Type 1 copper (T1Cu) proteins are electron transfer (ET) proteins involved in many important biological processes. While the effects of changing primary and secondary coordination spheres in the T1Cu ET function have been extensively studied, few report has explored the effect of the overall protein structural perturbation on active site configuration or reduction potential of the protein, even though the protein scaffold has been proposed to play a critical role in enforcing the entatic or “rack‐induced” state for ET functions. We herein report circular permutation of azurin by linking the N‐ and C‐termini and creating new termini in the loops between 1st and 2nd β strands or between 3rd and 4th β strands. Characterization by electronic absorption, electron paramagnetic spectroscopies, as well as crystallography and cyclic voltammetry revealed that, while the overall structure and the primary coordination sphere of the circular permutated azurins remain the same as those of native azurin, their reduction potentials increased by 18 and 124 mV over that of WTAz. Such increases in reduction potentials can be attributed to subtle differences in the hydrogen‐bonding network in secondary coordination sphere around the T1Cu center.  相似文献

Protein tyrosine phosphatase 1B (PTP1B) has been shown to regulate multiple cellular events such as differentiation, cell growth, and proliferation; however, the role of PTP1B in differentiation of embryonic stem (ES) cells into cardiomyocytes remains unexplored. In the present study, we investigated the effects of PTP1B inhibition on differentiation of ES cells into cardiomyocytes. PTP1B mRNA and protein levels were increased during the differentiation of ES cells into cardiomyocytes. Accordingly, a stable ES cell line expressing PTP1B shRNA was established. In vitro, the number and size of spontaneously beating embryoid bodies were significantly decreased in PTP1B-knockdown cells, compared with the control cells. Decreased expression of cardiac-specific markers Nkx2-5, MHC-α, cTnT, and CX43, as assessed by real-time PCR analysis, was further confirmed by immunocytochemistry of the markers. The results also showed that PTP1B inhibition induced apoptosis in both differentiated and undifferentiated ES cells, as presented by increasing the level of cleaved caspase-3, cytochrome C, and cleaved PARP. Further analyses revealed that PTP1B inhibition did not change proliferation and pluripotency of undifferentiated ES cells. Taken together, the data presented here suggest that PTP1B is essential for proper differentiation of ES cells into cardiomyocytes.  相似文献

The problem of deterioration of art works is particularly relevant in countries like Iran that are rich in cultural heritage. According to UNESCO data, Iran holds the tenth rank in a list of countries possessing the highest number of monuments belonging to the world cultural heritage. Archaeological areas, buildings, mosques, statues, museums and objects are all exposed to different biogenic and abiogenic stresses under generally aggressive climatic conditions. Lichens and fungi are known to actively decompose stone surfaces. This process is both physical and chemical in nature and often reaches deep below the stone surfaces. This is caused by chemical and physical interactions of the microbiota with the fluctuating and often drastically changing environmental conditions. Here, we describe and analyze the mainly physical degradation by invading fungal hyphae between stone crystals and a generally destabilizing stone texture. In addition to physical deterioration, organic acids produced by lichens enhance the chemical decomposing processes. In this work, penetration of hyphal bundles as well as individual fungi was studied, and the biodeteriorating patterns were documented and compared to general physical–chemical weathering phenomena. Several strains of aggressive black yeast-like fungi and bacteria were isolated and cultivated and will be described in a taxonomical context with many other isolates from different localities using physiological, morphological and molecular data.  相似文献

Ryanodine receptors (RyRs) are located primarily on the junctional sarcoplasmic reticulum (SR), adjacent to the transverse tubules and on the cell surface near the Z-lines, but some RyRs are on junctional SR adjacent to axial tubules. Neither the size of the axial junctions nor the numbers of RyRs that they contain have been determined. RyRs may also be located on the corbular SR and on the free or network SR. Because determining and quantifying the distribution of RyRs is critical for both understanding and modeling calcium dynamics, we investigated the distribution of RyRs in healthy adult rat ventricular myocytes, using electron microscopy, electron tomography, and immunofluorescence. We found RyRs in only three regions: in couplons on the surface and on transverse tubules, both of which are near the Z-line, and in junctions on most of the axial tubules—axial junctions. The axial junctions averaged 510 nm in length, but they occasionally spanned an entire sarcomere. Numerical analysis showed that they contain as much as 19% of a cell's RyRs. Tomographic analysis confirmed the axial junction's architecture, which is indistinguishable from junctions on transverse tubules or on the surface, and revealed a complexly structured tubule whose lumen was only 26 nm at its narrowest point. RyRs on axial junctions colocalize with Cav1.2, suggesting that they play a role in excitation-contraction coupling.  相似文献

The presence of Cs and Sr in culture medium of Azolla filiculoides caused about 27.4% and 46.3% inhibition of biomass growth, respectively, in comparison to A. filiculoides control weight which had not metals. Biosorption batch experiments were conducted to determine the Cs and Sr binding ability of native biomass and chemically modified biosorbents derived from Azolla namely ferrocyanide Azolla sorbents type 1 and type 2 (FAS1 and FAS2) and hydrogen peroxide Azolla sorbent (HAS). The best Cs and Sr removal results were obtained when A. filiculoides was treated by 2 M MgCl2 and 30 ml H2O2 8 mM at pH 7 for 12 h and it was then washed by NaOH solution at pH 10.5 for 6 h. Pretreatment of Azolla have been suggested to modify the surface characteristics which could improve biosorption process. The binding of Cs and Sr on the cell wall of Azolla was studied with micro-PIXE and FT-IR.  相似文献

Thirty-eight soil samples were collected from crude oil contaminated land in south of Iran. Initial screening of a total of 100 bacterial isolates, resulted in the selection of one isolate with maximum adsorption capacity of 52.7 mg vanadate/g dry weight. It was tentatively identified as Halomonas sp. according to morphological and biochemical properties and named strain GT-83. Removal of vanadate by biosorption with Halomonas sp. GT-83 was very sensitive to solution pH. Vanadate adsorption decreased with increasing pH, with maximum adsorption capacities achieved in at pH 3.0 in the absence and in the presence of increasing concentrations of salt. Vanadate-salt biosorption studies were also performed at this pH value. Equilibrium uptakes of vanadate increased with increasing vanadate concentration up to 600 mg/l. Maximum metal removal (91.8%) took place at pH 3.0 with initial vanadate concentration of 100 mg/l, which got reduced (84.8%) in the presence of 50 g/l salt. The equilibrium sorption data were analyzed by using Freundlich isotherm. The specific uptake of vanadate increased at low cell concentration and decreased when cell concentration exceeded 0.75 g/l. The paper also demonstrates the potential value of micro-PIXE in biosorption studies.  相似文献

Lipocalin-2 (Lcn2, NGAL) is a member of the lipocalin super family with diverse function such as the induction of apoptosis, the suppression of bacterial growth, and modulation of inflammatory response. Much interest has recently been focused on the physiological/pathological role of the lipocalin-2 that is considered to be a novel protective factor against oxidative stress. However, its precise biological roles in this protection are not fully understood. In this report we intended to test the effect of lipocalin-2 on the expression of heme oxygenase (1, 2) and superoxide dismutase (1, 2) which are two strong antioxidants. NGAL was cloned to pcDNA3.1 plasmid by using genetic engineering method. The recombinant vector was transfected to CHO and HEK293T to establish stable cell expressing NGAL and the expression of HO-1, 2 and SOD1, 2 were compared with appropriate controls by RT-PCR and western blot. On the other hand, expression of NGAL was suppressed by siRNA transfection in order to study the effect of lipocalin-2 on mentioned genes/proteins. The results showed that the expression of HO-1 and SOD1, 2 enzymes were higher in cells expressing recombinant lipocalin-2 compared with the control cells. Although the expression of HO-1 was lower in NGAL silencing cells, the expression of SOD1 and SOD2 were higher. Our data suggest that NGAL is a potent inducer of HO-1 and somewhat SOD1 and SOD2 and it appears that part of antioxidant property of NGAL could be attributed to the induction of HO-1and SOD1, 2.  相似文献

Hearts from streptozotocin (STZ)-induced diabetic rats have previously been shown to have impaired intercellular electrical coupling, due to reorganization (lateralization) of connexin43 proteins. Due to the resulting reduction in conduction reserve, conduction velocity in diabetic hearts is more sensitive to conditions that reduce cellular excitability or intercellular electrical coupling. Diabetes is a known risk factor for cardiac ischemia, a condition associated with both reduced cellular excitability and reduced intercellular coupling. Activation of Ca(2+)-independent phospholipase A(2) (iPLA(2)) is known to be part of the response to acute ischemia and may contribute to the intercellular uncoupling by causing increased levels of arachidonic acid and lysophosphatidyl choline. Normally perfused diabetic hearts are known to exhibit increased iPLA(2) activity and may thus be particularly sensitive to further activation of these enzymes. In this study, we used voltage-sensitive dye mapping to assess changes in conduction velocity in response to acute global ischemia in Langendorff-perfused STZ-induced diabetic hearts. Conduction slowing in response to ischemia was significantly larger in STZ-induced diabetic hearts compared with healthy controls. Similarly, slowing of conduction velocity in response to acidosis was also more pronounced in STZ-induced diabetic hearts. Inhibition of iPLA(2) activity using bromoenol lactone (BEL; 10 μM) had no effect on the response to ischemia in healthy control hearts. However, in STZ-induced diabetic hearts, BEL significantly reduced the amount of conduction slowing observed beginning 5 min after the onset of ischemia. BEL treatment also significantly increased the time to onset of sustained arrhythmias in STZ-induced diabetic hearts but had no effect on the time to arrhythmia in healthy control hearts. Thus, our results suggest that iPLA(2) activation in response to acute ischemia in STZ-induced diabetic hearts is more pronounced than in control hearts and that this response is a significant contributor to arrhythmogenic conduction slowing.  相似文献
