Ketogulonicigenium vulgare, though grows poorly when mono-cultured, has been widely used in the industrial production of the precursor of vitamin C with the coculture of Bacillus megaterium. Various efforts have been made to clarify the synergic pattern of this artificial microbial community and to improve the growth and production ability of K. vulgare, but there is still no sound explanation. In previous research, we found that the addition of reduced glutathione into K. vulgare monoculture could significantly improve its growth and productivity. By performing SEM and TEM, we observed that after adding GSH into K. vulgare monoculture, cells became about 4-6 folds elongated, and formed intracytoplasmic membranes (ICM). To explore the molecular mechanism and provide insights into the investigation of the synergic pattern of the co-culture system, we conducted a comparative iTRAQ-2-D-LC-MS/MS-based proteomic analysis of K. vulgare grown under reduced glutathione. Principal component analysis of proteomic data showed that after the addition of glutathione, proteins for thiamin/thiamin pyrophosphate (TPP) transport, glutathione transport and the maintenance of membrane integrity, together with several membrane-bound dehydrogenases had significant up-regulation. Besides, several proteins participating in the pentose phosphate pathway and tricarboxylic acid cycle were also up-regulated. Additionally, proteins combating intracellular reactive oxygen species were also up-regulated, which similarly occurred in K. vulgare when the co-cultured B. megaterium cells lysed from our former research results. This study reveals the demand for transmembrane transport of substrates, especially thiamin, and the demand for antioxidant protection of K. vulgare.  相似文献

Multidrug resistant microbes present in the environment are a potential public health risk. In this study, we investigate the presence of New Delhi metallo-β-lactamase 1 (NDM-1) producing bacteria in the 99 water samples in Beijing City, including river water, treated drinking water, raw water samples from the pools and sewage from 4 comprehensive hospitals. For the bla NDM-1 positive isolate, antimicrobial susceptibility testing was further analyzed, and Pulsed Field Gel Electrophoresis (PFGE) was performed to determine the genetic relationship among the NDM-1 producing isolates from sewage and human, as well as the clinical strains without NDM-1. The results indicate that there was a higher isolation of NDM-1 producing Acinetobacter baumannii from the sewage of the hospitals, while no NDM-1 producing isolates were recovered from samples obtained from the river, drinking, or fishpond water. Surprisingly, these isolates were markedly different from the clinical isolates in drug resistance and pulsed field gel electrophoresis profiles, suggesting different evolutionary relationships. Our results showed that the hospital sewage may be one of the diffusion reservoirs of NDM-1 producing bacteria.  相似文献

Two new species of Gesneriaceae, Wentsaiboea tiandengensis Yan Liu & B. Pan and Wentsaiboea luochengensis Yan Liu & W. B. Xu from Karst caves in Guangxi, China are described and illustrated. Wentsaiboea tiandengensis resembles W. renifolia, but differs in the leaf blade being cordate or broadly ovate, the margin crenate or repand, with conspicuous pinnate veins 3–5 on each side, corolla throat constricted, corolla lobes ovate with apex acute, staminodes 3, and ovary broadly ovoid. Wentsaiboea luochengensis is similar to W. renifolia, but can be distinguished from the latter by the coriaceous leaf blade with 2–3 inconspicuous pinnate veins on each side, elliptic or broadly ovate to sub‐rotund in shape, with sub‐entire margin and attenuate to cuneate or sub‐rotund base, broadly ovate or sub‐rotund corolla lobes, rotund or obtuse apex, and 3 staminodes.  相似文献

In order to study the physiological role of acetate metabolism in Escherichia coli, the growth characteristics of an E. coli W3100 pta mutant defective in phosphotransacetylase, the first enzyme of the acetate pathway, were investigated. The pta mutant grown on glucose minimal medium excreted unusual by-products such as pyruvate, D-lactate, and L-glutamate instead of acetate. In an analysis of the sequential consumption of amino acids by the pta mutant growing in tryptone broth (TB), a brief lag between the consumption of amino acids normally consumed was observed, but no such lag occurred for the wild-type strain. The pta mutant was found to grow slowly on glucose, TB, or pyruvate, but it grew normally on glycerol or succinate. The defective growth and starvation survival of the pta mutant were restored by the introduction of poly-beta-hydroxybutyrate (PHB) synthesis genes (phbCAB) from Alcaligenes eutrophus, indicating that the growth defect of the pta mutant was due to a perturbation of acetyl coenzyme A (CoA) flux. By the stoichiometric analysis of the metabolic fluxes of the central metabolism, it was found that the amount of pyruvate generated from glucose transport by the phosphoenolpyruvate-dependent phosphotransferase system (PTS) exceeded the required amount of precursor metabolites downstream of pyruvate for biomass synthesis. These results suggest that E. coli excretes acetate due to the pyruvate flux from PTS and that any method which alleviates the oversupply of acetyl CoA would restore normal growth to the pta mutant.  相似文献

哈维氏弧菌是引起大黄鱼疾病的主要致病菌之一,胞外蛋白酶是哈维氏弧菌GYC1108-1的主要致病因子。利用表型克隆技术,用Sau3AⅠ将提取的哈维氏弧菌基因组DNA酶切成短片段,纯化回收1-5 kb的片段与BamH I酶切的pUC118在T4连接酶的作用下进行连接,并转化感受态细胞TG1,在含0.5%脱脂奶和100μg/mL氨苄青霉素的LB平板上筛选胞外蛋白酶阳性株,命名为YZ。测定阳性株YZ的胞外蛋白酶活性,并对阳性株的重组质粒的插入片段进行序列测定,分析开放阅读框,确定胞外蛋白酶基因编码区。发现蛋白酶基因的ORF编码531个氨基酸,在起始密码子ATG上游存在一个核糖体结合位点(SD),其上游的启动区与大肠杆菌的σ70启动区和枯草芽孢杆菌的启动区高度同源。在终止子(TAG)下游,存在两个反向重复序列,为推导的2个转录终止子。哈维氏弧菌胞外蛋白酶基因的克隆与诠释为哈维氏弧菌致病机理和疫苗的开发奠定了基础。  相似文献

It has been demonstrated that antimony (Sb) at concentrations ranging from 1.0 to 10.0 mg L−1 inhibits O2 evolution. Deeper insight into the influence of Sb on PSII was obtained with measurements of in vivo chlorophyll fluorescence. The donor and the acceptor sides of PSII were shown to be the target of Sb. Sb treatment induces inhibition of electron transport from QA to QB/QB and accumulation of P680+. S2(QAQB) charge recombination and oxidation by PQ9 molecules became more important in QA reoxidation as the electron transfer in PSII was inhibited. Sb exposure caused a steady increase in the proportion of PSIIX and PSIIβ. These changes resulted in increased fluxes of dissipated energy and decreased index of photosynthesis performance, of maximum quantum yield, and of the overall photosynthetic driving force of PSII.  相似文献

Vitamin D receptor (VDR) is a nuclear protein which mediates the physiological actions of its hormone ligand, 1,25-dihydroxyvitamin D3 (1,25(OH)2D3). While it appears that the receptor-hormone complex regulates the expression of hormone-dependent genes involved in mineral homeostasis, its role in induction of differentiation of leukaemic cells is less clear. We have studied the expression of the VDR gene in several sublines of HL-60 leukaemic cells with varying responsiveness to 1,25(OH)2D3. Sublines which rapidly differentiated to monocytic forms were shown to contain elevated steady-state levels of VDR mRNA within 1 h of exposure to high concentration of 1,25(OH)2D3. This up-regulation of the expression of VDR was not apparent in sublines in which monocytic differentiation occurred after a delay of several days. Beginning at approximately 3 h after exposure to 1,25(OH)2D3 in most cases, there was a gradual decline in VDR mRNA levels. Measurement of steady-state levels of mRNA for c-myc and c-fos showed that in sublines of HL-60 cells which respond rapidly to 1,25(OH)2D3, elevation of VDR mRNA is evident prior to the changes in proto-oncogene expression. These data are consistent with the hypothesis that a change in VDR gene expression is one of the steps that promote monocytic differentiation.  相似文献

The contribution of zinc-mediated neuronal death in the process of both acute and chronic neurodegeneration has been increasingly appreciated. Phosphatase and tensin homologue, deleted on chromosome 10 (PTEN), the major tumor suppressor and key regulator of the phosphatidylinositol 3-kinase (PI3K)/Akt pathway, plays a critical role in neuronal death in response to various insults. NEDD4-1-mediated PTEN ubiquitination and subsequent degradation via the ubiquitin proteosomal system have recently been demonstrated to be the important regulatory mechanism for PTEN in several cancer types. We now demonstrate that PTEN is also the key mediator of the PI3K/Akt pathway in the neuronal response to zinc insult. We used primary cortical neurons and neuroblastoma N2a cells to show that zinc treatment results in a reduction of the PTEN protein level in parallel with increased NEDD4-1 gene/protein expression. The reduced PTEN level is associated with an activated PI3K pathway as determined by elevated phosphorylation of both Akt and GSK-3 as well as by the attenuating effect of a specific PI3K inhibitor (wortmannin). The reduction of PTEN can be attributed to increased protein degradation via the ubiquitin proteosomal system, as we show NEDD4-1 to be the major E3 ligase responsible for PTEN ubiquitination in neurons. Moreover, PTEN and NEDD4-1 appear to be able to counter-regulate each other to mediate the neuronal response to zinc. This reciprocal regulation requires the PI3K signaling pathway, suggesting a feedback loop mechanism. This study demonstrates that NEDD4-1-mediated PTEN ubiquitination is crucial in the regulation of PI3K/Akt signaling by PTEN during the neuronal response to zinc, which may represent a common mechanism in neurodegeneration.  相似文献

FAB1/PIKfyve是介导PI(3,5)P2 (磷脂酰肌醇3,5-二磷酸)生物合成的磷酸肌醇激酶。在动物和酵母(Saccharomyces cerevisiae)中, PI(3,5)P2参与调控胞内膜运输, 但在植物中的研究较少。该文通过分析拟南芥(Arabidopsis thaliana) FAB1的T-DNA插入突变体的表型解析PI(3,5)P2的生物学功能。拟南芥FAB1基因家族包含FAB1AFAB1BFAB1CFAB1D四个基因。研究发现, fab1a/b呈现雄配子体致死的表型。利用遗传杂交获得fab1b/c/d三突变体, 发现FAB1BFAB1CFAB1D功能缺失导致根毛相比野生型变短, 经FAB1特异性抑制剂YM201636处理后的野生型中也观察到相似的短根毛表型。此外, fab1b/c/d三突变体中DR5转录水平降低。同时, 外源施加生长素类似物2,4-D和NAA能部分恢复fab1b/c/d植株短根毛的表型, 但fab1b/c/d突变体对生长素转运抑制剂(1-NOA和TIBA)的敏感性与野生型相似。此外, FAB1B/C/D功能缺失使根毛中ROS的含量减少且影响肌动蛋白的表达。上述结果表明, FAB1B/C/D通过调控生长素分布、ROS含量和肌动蛋白的表达影响拟南芥根毛伸长。  相似文献
