The complete amino acid sequence of mohair protein, SCMKB-M1.2 (97 residues), was determined. The protein was isolated from reduced and carboxymethylated mohair by chromatography on DEAE-cellulose phosphate. Peptides for sequence determination were obtained by digestion with trypsin, pepsin, chymotrypsin, thermolysin and papain, and were fractionated by DEAE-cellulose chromatography, paper chromatography and electrophoresis. The sequence of the peptides were determined by the Edman degradation method (by use of both the Beckman Sequence and a non-automatic procedure), and by partial acid hydrolysis. The protein is closely homologous to wool protein SCMKB-IIIB2, and also contains acetylated alanine as N-terminal amino acid.  相似文献

Summary The habitat hectares approach is an explicit, quantitative method for assessing the quality of vegetation by adding scores that are assigned to 10 habitat attributes. We believe it will be more repeatable and transparent than other methods that rely on subjective judgement. However, we have four principal criticisms of the method as it is currently proposed: (i) measurement of some of the attributes may be subject to considerable error that varies among assessors; (ii) the comparison of each measure with a single benchmark does not accommodate appropriate disturbance regimes; (iii) the proposed combination of attributes leads to some apparent internal inconsistencies; and (iv) it is not clear how the method will actually be used in practice. We suggest modifications to address these concerns and improve the proposed method. Finally, we make additional suggestions about the method's potential application, including: separate reporting of the extent and quality of different vegetation types to avoid the inappropriate combination of measures of area and quality; valuing appropriate disturbance regimes in natural areas; and considering very carefully the application of compensation or mitigation measures.  相似文献

Using a minicircle DNA primer-template, the wild-type catalytic subunit of herpes simplex virus type 1 (HSV-1) DNA polymerase (pol) was shown to lack significant strand displacement activity with or without its processivity factor, UL42. However, an exonuclease-deficient (exo(-)) pol (D368A) was capable of slow strand displacement. Although UL42 increased the rate (2/s) and processivity of strand displacement by exo(-) pol, the rate was slower than that for gap-filling synthesis. High inherent excision rates on matched primer-templates and rapid idling-turnover (successive rounds of excision and polymerization) of exo-proficient polymerases correlated with poor strand displacement activity. The results suggest that the exo activity of HSV-1 pol modulates its ability to engage in strand displacement, a function that may be important to the viability and genome stability of the virus.  相似文献

Spatio‐temporal studies of hybrid zones provide an opportunity to test evolutionary hypotheses of hybrid zone maintenance and movement. We conducted a landscape genetics study on a classic hybrid zone of the south‐eastern Australian frogs, Litoria ewingii and Litoria paraewingi. This hybrid zone has been comprehensively studied since the 1960s, providing the unique opportunity to directly assess changes in hybrid zone structure across time. We compared both mtDNA and male advertisement call data from two time periods (present and 1960s). Clinal analysis of the coincidence (same center) and concordance (same width) of these traits indicated that the center of the hybrid zone has shifted 1 km south over the last 40 years, although the width of the zone and the rate of introgression remained unchanged. The low frequency of hybrids, the strong concordance of clines within a time period, and the small but significant movement across the study period despite significant anthropogenic changes through the region, suggest the hybrid zone is a tension zone located within a low‐density trough. Hybrid zone movement has not been considered common in the past but our findings highlight that it should be considered a crucial component to our understanding of evolution.  相似文献

Maternal effects have the potential to affect population dynamics and evolution. To affect population dynamics, maternal effects must influence offspring vital rates (birth, death, or movement). Here, we explore the magnitude of nongenetic maternal influence on the vital rates of an insect herbivore and explore predictability of maternal effects with reference to published studies. We experimentally studied the effects of maternal age, host plant species (two Asclepias spp.), and density on offspring vital rates in Aphis nerii, the oleander aphid. Older mothers produced offspring that lived shorter lives, consistent with the "Lansing Effect." Older mothers also produced offspring that matured at a younger age. As maternal age increased, offspring mass at maturity decreased when mothers were on Asclepias syriaca. However, offspring mass was highest from intermediate aged mothers on A. viridis. The absence of maternal density effects seems to exclude maternal density as a potential source of delayed density dependence in A. nerii. Our results indicate that maternal effects have some influence on A. nerii vital rates. However, references to published studies suggest that only the Lansing Effect is a predictable response to maternal age in insects. Moreover, the magnitude of observed effects was generally low.  相似文献

The bioacoustic attributes of vocalisations made by birds in urban environments often differ markedly from those of rural conspecifics. Whether such differences are result from genetic divergence between urban and rural populations, or from plasticity or cultural evolution of song remains poorly understood. Silvereyes (Zosterops lateralis) show evidence of acoustic adaptation to urban noise, modifying both their songs and calls in cities when compared to rural areas. We investigated whether these differences were associated with corresponding morphological and neutral genetic differences. Across six pairs of geographically separate urban and rural populations, all morphological traits measured were similar. Furthermore, genetic analyses of variation at nine microsatellite loci revealed high levels of genetic connectivity between populations, and similar levels of heterozygosity in both habitat types. Consistent directional shifts in song attributes of city birds across large geographic areas thus do not appear to be accompanied by associated morphological or neutral genetic divergence.  相似文献

Asymmetric competition in larval amphibians can influence population dynamics and community structure. This density-dependent regulatory mechanism may be of particular importance for rare or endangered species such as the northern crawfish frog, Rana areolata circulosa. Interspecific competition of R. areolata with two congenerics, R. blairi and R. sphenocephala, was examined in artificial ponds. Analysis of covariance (differential mortality covariate) indicated that interspecific competition increased larval period length and decreased metamorphic body mass of R. areolata. The number of metamorphs produced was lower for R. blairi ponds when reared with R. areolata at high density. Body mass at metamorphosis was larger for R. sphenocephala when reared with R. areolata, suggesting that R. areolata facilitates larval growth in R. sphenocephala. These results indicate that the larval performance of R. areolata was reduced in the presence of interspecific competitors. Although many conservation efforts emphasize the preservation of critical habitat or particular rare species, interactive effects of biotic components in the focal community may also be important demographic regulators. Artificial crossing using Rana blairi and R. sphenocephala frogs produced conspecific, interspecific and F1 backcross hybrid genotypes. Although hybrid males used in the crosses were sterile, crosses using hybrid females produced viable larvae. The larval performance of resultant parental and hybrid genotypes was measured in experimental ponds at two densities. Density significantly affected survival, body mass at metamorphosis, larval period length and metamorphosis for all genotypes. Survival was the same among genotypes, but decreased with increasing density. Body mass at metamorphosis was the same among genotypes, but decreased with increasing density. Larval period increased with increasing density. Among genotypes, larvae from the conspecific R. sphenocephala cross had the shortest larval period while larvae from the conspecific R. blairi cross had the longest larval period. All hybrid genotypes had larval periods longer than R. sphenocephala, but shorter than R. blairi. The percentage of individuals metamorphosing was highest for R. sphenocephala ponds and lowest for R. blairi ponds across densities. Ponds with hybrid larvae produced a greater proportion of metamorphs than those with R. blairi larvae, but a smaller proportion than R. sphenocephala ponds. Equivalent or increased relative larval performance of hybrid genotypes under the conditions of our experiment suggests that hybrid genotypes may possess similar or higher fitnesses relative to their progenitors in some environments. Reduced fertility of adult hybrid males is a powerful selective force against natural hybridization. Nevertheless, because of the successful reproduction by female hybrids, natural hybridization has the potential to serve as a mechanism for the introgression of novel genetic variation that can benefit both R. blairi and R. sphenocephala in fluctuating and unpredictable larval environments. Experimental determination of the fitness of parental and hybrid genotypes is crucial for a comprehensive understanding of the effects of hybridization on organismal evolution.  相似文献
