A novel series of steroidal compounds were designed and synthesized with various phosphorus-containing groups on the 17beta-side chain as progesterone receptor antagonists. The structure-activity relationships of these compounds are discussed. Selected compounds were tested in an rat progesterone-sensitive assay. An adult male Blainville's beaked whale (Mesoplodon densirostris) was found stranded on the Atlantic coast of the USA on 28 January 2004. Necropsy revealed a focal papilloma-like penile lesion, the cells from which revealed single 4-6 microm basophilic intranuclear inclusions. Total DNA extracted from lesion material was tested using a pan-herpes-virus PCR assay that targets the DNA polymerase gene and found to be positive. When the amplified DNA fragment was cloned, sequenced, and compared to GenBank-deposited herpesvirus DNA polymerase sequences, the detected virus was determined to be a distinct member of the Gammaherpesvirinae subfamily of herpesviruses. This new virus, tentatively named Ziphiid herpesvirus type 1, was associated with but not determined to be the cause of genital disease in the Blainville's beaked whale. 相似文献

TRPA1 is an excitatory ion channel targeted by pungent irritants from mustard and garlic. TRPA1 has been proposed to function in diverse sensory processes, including thermal (cold) nociception, hearing, and inflammatory pain. Using TRPA1-deficient mice, we now show that this channel is the sole target through which mustard oil and garlic activate primary afferent nociceptors to produce inflammatory pain. TRPA1 is also targeted by environmental irritants, such as acrolein, that account for toxic and inflammatory actions of tear gas, vehicle exhaust, and metabolic byproducts of chemotherapeutic agents. TRPA1-deficient mice display normal cold sensitivity and unimpaired auditory function, suggesting that this channel is not required for the initial detection of noxious cold or sound. However, TRPA1-deficient mice exhibit pronounced deficits in bradykinin-evoked nociceptor excitation and pain hypersensitivity. Thus, TRPA1 is an important component of the transduction machinery through which environmental irritants and endogenous proalgesic agents depolarize nociceptors to elicit inflammatory pain. 相似文献

We present a three-dimensional computer simulation of the dynamics of a vein valve. In particular, we couple the solid mechanics
of the vein wall and valve leaflets with the fluid dynamics of the blood flow in the valve. Our model captures the unidirectional
nature of blood flow in vein valves; blood is allowed to flow proximally back to the heart, while retrograde blood flow is
prohibited through the occlusion of the vein by the valve cusps. Furthermore, we investigate the dynamics of the valve opening
area and the blood flow rate through the valve, gaining new insights into the physics of vein valve operation. It is anticipated
that through computer simulations we can help raise our understanding of venous hemodynamics and various forms of venous dysfunction. 相似文献

A number of biological approaches are commonly used to assess the ecological integrity of stream ecosystems. Recently, it
is becoming increasingly common to use multiple organism groups in bioassessment. Advocates of the multiple organism approach
argue that the use of different organism groups should strengthen inference-based models and ultimately result in lower assessment
error, while opponents argue that organism groups often respond similarly to stress implying a high degree of redundancy.
Using fish, macroinvertebrate, macrophyte and benthic diatom data, site-specific parameters (e.g., water chemistry and substratum)
and catchment variables from European mountain (n = 77) and lowland (n = 85) streams we evaluated the discriminatory power and uncertainty associated with the use of a number of biological metrics
commonly used in stream assessment. The primary environmental gradient for both streams types was land use and nutrient enrichment.
Secondary and tertiary gradients were related to habitat quality or alterations in hydromorphology. Benthic diatom and macroinvertebrate
metrics showed high discriminatory power (R2 values often >0.50) and low error (<30%) with the primary (nutrient) gradient, while both fish and macrophyte metrics performed
relatively poorly. Conversely, both fish and macrophyte metrics showed higher response (high coefficients of determination)
than either benthic diatom or macroinvertebrate metrics to the second (e.g., alteration in habitat/hydromorphology) gradient.
However, the discriminatory power and error associated with individual metrics varied markedly, indicating that caution should
be exercised when selecting the ‘best’ organism group or metric to monitor stress. 相似文献

IS16 is a thiol-sensitive, Q-deficient mutant strain of Escherichia coli. Here, we show that IS16 harbors a mutation in the ubiG gene encoding a methyltransferase required for two O-methylation steps of Q biosynthesis. Complementation of IS16 with either ubiG or ubiX(K-12) reverses this phenotype, suggesting that UbiX may interact with UbiG. 相似文献

Burkholderia cenocepacia is an important opportunistic pathogen of patients with cystic fibrosis. This bacterium is inherently resistant to a wide range of antimicrobial agents, including high concentrations of antimicrobial peptides. We hypothesized that the lipopolysaccharide (LPS) of B. cenocepacia is important for both virulence and resistance to antimicrobial peptides. We identified hldA and hldD genes in B. cenocepacia strain K56-2. These two genes encode enzymes involved in the modification of heptose sugars prior to their incorporation into the LPS core oligosaccharide. We constructed a mutant, SAL1, which was defective in expression of both hldA and hldD, and by performing complementation studies we confirmed that the functions encoded by both of these B. cenocepacia genes were needed for synthesis of a complete LPS core oligosaccharide. The LPS produced by SAL1 consisted of a short lipid A-core oligosaccharide and was devoid of O antigen. SAL1 was sensitive to the antimicrobial peptides polymyxin B, melittin, and human neutrophil peptide 1. In contrast, another B. cenocepacia mutant strain that produced complete lipid A-core oligosaccharide but lacked polymeric O antigen was not sensitive to polymyxin B or melittin. As determined by the rat agar bead model of lung infection, the SAL1 mutant had a survival defect in vivo since it could not be recovered from the lungs of infected rats 14 days postinfection. Together, these data show that the B. cenocepacia LPS inner core oligosaccharide is needed for in vitro resistance to three structurally unrelated antimicrobial peptides and for in vivo survival in a rat model of chronic lung infection. 相似文献

The effect of cholesterol removal by methyl-beta-cyclodextrin on the dipole potential, psi(d), of membrane vesicles composed of natural membrane lipids extracted from the kidney and brain of eight vertebrate species was investigated using the voltage-sensitive fluorescent probe di-8-ANEPPS. Cyclodextrin treatment reduced cholesterol levels by on average 80% and this was associated with an average reduction in psi(d) of 50 mV. Measurements of the effect of a range of cholesterol derivatives on the psi(d) of DMPC lipid vesicles showed that the magnitude of the effect correlated with the component of the sterol's dipole moment perpendicular to the membrane surface. The changes in psi(d) observed could not be accounted for solely by the electric field originating from the sterols' dipole moments. Additional factors must arise from sterol-induced changes in lipid packing, which changes the density of dipoles in the membrane, and changes in water penetration into the membrane, which changes the effective dielectric constant of the interfacial region. In DMPC membranes, the cholesterol-induced change in psi(d) was biphasic, i.e., a maximum in psi(d) was observed at approximately 35-45 mol %, after which psi(d) started to decrease. We suggest that this could be associated with a maximum in the strength of DMPC-cholesterol intermolecular forces at this composition. 相似文献

The Chinese citrus fruit fly, Bactrocera (Tetradacus) minax (Enderlein), is one of the major citrus pests in Bhutan and can cause >50% mandarin (Citrus reticulata Blanco) fruit drop. As part of the development of a management strategy for the fly in mandarin orchards, population monitoring and experimental manipulations were carried out to determine: (i) adult emergence period; (ii) adult phenology patterns; (iii) period of crop susceptibility; and (iv) period from fruit drop to pupation. In western Bhutan, adult flies emerge from the overwintering pupal stage in late April/early May. Most flies are mature by the end of May and it is inferred that mating occurs at this time: from the beginning of June males rapidly disappear from the population and by mid- to late June are rare or absent from traps. Mature females are present in the mandarin crop at the beginning of June, but very little oviposition occurs until mid-June, while most damage has occurred by mid-July. Initiation of oviposition into mandarins is almost certainly linked to crop phenology. Adult flies disappear from the orchard system during August. After fruit drop, larvae were recorded leaving the fruit to pupate within 13 days. The use of early to mid-season protein bait sprays and/or targeted use of systemic insecticides during the one month oviposition period, plus the removal of fallen fruit once every 10 days, are recommended as control strategies. 相似文献
