Matrilysin (matrix metalloproteinase-7) is expressed by mucosal epithelia throughout the body and functions in host defense by activating murine intestinal alpha-defensins. In normal adult human lung, matrilysin is expressed at low levels in the airway epithelium, but is markedly up-regulated in cystic fibrosis (CF). Because CF lungs support a heavy bacterial load, we assessed if relevant CF pathogens regulate matrilysin expression in human lung epithelial cells. Indeed, acute infection with Pseudomonas aeruginosa (but not Staphylococcus aureus, Haemophilus influenzae, or Klebsiella pneumoniae) induced the expression of matrilysin in Calu-3 lung epithelial cells. Increased matrilysin mRNA levels were detectable at 3 h post-infection and peaked at a 25-fold induction between 6 and 8 h. Both P. aeruginosa CF isolates and laboratory strains induced matrilysin expression to similar levels. Flagellin, the monomeric precursor of bacterial flagella, was identified as the inductive factor released by P. aeruginosa that regulated matrilysin expression. In addition, flagellin-null mutants failed to stimulate matrilysin expression in cultured cells or in lungs infected in vivo. These data show that P. aeruginosa (and specifically flagellin) potently stimulates matrilysin expression in lung epithelial cells and may mediate the overexpression of this proteinase in CF lungs.  相似文献

Unlike most marine teleosts, the coral reef-dwelling spiny damsel, Acanthochromis polyacanthus, lacks a pelagic larva dispersal phase and represents one of few examples of self recruitment onto a natal reef by a marine teleost immediately after hatching. Benthic eggs are protected by the parents, and upon hatching the young remain under parental care for several months. Visual morphogenesis of spiny damsel embryos and juveniles was examined to evaluate the potential visual capabilities of the young after emergence onto the reef. The optic primordia were visible in the embryo as hollow spheres of undifferentiated neuroblasts 2 days after fertilization (daf). Visual morphogenesis proceeded rapidly thereafter in the embryo such that at hatching (between 10 and 12daf) gross visual morphology was consistent with that reported in the majority of juvenile marine teleosts, reflecting direct development of the retina of the spiny damsel within the egg. At hatching, the outer nuclear layer comprised 2 classes of photoreceptors; cones and rods. Tangential sections of the retina revealed a square cone mosaic in which 4 double cones surrounded a single cone. This arrangement remained unchanged in all later life history intervals examined. Absolute eye size was large compared to larvae of marine pelagic spawners. Eye and lens diameters increased from 0.69 and 0.23mm, respectively, on the day of hatching (12daf), to 3.77 and 1.52mm, respectively, in a fish 131daf. Angular density of cones increased from 0.25 cones 10 visual arc–1 in an embryo 8daf, to 1.14 cones 10 visual arc–1 in a fish 131daf, demonstrating the potential for significant increase in spatial resolution with increasing eye size. Convergence ratios of cones to ganglion cells remained relatively constant from the time of hatching, suggesting that the determinate ganglion cell photopic receptive field was established early in development. The increase in the convergence ratios of rods: ganglion cells from 1.4 in the late stages of embryogenesis (10daf; 2 days prior to hatching) to 4.9 in a fish 103daf, demonstrated increasing scotopic ganglion cell receptive field size, with increasing age. This was a result of rod cell addition with growth. An increase in the angular density of rods from 0.18 rods 10 visual arc–1 in an embryo 8daf, to 4.07 rods 10 visual arc–1 in a fish 131daf, and the increase in mean scotopic light path-length from 13.3±1.1m in an embryo 8dpf, to 55±5.2m a fish 22dpf, collectively indicate the potential for increasing scotopic sensitivity during growth. On the basis of visual morphology it is predicted that newly hatched spiny damsel juveniles have substantially greater visual capabilities than first feeding larvae with a pelagic dispersal phase. In addition, we propose that the developmental trajectory of the spiny damsel is different from that of pelagic dispersing larvae and does not simply reflect displacement along a common developmental continuum by an extended embryonic duration.  相似文献

Hospital tap water is a recognised source of Pseudomonas aeruginosa. UK guidance documents recommend measures to control/minimise the risk of P. aeruginosa in augmented care units but these are based on limited scientific evidence. An experimental water distribution system was designed to investigate colonisation of hospital tap components. P. aeruginosa was injected into 27 individual tap ‘assemblies’. Taps were subsequently flushed twice daily and contamination levels monitored over two years. Tap assemblies were systematically dismantled and assessed microbiologically and the effect of removing potentially contaminated components was determined. P. aeruginosa was repeatedly recovered from the tap water at levels above the augmented care alert level. The organism was recovered from all dismantled solenoid valves with colonisation of the ethylene propylene diene monomer (EPDM) diaphragm confirmed by microscopy. Removing the solenoid valves reduced P. aeruginosa counts in the water to below detectable levels. This effect was immediate and sustained, implicating the solenoid diaphragm as the primary contamination source.  相似文献

The purpose of the present study was to test the hypothesis that lecithin:cholesterol acyltransferase (LCAT) deficiency would accelerate atherosclerosis development in low density lipoprotein (LDL) receptor (LDLr-/-) and apoE (apoE-/-) knockout mice. After 16 weeks of atherogenic diet (0.1% cholesterol, 10% calories from palm oil) consumption, LDLr-/- LCAT-/- double knockout mice, compared with LDLr-/- mice, had similar plasma concentrations of free (FC), esterified (EC), and apoB lipoprotein cholesterol, increased plasma concentrations of phospholipid and triglyceride, decreased HDL cholesterol, and 2-fold more aortic FC (142 +/- 28 versus 61 +/- 20 mg/g protein) and EC (102 +/- 27 versus 61+/- 27 mg/g). ApoE-/- LCAT-/- mice fed the atherogenic diet, compared with apoE-/- mice, had higher concentrations of plasma FC, EC, apoB lipoprotein cholesterol, and phospholipid, and significantly more aortic FC (149 +/- 62 versus 109 +/- 33 mg/g) and EC (101 +/- 23 versus 69 +/- 20 mg/g) than did the apoE-/- mice. LCAT deficiency resulted in a 12-fold increase in the ratio of saturated + monounsaturated to polyunsaturated cholesteryl esters in apoB lipoproteins in LDLr-/- mice and a 3-fold increase in the apoE-/- mice compared with their counterparts with active LCAT. We conclude that LCAT deficiency in LDLr-/- and apoE-/- mice fed an atherogenic diet resulted in increased aortic cholesterol deposition, likely due to a reduction in plasma HDL, an increased saturation of cholesteryl esters in apoB lipoproteins and, in the apoE-/- background, an increased plasma concentration of apoB lipoproteins.  相似文献

Human epithelial cells infected with the parainfluenza virus simian virus 5 (SV5) show minimal activation of host cell interferon (IFN), cytokine, and cell death pathways. In contrast, a recombinant SV5 P/V gene mutant (rSV5-P/V-CPI-) overexpresses viral gene products and is a potent inducer of IFN, proinflammatory cytokines, and apoptosis in these cells. In this study, we have compared the outcomes of wild-type (WT) SV5 and rSV5-P/V-CPI- infections of primary human dendritic cells (DC), important antigen-presenting cells for initiating adaptive immune responses. We have tested the hypothesis that a P/V mutant which activates host antiviral responses will be a more potent inducer of DC maturation and function than WT rSV5, which suppresses host cell responses. Infection of peripheral blood mononuclear cell-derived immature DC with WT rSV5 resulted in high levels of viral protein and progeny virus but very little increase in cell surface costimulatory molecules or secretion of IFN and proinflammatory cytokines. In contrast, immature DC infected with the rSV5-P/V-CPI- mutant produced only low levels of viral protein and progeny virus, but these infected cells were induced to secrete IFN-alpha and other cytokines and showed elevated levels of maturation markers. Unexpectedly, DC infected with WT rSV5 showed extensive cytopathic effects and increased levels of active caspase-3, while infection of DC with the P/V mutant was largely noncytopathic. In mixed-culture assays, WT rSV5-infected DC were impaired in the ability to stimulate proliferation of autologous CD4+ T cells, whereas DC infected with the P/V mutant were very effective at activating T-cell proliferation. The addition of a pancaspase inhibitor to DC infected with WT rSV5 reduced cytopathic effects and resulted in higher surface expression levels of maturation markers. Our finding that the SV5 P/V mutant has both a reduced cytopathic effect in human DC compared to WT SV5 and an enhanced ability to induce DC function has implications for the rational design of novel recombinant paramyxovirus vectors based on engineered mutations in the viral P/V gene.  相似文献

The rate of production of Clostridium pasteurianum rubredoxin overexpressed in Escherichia coli was examined by electrospray ionization-Fourier transform ion cyclotron resonance (ESI-FTICR) mass spectrometry. Previous work had shown that this heterologous expression resulted in isolation of both iron-containing (FeRd) and zinc-containing (ZnRd) rubredoxins. In the present work, minimally processed cell lysates of E. coli were analyzed in order to monitor the production of FeRd and ZnRd. The sensitivity of the measurement favored FeRd relative to ZnRd, and this differential sensitivity was quantitated using previously separated and purified rubredoxins. A time course study indicated that ZnRd and FeRd are produced simultaneously during overexpression, but at different rates. The ratio of the concentration of ZnRd to FeRd increased in a linear fashion during 3 h following induction of overexpression. Since only FeRds have been reported from native bacteria and archaea, the data suggest that either Zn2+ is sequestered from rubredoxins during native biosynthesis or that ZnRds may have escaped detection in the native microorganisms. ESI-FTICR mass spectrometry is shown to be a useful tool for monitoring metal insertion during protein biosynthesis.  相似文献

A two-layer model is used to simulate the mechanical behavior of an airway or other biological vessel under external compressive stress or smooth muscle constriction sufficient to cause longitudinal mucosal buckling. Analytic andfinite element numerical methods are used to examine the onset of buckling. Post-buckling solutions are obtained by finite element analysis, then verified with large-scale physical model experiments. The two-layer model provides insight into how the stiffness of a vessel wall changes due to changes in the geometry and intrinsic material stiffnesses of the wall components. Specifically, it predicts that the number of mucosal folds in the buckled state is diminished most by increased thickness of the inner collagen-rich layer, and relatively little by increased thickness of the outer submucosal layer. An increase in the ratio of the inner to outer material stiffnesses causes an intermediate reduction in the number of folds. Results are cast in a simple form that can easily be used to predict buckling in a variety of vessels. The model quantitatively confirms that an increase in the thickness of the inner layer leads to a reduction in the number of mucosal folds, and further, that this can lead to increased vessel collapse at high levels of smooth muscle constriction.  相似文献

The plasticity of growth of animals in time, due to the resilience of their response to the ways they can be fed, suggests the difficulty of describing growth by a stochastic model in the time domain. A model is presented which avoids this difficulty by describing growth as a Markov process in the food-consumed domain, assuming that, at conception, (1) the maximum mature weight as a number α of biomass units of mass μ, and (2) the probability B of production of a biomass unit per unit of food consumed, are specified. Constancy of α, μ and B, as the animal feeds, is the basis of the proposed Markov process. The mean growth from infancy to maturity in the food-consumed domain is then the old law of diminishing returns empirically formulated first by Spillman (1924) for cattle and swine, and confirmed by Titus, Jull &; Hendricks (1934) for fowl, and by Parks (1972) across species from mice to steers. The solution also leads to the possibility that the distribution of weights in a population of growing animals of the same species, is related to the distribution of mature weights among the individuals. An experiment by Lister &; McCance (1967) with well-fed and severely undernourished pigs, shows the stability of growth in the foodconsumed domain compared to the plasticity in the time domain.
