Transformation is an alternative to normal skeletal muscle development   总被引:6,自引:0,他引:6  
The differentiation of skeletal muscle is characterized by cessation of proliferation and fusion of single myoblasts to form non-replicating multinucleate fibers (myotubes). If termination of proliferation is an obligate requirement for further differentiation, myoblasts defective in this stage of development should fail to fuse or exhibit any further characteristics of myotubes. Furthermore, myoblasts which have lost the ability to control and cease proliferation may represent a transformed, potentially tumorigenic population. Formation of the neoplastic state may therefore be viewed as an alternate path, antithetical to the normal differentiation of skeletal muscle. To test this hypothesis, we isolated 13 clones of non-fusing cells from the myogenic L8 line of rat myoblasts. In contrast to the L8 line, all of the non-fusing clones maintain their proliferative capacity, do not form myotubes, nor elevated creatine kinase activity nor increased myosin, but do develop into tumors when injected into athymic mice. L8 cells do not produce tumors in these mice. Analysis of cell growth and serum requirements, plasminogen activator, hexose transport, adhesiveness, LETS protein and growth in soft agar, indicates that these non-fusing cells are transformed and clearly distinguished from the parent L8 cells. Whereas the L8 line maintains a near diploid complement of chromosomes, all non-fusing clones were polyploid. In addition, 12 of 13 non-fusing clones (but not the L8 cells) express an endogenous type C virus. Although all clones defective in differentiation formed tumors, no single in vitro characteristic was found to be a constant index of this tumorigenic capacity. We conclude that cessation of proliferation is an obligate requirement for skeletal myogenesis, that transformation is an alternative to normal skeletal muscle development and that the phenotype of these transformed cells may be quite varied.  相似文献

The separation of membrane fragments was investigated in extracts of phototropically grown Rhodopseudomonas sphaeroides to determine if the plasma membrane contains discrete regions. A highly purified fraction of bacteriochlorophyll alpha-deficient membrane fragments was isolated by differential centrifugation, chromatography on Sepharose 2B, reaggregation, and isopycnic sedimentation on sucrose gradients. Significant levels of b- and c-type cytochromes and succinate dehydrogenase were demonstrated in the isolated membrane fragments and their appearance in electron micrographs, their polypeptide profile in dodecyl sulfate-polyacrylamide gel electrophoresis, and overall chemical composition were essentially identical to a similar fraction isolated from aerobically grown cells. Their polypeptide profiles were distinct from those of the intracytoplasmic chromatophore and outer membranes, and on the basis of bacteriochlorophyll content the phototrophic fraction was contaminated with chromatophores by less than 9%. The membrane fragments contained no diaminopimelic acid or glucosamine. It is condluded that the membrane fragments isolated from phototrophically growing Rp. sphaeroides have arisen from photosynthetic pigment-depleted regions of the plasma membrane structurally and functionally differentiated from the intracytoplasmic chromatophore membrane. These regions represent conserved chemotrophic cytoplasmic membrane whose synthesis continues under photoheterotrophic conditions.  相似文献

The establishment of cultured cell lines from skin biopsies stored at -196 degrees C for periods up to 1 year has been investigated. Attempts to initiate cell cultures from the frozen tissue samples were uniformly successful. There was no alteration in chromosome constitution, morphological appearance, or specific activities of lysosomal enzymes in cells cultured from the stored samples. This process can safeguard against failure of the initial tissue culture and provide an alternate means of storing viable cells when it is impossible or impractical to initiate a cell culture immediately.  相似文献

ABSTRACT: BACKGROUND: The Comprehensive Geriatric Assessment (CGA) is an analytical tool increasingly implemented in clinical practice. Breast cancer is primarily a disease of older people; however, most evidence-based research is aimed at younger patients. METHODS: A systematic review of literature was carried out to assess the use of CGA in older breast cancer patients for clinical decision making. The PubMed, Embase and Cochrane databases were searched. RESULTS: A total of nine useful full text article results were found. Only five of these were exclusively concerned with early breast cancer; thus, studies involving a variety of cancer types, stages and treatments were accepted, as long as they included early breast cancer. The results comprised a series of low sources of evidence. However, all results shared a common theme: the CGA has a use in determining patient suitability for different types of cancer treatment and subsequently maximizing the patient's quality of life. CONCLUSIONS: There is not yet sufficient high level evidence to instate CGA guidelines as a mandatory practice in the management of breast cancer, due to the heterogeneity of available studies. More studies need to be conducted to cement current work on the benefits of the CGA. An area of particular interest is with regard to treatment options, especially surgery and chemotherapy, and identifying patients who may be suitable for these treatments.  相似文献
