Rabbit alveolar macrophages exposed to the ionophore A23187 in the presence of extracellular Ca2+ take up about 12 nmoles of Ca2+/1 × 106 cells. This uptake induces a slight, but significant, extracellular release of granule enzymes, β-glucuronidase and lysozyme, but not of the cytoplasmic marker, lactate dehydrogenase. If either EGTA is added to the medium or Mg2+ replaces Ca2+ no stimulation of secretion is observed. If the energy supply is decreased by treating the macrophages with mitochondrial inhibitors such as oligomycin, cyanide, or rotenone and antimycin, Ca2+-dependent secretion is potentiated several fold. Human membrane bound O-acyltransferase domain-containing 7 (MBOAT7), also known as lysophosphatidylinositol acyltransferase 1 (LPIAT1), is an enzyme involved in the acyl-chain remodeling of phospholipids via the Lands' cycle. The MBOAT7 rs641738 variant has been associated with the entire spectrum of fatty liver disease (FLD) and neurodevelopmental disorders, but the exact enzymatic activity and the catalytic site of the protein are still unestablished.Human wild type MBOAT7 and three MBOAT7 mutants missing in the putative catalytic residues (N321A, H356A, N321A + H356A) were produced into Pichia pastoris, and purified using Ni-affinity chromatography. The enzymatic activity of MBOAT7 wild type and mutants was assessed measuring the incorporation of radiolabeled fatty acids into lipid acceptors.MBOAT7 preferentially transferred 20:4 and 20:5 polyunsaturated fatty acids (PUFAs) to lysophosphatidylinositol (LPI). On the contrary, MBOAT7 showed weak enzymatic activity for transferring saturated and unsaturated fatty acids, regardless the lipid substrate. Missense mutations in the putative catalytic residues (N321A, H356A, N321A + H356A) result in a loss of O-acyltransferase activity.Thus, MBOAT7 catalyzes the transfer of PUFAs to lipid acceptors. MBOAT7 shows the highest affinity for LPI, and missense mutations at the MBOAT7 putative catalytic dyad inhibit the O-acyltransferase activity of the protein. Our findings support the hypothesis that the association between the MBOAT7 rs641738 variant and the increased risk of NAFLD is mediated by changes in the hepatic phosphatidylinositol acyl-chain remodeling. ABSTRACT A phytosociological study of the shrubland communities which colonise abandoned terraced olive groves in the Tyrrhenian district of Central Italy is presented here. The vegetational analysis was carried out using the Braun Blanquet phytosociological approach. Two main types of shrubland communities characterising distinct environmental conditions occur within abandoned terraced olive groves. On the one hand, there are those scrub types which tend to occupy the central part of the terrace; on the other hand, there are the scrubs types which develop along the stone walls bordering the terrace. Different phytosociological associations, sub-associations, and variants ranging from Pistacio-Rhamnetalia alaternii to Prunetalia spinosae are recognised and described. In the warm dry areas, both Myrto-Lentiscetum and Rhamno-Euphorbietum dendroidis occur. Also, a new Oleo-Ceratonion sub-association (Myrto-Lentiscetum spartietosum junceii) is presented here for the first time. In the inner zones of the study area, a range of aspects characterising a new Pruno-Rubion ulmifolii community type (Roso-Rubetum ulmifolii ass. nova) are identified. 相似文献

A long-range surface plasmon polariton variable optical attenuator based on available nematic liquid crystals and polymers is proposed and theoretically investigated. It is demonstrated that the electro-optic control of the nematic molecular orientation is capable of tuning the level of index asymmetry of an Au stripe waveguide and the key properties of the fundamental long-range plasmonic mode, such as modal profile and propagation losses. By proper structural design and material selection, plasmonic in-line intensity modulators are designed, which exhibit very low power consumption, extinction ratios in excess of 30 dB, and insertion losses as low as 1 dB for a device length in the millimeter range. The hepatitis delta virus (HDV) is a small, defective RNA virus that requires the presence of the hepatitis B virus (HBV) for its life cycle. Worldwide more than 15 million people are co-infected with HBV and HDV. Although much effort has been made, the early steps of the HBV/HDV entry process, including hepatocyte attachment and receptor interaction are still not fully understood. Numerous possible cellular HBV/HDV binding partners have been described over the last years; however, so far only heparan sulfate proteoglycans have been functionally confirmed as cell-associated HBV attachment factors. Recently, it has been suggested that ionotrophic purinergic receptors (P2XR) participate as receptors in HBV/HDV entry. Using the HBV/HDV susceptible HepaRG cell line and primary human hepatocytes (PHH), we here demonstrate that HDV entry into hepatocytes depends on the interaction with the glycosaminoglycan (GAG) side chains of cellular heparan sulfate proteoglycans. We furthermore provide evidence that P2XR are not involved in HBV/HDV entry and that effects observed with inhibitors for these receptors are a consequence of their negative charge. HDV infection was abrogated by soluble GAGs and other highly sulfated compounds. Enzymatic removal of defined carbohydrate structures from the cell surface using heparinase III or the obstruction of GAG synthesis by sodium chlorate inhibited HDV infection of HepaRG cells. Highly sulfated P2XR antagonists blocked HBV/HDV infection of HepaRG cells and PHH. In contrast, no effect on HBV/HDV infection was found when uncharged P2XR antagonists or agonists were applied. In summary, HDV infection, comparable to HBV infection, requires binding to the carbohydrate side chains of hepatocyte-associated heparan sulfate proteoglycans as attachment receptors, while P2XR are not actively involved. 相似文献
