The Emerson-Trinder reaction has been optimized in this work using an initial rate spectrophotometric method and response surface methodology (RSM). In this investigation, the variation range of critical variables along with the fixed parameters were selected based on a preliminary 'one at a time' (OVAT) procedure for the subsequent RSM chemometric analysis as follows: pH (6-10), buffer concentration (50-250 mM), 4-aminoantipyrine (4-AAP) concentration (1-5 mM), temperature (25-45°C). The optimum values of fixed parameters were: 4-fluorophenol (4-FP, 30 mM), horseradish peroxidase (HRP) enzyme activity (0.12 U mL-1), and the fixed concentration of the H2O2 in the chemometric experiments was 11.4 µM. The non-linear nature of the experimental response of the reaction system was explained by a second-order polynomial equation, which revealed the impact of the experimental factors, their interactions and also their optimum values. The results of the reported RSM analysis proved to be quite appropriate for the design and optimization of this reaction, as illustrated by the relatively high value of the determination coefficient (R2=96.7%) for the fitting of quadratic model, along with the satisfactory results generated by the analysis of variance (ANOVA). All the evaluated analytical characteristics of this method: typical reaction progress curves, resulting linear calibration curve, within-day precisions at low and at high levels, and the upper and lower detection limits were, also, reported. Hordem murinum is a widespread weedy/wild species, growing in different ecological conditions in Iran. Populations of H. murinum subsp. glaucum (2n = 2x = 14) and subsp. leporinum (2n = 4x, 6x = 28, 42) are found in this region. Inter-retroelement amplified polymorphism (IRAP) markers were used to analyse the genetic diversity of 57 accessions of H. murinum from different regions of Iran, and to examine patterns of diversity related to the taxonomy and geography. Eight IRAP primer combinations amplified a total of 241 distinct DNA fragments sized 150-1400 bp, from which 236 (97.9%) were polymorphic. On average, each primer combination amplified about 30.12 fragments (ranged from 23 to 34) in PCR. The patterns of genetic diversity were closely correlated with taxonomic groups, ploidy levels and geographic origin. Along with the high genetic diversity, three geographic sub-genepools were evident, 1: in the North-Northeast region along the Alborz Mountains, 2: in the West-Northwest region along the Zagros Mountains, and 3: in the Central — Southern region. The detailed structural basis for the cryptic nature (crypticity) of a B cell epitope harbored by an autoantigen is unknown. Because the immune system may be ignorant of the existence of such "cryptic" epitopes, their exposure could be an important feature in autoimmunity. Here we investigated the structural basis for the crypticity of the epitopes of the Goodpasture autoantigen, the alpha3alpha4alpha5 noncollagenous-1 (NC1) hexamer, a globular domain that connects two triple-helical molecules of the alpha3alpha4alpha5 collagen IV network. The NC1 hexamer occurs in two isoforms as follows: the M-isoform composed of monomer subunits in which the epitopes are accessible to autoantibodies, and the D-isoform composed of both monomer and dimer subunits in which the epitopes are cryptic. The D-isoform was characterized with respect to quaternary structure, as revealed by mass spectrometry of dimer subunits, homology modeling, and molecular dynamics simulation. The results revealed that the D-isoform contains two kinds of cross-links as follows: S-hydroxylysyl-methionine and S-lysyl-methionine cross-links, which stabilize the alpha3alpha5-heterodimers and alpha4alpha4-homodimers, respectively. Construction and analysis of a three-dimensional model of the D-isoform of the alpha3alpha4alpha5 NC1 hexamer revealed that crypticity is a consequence of the following: (a) sequestration of key residues between neighboring subunits that are stabilized by domain-swapping interactions, and (b) by cross-linking of subunits at the trimer-trimer interface, which stabilizes the structural integrity of the NC1 hexamer and protects against binding of autoantibodies. The sequestrated epitopes and cross-linked subunits represent a novel structural mechanism for conferring immune privilege at the level of quaternary structure. population genetics for the evolution of ageing in the 1970’s. In the 1980’s, experiments using Drosophila showed that the rate of ageing evolves as predicted by Hamilton’s’ forces of natural selection’. The discovery of the cessation
of ageing late in life in the 1990’s was followed by its explanation in terms of evolutionary theory based on Hamilton’s forces.
Recently, it has been shown that the cessation of ageing can also be manipulated experimentally using Hamilton’s’ forces of

Nitric oxide (NO) is dynamic molecule implicated in diverse biological functions demonstrating its protective effect against damages provoked by abiotic stresses. The present study investigated that exogenous NO pretreatment (500?µM sodium nitroprusside, 24?h) prevented the adverse effect of drought stress [induced by 10% and 20% polyethylene glycol (PEG), 48?h] on rapeseed seedlings. Drought stress resulted in reduced relative water content with increased proline (Pro) level. Drought stress insisted high H2O2 generation and consequently increased membrane lipid peroxidation which are clear indications of oxidative damage. Drought stress disrupted the glyoxalase system too. Exogenous NO successfully alleviated oxidative damage effects on rapeseed seedlings through improving the levels of nonenzymatic antioxidant pool and upregulating antioxidant enzymes’ activities. Improvement of glyoxalase system (glyoxalase I and glyoxalase II activities) by exogenous NO was significant to improve plants’ tolerance. The objective of this work was to examine the time-dependent pro-oxidant versus antioxidant effect of various doses of vitamin E used commonly in experimental studies. Erythrocyte activity of superoxide dismutase (SOD), glutathione peroxidase (GPX), catalase (CAT) and plasma lipid peroxidation levels were investigated following biweekly intramuscular administration of 100, 300 and 600 mg/kg of vitamin E at a baseline time point, and additionally at 2, 4 and 6 weeks after initiating treatment. Vitamin E had an antioxidant effect when administered at low doses over short time periods, and increased the activity of antioxidant enzymes. At higher doses and over longer time periods, it increased the level of lipid peroxidation, and attenuated the activity of antioxidant enzymes. These results suggest that time-dependent variations in vitamin E effects should be considered in design and interpretation of experimental antioxidant studies, as well as during clinical trials. 相似文献

SCO6571 protein from Streptomyces coelicolor A3(2) was overexpressed and purified using Rhodococcus erythropolis as an expressing host. Crystals of selenomethionine-substituted SCO6571 have been obtained by vapor diffusion method. SCO6571 crystals diffract to 2.3 A and were found to belong to the orthorhombic space group P2(1)2(1)2(1) with unit cell parameters a = 84.5, b = 171.6, c = 184.8 A. Six molecules in the asymmetric unit give a crystal volume per protein mass (V(M)) of 2.97 A (3) Da(-1) and solvent content of 58.6 %. The structure was solved by the single wavelength anomalous diffraction (SAD) method. SCO6571 is a TIM-barrel fold protein that assembles into a hexameric molecule with D(3) symmetry. 相似文献
