Pythium indigoferae and Pythium irregulare, identified based on morphological and physiological characteristics, were isolated from necrotic roots, crown tissues and the rhizosphere of apple trees in Tunisia from 23 apple orchards in spring and autumn 2007–2009. The virulence assays on excised twigs, using different Pythium species isolated demonstrated that these oomycetes were pathogenic on the Anna, Lorka and Meski varieties and the MM106 rootstock. However, the biggest lesion area was noted on MM106 rootstock. Thus, it appeared that this rootstock is more susceptible to Pythium infections than Anna, Meski and Lorka apple varieties. Furthermore, it is important to note that in vitro tests showed that P. indigoferae seems to be more virulent than P. irregulare. 相似文献

Hitherto, aerobiology has mainly focused on studying and analysing the biologic material found in the atmosphere (pollen grains
and fungal spores), due to its allergologic and agricultural interest. However, the increase in respiratory tract problems
caused by other solid particles suspended in the air has made studies including biologic and non-biologic material more frequent.
This paper describes a simple technique based on spectrophotometric analysis. The results thus obtained can complement the
outcome obtained by traditional aerobiologic techniques if the method is used properly and the equipment properly calibrated
and an idea of the solid particles in the air can be obtained. The combination of both aerobiological and spectrophotometric
analysis makes it possible to estimate the concentration of solid-particle material in the air, and we can also estimate the
percentage of pollen grains and fungal spores among the general spectrum of particles. The results also show that a great
part of the solid particulates originate from human activities. However, it was also observed that they emerge from natural

Objectives:The purpose of the present study is to assess the effects of an intense cycling training session on the stability of the lumbopelvic-hip complex through two dynamic exercise tests - the single-leg-deadlift (SLD) and a variation of the bird-modified dog (BD), via the OCTOcore application.Methods:Thirty-one elite female road cyclists were self-evaluated with their own smartphones, before and immediately after finishing their training sessions. Right, left and composite were measured for each exercise test.Results:There was a significant time effect on performance for both the SLB and BD tests (p<0.05; η2=0.137), and the SLD and BD tests were increased with respect to the pre-test at 15% and 17%, respectively.Conclusion:An intense cycling training session produced significant alterations in lumbopelvic behavior in the elite female cyclists. The OCTOcore application demonstrated that it was a sensitive tool in detecting these changes and it could easily be used by the cyclists themselves. 相似文献

Phosphocalmodulin has been shown to have a differential biological activity compared to nonphosphorylated calmodulin when assayed on a variety of calmodulin-dependent systems. However, the phosphocalmodulin preparations used so far in those experiments were not necessarily free of nonphosphorylated calmodulin. Therefore, the results obtained may not unquestionably show the real effect of pure phosphocalmodulin on the systems under study. To solve this problem, we describe here a method for the purification of phospho(Tyr)calmodulin free of nonphosphorylated calmodulin. The procedure consists of the following steps: (i) phosphorylation of calmodulin by a fraction enriched in epidermal growth factor receptor tyrosine kinase from rat liver isolated by calmodulin affinity chromatography, (ii) isolation of a calmodulin/phosphocalmodulin mixture by Ca(2+)-dependent chromatography in phenyl-Sepharose, (iii) purification of phospho(Tyr)calmodulin using an anti-phosphotyrosine antibody immobilized in agarose upon elution with phenyl phosphate, and (iv) removal of phenyl phosphate from the phospho(Tyr)calmodulin preparation by filtration chromatography in a Bio-Gel P-2 column. The obtained phospho(Tyr)calmodulin preparation was highly pure and essentially free of nonphosphorylated calmodulin because of the use of anti-phosphotyrosine affinity chromatography. We demonstrate that this ultrapure phospho(Tyr)calmodulin preparation is totally incapable of activating the calmodulin-dependent cyclic nucleotide phosphodiesterase. In contrast, when a nonpurified phospho(Tyr)calmodulin preparation was used a partial activation of this enzyme was observed. 相似文献

Cognitive impairment is a major concern in temporal lobe epilepsy (TLE). While different experimental models have been used to characterize TLE-related cognitive deficits, little is known on whether a particular deficit is more associated with the underlying brain injuries than with the epileptic condition per se. Here, we look at the relationship between the pattern of brain damage and spatial memory deficits in two chronic models of TLE (lithium-pilocarpine, LIP and kainic acid, KA) from two different rat strains (Wistar and Sprague-Dawley) using the Morris water maze and the elevated plus maze in combination with MRI imaging and post-morten neuronal immunostaining. We found fundamental differences between LIP- and KA-treated epileptic rats regarding spatial memory deficits and anxiety. LIP-treated animals from both strains showed significant impairment in the acquisition and retention of spatial memory, and were unable to learn a cued version of the task. In contrast, KA-treated rats were differently affected. Sprague-Dawley KA-treated rats learned less efficiently than Wistar KA-treated animals, which performed similar to control rats in the acquisition and in a probe trial testing for spatial memory. Different anxiety levels and the extension of brain lesions affecting the hippocampus and the amydgala concur with spatial memory deficits observed in epileptic rats. Hence, our results suggest that hippocampal-dependent spatial memory is not necessarily affected in TLE and that comorbidity between spatial deficits and anxiety is more related with the underlying brain lesions than with the epileptic condition per se. 相似文献

Retama sphaerocarpa (L.) Boiss. is aMediterranean shrub with a remarkably simplified metameric structure. Terminalyoungest shoots act as units of modular growth, being able to produce newshootsby basal axillary buds (at the base of the shoot) and inflorescencesby lateral axillary buds. In this study, we have analysed the structural andgrowth potential features of these modules, as well as theirdemographic proportions, regarding the allocation of newvegetative and reproductive growth in plants of different age. Reproductiveeffort is proportionally higher in older plants. This shift in the allocationstrategy with plant ontogeny is not attained with changes in the shoot modules(which maintain a constant size, nutrient composition and show a similarnew growth investment per module) but through a differentdemographic composition of the population ofmodulesaccording to their developmental fate (vegetative or reproductive).This indicates a high level of iterativity and a purely modular growth, sincethe attributes of the individual (age of the plants) do not seem toaffect those of the integrating modular units (growth performance of theshoots). 相似文献

Serum deprivation of the immortalized brown adipocyte cell line resulted in growth arrest inG0/G1phases of the cell cycle and apoptosis, as detected by DNA laddering, nuclei condensation and fragmentation, and an increase in the percentage of hypodiploid cells. In addition, apoptosis in these cells is accompanied by an induction of the expression of the apoptotic form of the Bcl-x gene, the isoform Bcl-xS, and by a decrease of Bcl-2 expression, Bcl-xL remaining almost undetectable. The loss of mitochondrial membrane potential was associated with apoptosis. Z-VAD, a cell-permeable inhibitor of caspases, but not cycloheximide, precludes DNA laddering under serum deprivation. Moreover, Z-VAD rescues serum-deprived brown adipocytes from apoptosis, decreasing the percentage of hypodiploid cells, the percentage of apoptotic cells under Tunnel assay, and the external display of phosphatidylserine. More importantly, Z-VAD survival effects on immortalized brown adipocytes concur with a downregulation of Bcl-xS mRNA/protein and an upregulation of Bcl-2 protein content. Ultimately, Z-VAD prevents the loss of mitochondrial membrane potential. 相似文献
