Four heavy metal salts, nickel sulphate, mercuric chloride, cadmium sulphate and zinc sulphate, were tested for induction of sister chromatid exchange (SCE) in root meristem cells ofAllium cepa. A simple modified Feulgen staining procedure was employed for SCE-analysis. Maleic hydrazide and paraquat were included for comparison. An evaluation of genotoxicity of the above test chemicals made on the basis of SCE-assay was found positive for all the test chemicals with exception of zinc sulphate which gave a weak positive result.  相似文献

The effects of sanguinarine on IgE mediated early signaling mechanisms leading to inflammatory mediators release were investigated. Pretreatment of RBL 2H3 cells with sanguinarine inhibited IgE induced activation of type II PtdIns 4-kinase activity. Concomitant with type II PtdIns 4-kinase inhibition, sanguinarine also inhibited IgE induced degranulation and β hexosaminidase release in RBL 2H3 cells. In vitro assays showed sanguinarine inhibited type II PtdIns 4-kinase activity in a dose dependent fashion with no effect on PtdIns 3-kinase activity. Fluorescence spectroscopic studies suggested that sanguinarine binds to type II PtdIns 4-kinases α and β isoforms with a Kd of 2.4 and 1.8 μM, respectively. Kinetic studies showed that sanguinarine competes with PtdIns binding site of type II PtdIns 4-kinase β. These results suggest that the anti-inflammatory effects of sanguinarine on PtdIns 3-kinase signaling pathway are more likely an indirect effect and emphasize the importance of the cross talk between type II PtdIns 4-kinases and PtdIns 3-kinases.  相似文献

Abstract

Hispolon is a polyphenolic compound derived from black hoof mushroom (Phellinus linteus) or shaggy bracket mushroom (Inonotus hispidus) which induces the inhibition of cancer-promoting nuclear factor-kappa beta (NF-κβ) complex. To develop more potent lead molecules with enhanced anticancer efficiency, the mechanism of hispolon-mediated nuclear factor-κβ inhibition has been investigated by molecular modelling and docking. Ten derivatives of hispolon (DRG1-10) have been developed by pharmacophore-based design with a view to enhance the anticancer efficacy. Hispolon and its derivatives were further screened for different pharmacological parameters like binding free energy, drug likeliness, absorption–digestion–metabolism–excretion (ADME), permeability, mutagenicity, toxicity and inhibitory concentration 50 (IC50) to find a potent lead molecule. Based on pharmacological validation, comparative molecular dynamics (MD) simulations have been performed for three lead molecules: Hispolon, DRG2 and DRG7 complexed with human NF-κβ up to 50?ns. By analysing different factors like root mean square deviation (RMSD), root mean square fluctuation (RMSF), radius of gyration (Rg), solvent-accessible surface area (SASA) and principal component analysis (PCA), Gibb’s free energy plots DRG2 have more binding efficiency compared to hispolon and DRG7. In RMSD plot, hispolon-bound NF-κβ has the most deviation within a range between 0.125 and 0.45?nm, and DRG2-bound complex showed the range between 0.125 and 0.25?nm. The residues of NF-κβ responsible for hydrophobic interactions with ligand, e.g. Met469, Leu522 and Cys533, have the lowest fluctuation values in DRG2-bound complex. The average Rg fluctuation for DRG2-bound NF-κβ has been recorded under 2.025?nm for most of the simulation time which is much less compared to hispolon and DRG7. Gibb’s free energy plots also define the highest stability of DRG2-bound NF-κβ.

Body temperature (Tb), food intake and sleep cycle follows a circadian rhythm. Circadian variations of Tb may be influenced by changes in sleep time, late night consumption of high calorie diet and less physical activities. Deviations in any of these daily habits may alter the acrophase, mesor and amplitude of the cosinor curve of Tb. Changes in these confounding factors may also contribute to the body mass index (BMI) of the individual. This study was conducted on 29 high school going adolescents. We have employed an ideal cosinor model to study the changes in the 24 h cyclical changes in Tb (n = 12). Academic overload during examination days was seen in these high school going adults. Sleep duration and dietary habits were also studied in them to find out the relationship with BMI and circadian variations if any (n = 17). Higher BMI was observed in individuals having shorter sleep duration. BMI was high in students consuming high calorie diet at the time of the day when the body metabolism is normally low. A mathematical model exhibiting altered circadian rhythms is proposed that may be used to modify strategies to restore biorhythms for better health.
