small RNAs in Aedes aegypti Aag2 cells infected with dengue virus 2 (DENV), a member of the Flaviviridae family. Aag2 cells are competent in producing all three types of small RNAs and provide a powerful tool to explore the crosstalk between arboviral infection and the distinct RNA silencing pathways. Interestingly, besides the well-characterized DENV-derived siRNAs, a specific population of viral piRNAs was identified in infected Aag2 cells. Knockdown of Piwi5, Ago3 and, to a lesser extent, Piwi6 results in reduction of vpiRNA levels, providing the first genetic evidence that Aedes PIWI proteins produce DENV-derived small RNAs. In contrast, we do not find convincing evidence for the production of virus-derived miRNAs. Neither do we find that host miRNA expression is strongly changed upon DENV2 infection. Finally, our deep-sequencing analyses detect 30 novel Aedes miRNAs, complementing the repertoire of regulatory small RNAs in this important vector species. 相似文献

Escherichia coli B/pTG201 recombinant cells were immobilized by entrapment in a carrageenan gel and cultivated in nonselective media to investigate the effect of agitation rate on plasmid stability, biomass concentration, and enzyme productivity. These parameters were studied in continuous cultures for free and immobilized cells, respectively. Immobilized recombinant cells exhibit an increase in the stability of the plasmid pTG201 compared to free cells, even under conditions where the tendency of plasmid stability for free cells decreased generally more rapidly under a higher agitation rate. Intensive agitation, resulting also in a strong shear stress, greatly reduced cell concentration within gel beads throughout the course of growth. Higher enzyme expression of catechol 2–3, dioxygenase was also obtained in leaked cells due to better maintenance of plasmid stability and higher plasmid copy number with regard to free cells. Enzyme productivity of leaked and free cells in minimal medium decreased with the increase in agitation rate, due to decreased plasmid stability; however, in LB medium, it increased in the presence of higher agitation rate related to important cell concentration. 相似文献

The way misses happen in oxygen evolution is subject to debate (Govindjee et al. 1985). We recently observed a linear lowering of the miss probability with the flash number (Meunier and Popovic 1989). Therefore, we investigated in Dunaliella tertiolecta the link between the average miss probability and the redox state of plastoquinone after n flashes. The effect of flashes was to oxidize the plastoquinone pool; we found that the oxidation of plastoquinone highly correlated (linear regression: R
2=0.996) with the lowering of the miss probability. The flash frequency was found to affect both the miss probability and the redox state of plastoquinone. When pre-flashes were given using a high flash frequency (10 Hz), the plastoquinone pool was oxidized and misses were low; however, if long dark intervals between flashes were used, the oxidizing effect of flashes was lost and the misses were high. We could not explain our results by assuming equal misses over all S-states; but unequal misses, caused by deactivations, were coherent with our results. We deduced that chlororespiration was responsible for the reduction of plastoquinone in the dark interval between flashes. We compared oxygen evolution with and without benzoquinone, using a low flash frequency (0.5 Hz) for maximum misses. Benzoquinone lowered the misses from 34% to 3%, and raised the amplitude of oxygen evolution by more than a factor of two (2). From this we deduced that the charge carrier C postulated to explain misses (Lavorel and Maison-Peteri 1983) did not account for more than 3% of miss probability in Dunaliella tertiolecta. Summary A cell entrapment process using -carrageenan — locust bean gum gel is presented. Streptococcus thermophilus, Lactobacillus bulgaricus and S. lactis were immobilized in small gel beads (0.5–1.0 mm and 1.0–2.0 mm diameter) and fermentations in bench bioreactors were conducted. Viability of entrapped cells, lactose utilization, lactic acid production and cell release rates were measured during fermentation. The procedure was effective for S. thermophilus, L. bulgaricus and S. lactis, and the viability of these bacteria remained very high throughout entrapment steps and subsequent storage. Bead diameter influenced the fermentation rate: smaller beads (0.5–1.0 mm) permitted an increase in release rates, lactose utilization and acid production by entrapped cells, approximating values attained with free cells. 相似文献
