A non-nodulating auxotroph (L4-73) derived from an effective strain (L4) of Rhizobium leguminosarum has a growth requirement for adenine and thiamine. The auxotroph was able to infect the roots of the host plant Pisum sativum L. but formed root nodules (ineffective in nitrogen fixation) only when adenine and, to a lesser extent, thiamine were added to the plant substrate. Nodules formed in the presence of adenine were structurally abnormal, containing small cells in which infection threads appeared to have aborted. In the presence of thiamine the auxotroph produced a smaller number of nodules which were slightly more developed and were able to reduce trace amounts of acetylene to ethylene. The adenine effect predominated when both growth factors were added together or separately in different sequences. Adenine amendment was required during the first 6 days following inoculation to achieve the maximum number of nodules. The block in symbiosis could not be fully overcome by sequential addition or removal from the root medium of either compound or by addition of some other adeninecontaining compounds. Partial prototroph revertants requiring adenine but not thiamine induced a nodulation response similar to that of the original auxotroph in the presence of thiamine; partial prototrophs requiring thiamine only were almost fully effective. Bioassay of pea root tissue indicated the presence of significant amounts of both adenine and thiamine or related substances in the roots. The auxotroph was able to compete with the parent strain L4 in nodulation on roots of pea only in the presence of exogenous adenine.  相似文献

Summary The effect of nutrient supply on nodule formation and competition between Rhizobium strains for nodulation ofLotus pedunculatus was studied. Limiting plant growth by decreasing the supply of nutrients in an otherwise nitrogen-free medium, increased the size but decreased the number and the nitrogenase activity of nodules formed by a fast-growing strain of Lotus Rhizobium (NZP2037). In contrast decreasing nutrient supply caused only a small decline in the size, number and nitrogenase activity of nodules formed by a slow-growing strain (CC814s). Providing small quantities of NH4NO3 (50 to 250 g N) to plants grown with a normal supply of other nutrients stimulated nodule development by both Rhizobium strains and increased the nitrogenase activity of the NZP2037 nodules. Differences in the level of effectiveness (nitrogen-fixing ability) of nodules formed by different Rhizobium strains on plants grown with a normal supply of nutrients were less apparent when the plants were grown with decreased nutrient supply or when the plants were supplied with low levels of inorganic N.Inter-strain competition for nodulation ofL. pedunculatus between the highly effective slow-growing strain CC814s and 7 other fast- and slow-growing strains, showed CC814s to form 42 to 100% of the nodules in all associations. The greater nodulating competitiveness of strain CC814s prevailed despite changes in the nutrient supply to the host plant. A tendency was observed for partially effective Lotus Rhizobium strains to become more competitive in nodule formation when plant growth was supplemented with low levels of inorganic nitrogen.  相似文献

The reproductive biology of coral trout, Plectropomus leopardus, from the Great Barrier Reef (Australia) was investigated by correlating gonadal condition with plasma levels of gonadal steroids. Female fish were found to be regressed from mid-summer to early spring, after which rapid and cyclical increases in gonado-somatic index (I G), maximum oocyte diameter (MOD) and plasma concentrations of estradiol-17β and testosterone were detected. Male fish, in contrast, commenced recrudescence slightly earlier in winter and responded with less dramatic increases in both I G and plasma concentrations of testosterone and 11-ketotestosterone. The mode of oocyte development was multiple group-synchronous, and cyclical fluctuations in reproductive parameters (I G, MOD and gonadal steroid concentrations) were synchronized with new-moon lunar phases. It is likely, therefore, that individual P. leopardus have the capacity to spawn on multiple occasions, with lunar periodicity. However, evidence suggests that early bouts of reproduction may be more important in terms of reproductive investment than subsequent bouts later in the same season. It is concluded that patterns of gametogenesis and steroidogenesis in P. leopardus are similar to the patterns displayed by other tropical groupers, suggesting that management regimes and propagation protocols developed for these fishes may also be appropriate for use with P. leopardus.  相似文献

In order to compare the effects on reproductive performance of short-term or prolonged exposure to elevated temperatures during vitellogenesis, female Atlantic salmon Salmo salar were held at a water temperature of 22° C for periods of 4 or 6 weeks during the austral summer and autumn. Plasma levels of 17β-oestradiol (E2), testosterone (T) and vitellogenin (Vtg) were monitored and reproductive success was compared to that in groups of fish maintained at 14 or 22° C for 12 weeks from mid-January. Significant endocrine effects were observed within as few as 3 days of the commencement of exposure to 22° C, when plasma levels of E2 ( c. 0·5 ng ml−1) and Vtg ( c. 1·4 mg ml−1) were approximately half those observed in fish maintained at 14° C ( c. 1·0 ng ml−1 and 2·7 mg ml−1 respectively). The fertility and survival to the eyed stage of ova from fish held at 14° C exceeded 85 and 70% respectively, whereas ova from fish held at 22° C for 6 or 12 weeks exhibited significantly reduced fertility (<70 and <45% respectively) and survival ( c. 40 and 13% respectively). In spite of significant endocrine effects at all stages, a 4 week exposure to 22° C only generated significant reductions in egg fertility (<65%) and survival ( c. 30%) when it occurred between mid-February and mid-March. Together, these data confirm that high temperature spikes can affect reproductive success as strongly as more prolonged exposures, and indicate that there is a critical period of reproductive sensitivity to elevated temperature in late February and early March in this stock of Atlantic salmon.  相似文献

Greenback flounder were treated with either an injection of des Gly10 [D‐Ala6] LHRH ethylamide (LHRH‐a), a silastic pellet implant of testosterone (T) or LHRH‐a+T. LHRH‐a and LHRH‐a+T induced more ovulatory events than in control fish or fish treated with T alone. T treatment did not significantly increase the number of ovulatory events above control levels. Treatment with LHRH‐a+T stimulated more ovulatory events than treatment with LHRH‐a alone, suggesting T increases pituitary responsiveness to LHRH‐a. Plasma levels of 17β‐estradiol (E 2) were significantly higher in fish treated with LHRH‐a and LHRH‐a+T than in control fish or fish treated with T. Plasma levels of 17,20β‐dihydroxy‐4‐pregnen‐3‐one were not elevated above control levels. Daily ovulations, elevated E 2 and T levels and presumably elevated GtH‐II levels, persisted well after the expected clearance time of exogenous LHRH‐a. Positive steroid feedback may have contributed to the processes of repeated final oocyte maturation and ovulation.  相似文献

Dissection of freshly eaten prey suggests that the North Pacific daggertooth Anotopterus nikparini sever the spine of small prey with a single slashing blow. The bite marks indicate that the immobilized prey is manipulated and swallowed head first. Frequent evidence suggests daggertooth wound prey which are too large to consume whole by cutting off strips of flesh. In either case the wounds consist of a single prominent slash mark on one side and a series of inconspicuous marks on the opposite side. Retinal morphology provides supporting evidence that daggertooths are visual ambush predators that attack prey from below, and should be considered as epipelagic rather than mesopelagic fish. Visual acuity appears to be high, suggesting that frequent attacks on prey too large to be killed immediately is not error, but a well-defined feeding behaviour. The daggertooth may be a significant source of mortality for economically important fish such as all sizes of salmon.  相似文献

Rhizobium loti NZP2037 and NZP2213, each cured of its single large indigenous plasmid, formed effective nodules on Lotus spp., suggesting that the symbiotic genes are carried on the chromosome of these strains. By using pSUP1011 as a vector for introducing transposon Tn5 into R. loti NZP2037, symbiotic mutants blocked in hair curling (Hac), nodule initiation (Noi), bacterial release (Bar), and nitrogen fixation (Nif/Cof) on Lotus pedunculatus were isolated. Cosmids complementing the Hac, Noi, and Bar mutants were isolated from a pLAFR1 gene library of NZP2037 DNA by in planta complementation and found to contain EcoRI fragments of identical sizes to those into which Tn5 had inserted in the mutants. The cosmids that complemented the mutants of these phenotypic classes did not share common fragments, nor did cosmids that complemented four mutants within the Noi class, suggesting that these symbiotically important regions are not tightly linked on the R. loti chromosome.  相似文献

Rhizobium loti strains NZP2037 and NZP2213 were each found to contain a single large plasmid: pRlo2037a (240 MDal) and pRlo2213a (120 MDal), respectively. Plasmid DNA present in crude cell lysates of each strain and purified pRlo2037a DNA did not hybridize with pID1, a recombinant plasmid containing part of the nitrogen fixation (nif) region of R. meliloti, indicating that nif genes were not present on these plasmids. The transposon Tn5 was inserted into pRlo2037a and this plasmid was then transferred into R. leguminosarum, R. meliloti and Agrobacterium tumefaciens. All transconjugants failed to nodulate Lotus pedunculatus, suggesting that the ability to nodulate this legume was also not carried on pRlo2037a. Transfer of pRlo2037a to R. loti strain NZP2213 did not alter the Nod+ Fix- phenotype of this strain for L. pedunculatus. Determinants for flavolan resistance, believed to be necessary for effective nodulation of L. pedunculatus, were not carried on pRlo2037a. These data suggest that nodulation, nitrogen fixation and flavolan resistance genes are not present on the large plasmid in R. loti strain NZP2037.  相似文献

Synopsis The social and reproductive biology of the sand tilefish,Malacanthus plumieri (Malacanthidae), was studied at Glover's Reef, Belize, where this species occurs in colonies over sand-rubble flats. Individuals each occupy a home burrow refuge and a surrounding home range. Home range overlap among adjacent fish of the same sex is low, and individuals defend exclusive use of much of their home range against all conspecifics except mates (i.e., territoriality). Areas defended by males overlap the territories of up to 6 females; and male territory area is positively related to the number of female residents. Males maintain dominance over females within their territories by aggression, including intervention into some female disputes. Females spawn pelagically-dispersed eggs as frequently as every day. Each female spawns near her burrow, almost exclusively with the male whose defended area encompasses her territory (harem polygyny). Tilefish colonies therefore consist of a mosaic of female territories over which adjacent male territories are superimposed. Histological evidence and observation of behavioral sex change in one female revealed thatM. plumieri is capable of protogynous sex reversal. Females did not change sex in response to removal of one male. Occurrence of small transitional fish indicates that the onset of sex change is controlled by factors other than size-related social hierarchies within harems or colonies.  相似文献

A riboflavin-requiring auxotroph of Rhizobium trifolii (T1/D-his(r)-15) formed ineffective root nodules on red clover and on two cultivars of subterranean clover, but produced almost fully effective nodules on several other cultivars of subterranean clover. Fluorescence and bioassay measurements of the flavin content of the roots and shoots of these cultivars revealed no differences between cultivars which could be correlated with the differences in symbiotic response. The concentration of flavin in nodules formed by the auxotroph (in the absence of riboflavin), by the effective parent strain (T1), or by a partly effective mutant (penicillin-resistant) of T1 was roughly proportional to the effectiveness of the nodules. Effective nodules contained 20 times as much flavin, and ineffective nodules 3 to 4 times as much flavin as non-nodulated root tissue. Approximately 20 to 30% of the flavins in both root and nodule tissue was flavin adenine dinucleotide and 70 to 80% was riboflavin + flavin mononucleotide. Most of the flavin adenine dinucleotide in macerated nodules was associated with host cell fragments, and none was detected in a cell-free fraction. Bacteroids accounted for approximately 20% of flavins in effective nodules and also contained more riboflavin + flavin mononucleotide than cultured rhizobial cells. The total flavin content of noninoculated roots increased from about 1.2 nmoles to 1.7 nmoles flavin/g of tissue after 3 days' exposure to 80 mum riboflavin. Exposure of only the upper or lower portion of preinoculated roots indicated negligible translocation, as effective nodulation occurred only on parts of the root in direct contact with riboflavin. Plants grown in a medium containing combined nitrogen (100 or 300 mum nitrogen added as (NH(4))(2)SO(4)), but no added riboflavin showed an increased root flavin content (about 2.1 nmoles flavin/g tissue) and a partly effective response when inoculated with the mutant. Nitrogen also promoted some upward translocation of exogenous riboflavin in the roots.  相似文献
