Traditional farm-based products based on livestock are one of the main contributors to greenhouse gas emissions. Cultivated meat is an alternative that mimics animal meat, being produced in a bioreactor under controlled conditions rather than through the slaughtering of animals. The first step in the production of cultivated meat is the generation of sufficient reserves of starting cells. In this study, bovine adipose-derived stem cells (bASCs) were used as starting cells due to their ability to differentiate towards both fat and muscle, two cell types found in meat. A bioprocess for the expansion of these cells on microcarriers in spinner flasks was developed. Different cell seeding densities (1,500, 3,000, and 6,000 cells/cm2) and feeding strategies (80%, 65%, 50%, and combined 80%/50% medium exchanges) were investigated. Cell characterization was assessed pre- and postbioprocessing to ensure that bioprocessing did not negatively affect bASC quality. The best growth was obtained with the lowest cell seeding density (1,500 cells/cm2) with an 80% medium exchange performed (p < .0001) which yielded a 28-fold expansion. The ability to differentiate towards adipogenic, osteogenic, and chondrogenic lineages was retained postbioprocessing and no significant difference (p > .5) was found in clonogenicity pre- or postbioprocessing in any of the feeding regimes tested.  相似文献

Competitive interactions between coinfecting genotypes of the same pathogen can impose selection on virulence, but the direction of this selection depends on the mechanisms behind the interactions. Here, we investigate how host immune responses contribute to competition between clones in mixed infections of the rodent malaria parasite Plasmodium chabaudi. We studied single and mixed infections of a virulent and an avirulent clone and compared the extent of competition in immunodeficient and immunocompetent mice (nude mice and T cell-reconstituted nude mice, respectively). In immunocompetent mice, the avirulent clone suffered more from competition than did the virulent clone. The competitive suppression of the avirulent clone was alleviated in immunodeficient mice. Moreover, the relative density of the avirulent clone in mixed infections was higher in immunodeficient than in immunocompetent mice. We conclude that immune-mediated interactions contributed to competitive suppression of the avirulent clone, although other mechanisms, presumably competition for resources such as red blood cells, must also be important. Because only the avirulent clone suffered from immune-mediated competition, this mechanism should contribute to selection for increased virulence in mixed infections in this host-parasite system. As far as we are aware, this is the first direct experimental evidence of immune-mediated apparent competition in any host-parasite system.  相似文献

OBJECTIVE: To investigate (1) the feasibility of preparing cell blocks by inverted filter sedimentation (IFS-CB) from endometrial samplings processed by the ThinPrep (TP) technique (Cytyc Corp., Boxborough, Massachusetts, U.S.A.), and (2) the possibility of increasing the diagnostic accuracy of TP endometrial cytology by examining the tissue architecture as an adjunctive method of detecting endometrial lesions. STUDY DESIGN: Three hundred one endometrial samplings were obtained, using the Endogyn endometrial device (Biogyn S. n.c., Italy), from perimenopausal and postmenopausal women. The endometrial samplings were collected in a vial with liquid fixative for the TP processing. One TP slide was prepared from each case. If adequate material remained in the vial after the TP slide preparation, it was processed for IFS-CB preparation. RESULTS: IFS-CB preparation was processed in 263 cases (87%) with adequate material. Diagnoses on IFS-CB preparations obtained by endometrial sampling matched those of the hysterectomy specimens. The addition of IFS-CB histology to the cytologic diagnosis by TP increased the diagnostic accuracy of endometrial cytology to 96.3% and 100% for benign/atrophic endometrium and adenocarcinoma, respectively (p = 0.39 and 0.46). In hyperplasia without atypia and hyperplasia with atypia, the diagnostic accuracy increased significantly, to 96% and 95.3%, respectively (p = 0.037 and < 0.001). CONCLUSION: This study illustrates the merit of linking TP cytology with direct endometrial sampling, including small tissue fragments and material adequate for IFS-CB preparation. TP cytology provides an accurate cytologic diagnosis and the possibility of IFS-CB preparation, which could be a valuable diagnostic adjunct to TP cytology.  相似文献

The role of hormones in the determination of sexual characteristics has been known for several decades. It has been shown, for example, that several products, including sex steroids, may influence the body development pattern, metabolic pathways, fat and muscle distribution and vocal cord anatomy, thus producing an overall outcome consistent with a masculine or feminine phenotypic pattern. These qualities are usually described as secondary sexual traits, so as to be distinguished from primary sex traits, usually referring to the gonads and external genitalia. However, it must be noted that hormonal regulation may not explain the full range of the sexual phenotype, since the central nervous system retains a significant role in the establishment of sexual identity, thus giving rise to a higher sex determination stage exclusively described in humans, namely behavioral or psychological sex. Recently, it has been suggested that differences among the sexes are not limited to brain function but they may also refer to anatomical differences and different biochemical profiles, including a distinct pattern of AR and ER distribution. This new aspect of sexual dimorphism suggests a whole system of meta-hormonal regulation, recently described as the sexual brain model. The role of local androgen and/or estrogen concentrations in the initial establishment of brain sexual dimorphism is still under evaluation, since the first results are relatively inconclusive and no direct cause and effect relationship has been proven so far. On the other hand, sex hormones have recently been found to participate in processes well beyond their initially suggested spectrum of action. For instance, ER interacts with EGFR in a number of ways, affecting development of a number of epithelial structures. Estrogen receptors have also been detected in a number of non-classic targets of steroids, such as the brain and the lungs. This observation may imply that sexual dimorphism goes a lot deeper than previously estimated, affecting virtually every organic system, suggesting, in essence, the existence of two different functional models for the whole human body, formulated and conserved throughout the evolutionary progress.  相似文献

Dimorphism between testis and ovary in germ cells proliferative behavior, shows remarkable differences in foetal and neonatal period [14.5 days post conception (dpc)--7 days post partum (dpp)]. Immunostaining of the foetal testis, with the PCNA and Ki-67 antibodies [estimation of Labeling Index (LI)], reveals increasing germ cells population until birth. Afterwards, a sharp decline in the first 3 days of postnatal life and a transient increase, between 3 and 5 dpp, is observed. Then, the mitotic activity of germ cells ceases. In the foetal ovary, germ cells proliferation reaches a peak value before birth, decreasing thereafter Somatic (Sertoli or follicular) cells behave similarly in both sexes. Increased mitotic activity is observed throughout the examined period. Thus, the gonadal dimorphism in proliferative behavior, concerns only germ cell lineage and is established during the foetal and neonatal period.  相似文献

Cytosolic glutathione transferases (GSTs) are a major reserve of high-capacity ligand binding proteins which recognise a large variety of hydrophobic compounds. In the present study, the binding of non-substrate xenobiotic compounds (herbicides and insecticides) to maize GST I was investigated by employing kinetic inhibition studies, site-directed mutagenesis and molecular modelling studies. The results showed that the xenobiotics bind at the substrate binding site. Based on in silico docking analysis, two residues were selected for assessing their contribution to xenobiotic binding. The mutant Gln53Ala of GST I Exhibits 9.2-fold higher inhibition potency for the insecticide malathion, compared to the wild-type enzyme. A potentiometric assay was developed for the determination of malathion using the Gln53Ala mutant enzyme. The assay explores the ability of the xenobiotic to promote inhibition of the GST-catalysing 1-chloro-2,4-dinitrobenzene (CDNB)/glutathione (GSH) conjugation reaction. The sensing scheme is based on the pH change occurring in a low buffer system by the GST reaction, which is measured potentiometrically using a pH electrode. Calibration curve was obtained for malathion, with useful concentration range 0-20muM. The method's reproducibility was in the order of +/-3-5% and malathion recoveries were 96.7+/-2.8%. Immobilized Gln53Ala mutant GST was used to assemble a biosensor for malathion. The enzyme was immobilized by crosslinking with glutaraldehyde and trapped behind a semipermeable membrane in front of the pH electrode. The results demonstrated that the immobilized enzyme behaved similar to free enzyme.  相似文献

OBJECTIVE: To develop and validate a computer-based approach for the quantitative assessment of estrogen receptor (ER) status in breast tissue specimens for breast cancer management. STUDY DESIGN: Microscopy images of 32 immunohistochemically (IHC) stained specimens of breast cancer biopsies were digitized and were primarily assessed for ER status (percentage of positively stained nuclei) by a histopathologist. A pattern recognition system was designed for automatically assessing the ER status of the IHC-stained specimens. Nuclei were automatically segmented from background by a pixel-based unsupervised clustering algorithm and were characterized as positively stained or unstained by a supervised classification algorithm. This cascade structure boosted the system's classification accuracy. RESULTS: System performance in correctly characterizing the nuclei was 95.48%. When specifying each case's ER status, system performance was statistically not significantly different to the physician's assessment (p = 0.13); when ranking each case to a particular 5-scale ER-scoring system (giving the chance of response to endocrine treatment), the system's score and the physician's score were in agreement in 29 of 32 cases. CONCLUSION: The need for reliable and operator independent ER-status estimation procedures may be served by the design of efficient pattern recognition systems to be employed as support opinion tools in clinical practice.  相似文献

The purpose of the present study was to investigate the effect of short-term resistance training and detraining on shot put throwing performance. Eleven young healthy subjects with basic shot put skills participated in 14 weeks of resistance training, which was followed by 4 weeks of detraining. Shot put performance in four field tests was measured before (T1) and after (T2) resistance training and after detraining (T3). At the same time points, one repetition maximum (1RM) was measured in squat, bench press, and leg press. Fat-free mass (FFM) was determined with dual x-ray absorptiometry and muscle biopsies obtained from vastus lateralis for the determination of fiber type composition and cross-sectional area (CSA). 1RM strength increased 22-34% (p < 0.01) at T2 and decreased 4-5% (not significantly different) at T3. Shot put performance increased 6-12% (p < 0.05) after training and remained unaltered after detraining. FFM increased at T2 (p < 0.05) but remained unchanged between T2 and T3. Muscle fiber CSA increased 12-18% (p < 0.05) at T2. Type I muscle fiber CSA was not altered after detraining, but type IIa and IIx fiber CSA was reduced 10-12% (p < 0.05). The percentage of type IIx muscle fibers was reduced after training (T1 = 18.7 +/- 4, T2 = 10.4 +/- 1; p < 0.05), and it was increased at T3 compared with T2 (T3 = 13.7 +/- 1; p < 0.05). These results suggest that shot put performance remains unaltered after 4 weeks of complete detraining in moderately resistance-trained subjects. This might be linked to the concomitant reduction of muscle fiber CSA and increase in the percentage of type IIx muscle fibers.  相似文献
