【背景】Effective microorganisms (EM)复合菌在我国农业种植上的应用越来越广泛,但对色素辣椒的促生作用与根际细菌群落结构的影响未见报道。【目的】评估EM复合菌对新疆色素辣椒的促生长作用,并分析其对色素辣椒根际细菌群落组成的影响。【方法】通过随水灌溉方式将EM复合菌接种到色素辣椒根部,在收获期测定辣椒生长指标、土壤养分和酶活活性,明确EM复合菌对辣椒生长和土壤质量的影响;利用16S rRNA基因高通量测序技术测定EM复合菌对辣椒根系细菌群落组成和结构的影响。【结果】与对照组相比,EM复合菌的施用使辣椒株高、鲜重、单个果重和单株结果数分别提高23.89%、85.41%、42.31%和46.04%;土壤中碱解氮和速效磷含量分别提高5.83%和13.39%,土壤中脲酶、蔗糖酶和过氧化物酶的活性分别提高11.47%、9.42%和21.43%;施用EM复合菌显著改变辣椒根际微生物群落的α多样性和β多样性,提高有益菌群变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)、芽单胞菌门(Gemmatimonadetes)、放线菌门(Actinobacteria)和厚壁菌门(Firmicutes)的相对丰度,其中变形菌门黄单胞菌科(Xanthomonadaceae)的相对丰度增加119.32%;在属的水平上,施用EM复合菌显著增加了藤黄色杆菌属(Luteitalea)、藤黄单胞菌属(Luteimonas)、鞘脂单胞菌属(Sphingobacterium)和盐单胞菌属(Halomonas)的相对丰度,尤其是藤黄单胞菌属的丰度提高244.17%,同时显著降低黄杆菌属(Flavobacterium)的相对丰度。此外,与土壤理化指标呈正相关的微生物菌群相对丰度也显著升高。【结论】EM复合菌能够通过提高土壤营养成分与酶活活性,调控根系微生物群落结构,富集大量在盐碱地生存能力较强的有益菌群,进而起到促进色素辣椒生长的功效。  相似文献

As derived from a cDNA clone, the structure of the b-32 protein ofZea mays, a putative regulatory factor of zein expression, has a central acidic region separated by two domains covered by secondary structure motifs. In this work, three b-32 genomic clones were selected from two genomic libraries obtained from the maize inbred lines W64A and A69Y. The nucleotide sequences of the complete coding region of eachb-32 gene, as well as long stretches of their 5 and 3 flanking regions, were determined. Introns are not present in the b-32 genomic sequences. Minor variations among the three genes and an earlier reported b-32 cDNA indicates that they constitute a gene family showing a characteristic polymorphism. Such a polymorphism is highly evident in large segments of the upstream regulatory sequences. Interestingly, when compared with cDNA (W64A) or with geneb-32.120 (W64A), the genesb-32.129 (W64A) andb-32.152 (A69Y) show three jumps of the reading frame in the central part of the coding region, resulting in a completely different sequence of the b-32 protein central domain. In all cases, variations in the N- and C-terminal domains account only for microheterogeneity.  相似文献

基于2000—2015年MOD17A3数据及各驱动因素数据,辅以地统计学理论,利用趋势分析、Hurst指数及相关分析等方法,剖析秦巴山区近16年植被净初级生产力(NPP)时空变化特征,研究气候变化、土壤类型、地形因子、植被类型及人类活动等对植被NPP的影响.结果表明: 秦巴山区植被NPP空间上呈西南高、东北低的分布格局;时间上,近16年呈西北增、东北减的变化特征,在未来呈北部持续性、南部反持续性的发展态势.秦巴山区植被NPP与降雨量和气温呈正相关;暗棕壤、黄壤、紫色土和水稻土4种土壤类型的NPP均值明显高于其他土壤类型;不同植被类型的NPP存在空间分布和变化趋势的差异;NPP的高值主要分布在坡度25°~50°、海拔500~1000 m 以及大于2500 m的区域内;人类活动对NPP具有双重扰动性,表现为正向促进,反向抑制.  相似文献

人睫状神经营养因子(human ciliary neurotrophic factor,hCNTF)cDNA克隆入具有PL启动子的表达载体,转化大肠杆菌HB101,构建成表达hCNTF菌株。热诱导后,hCNTF的表达量达菌体总蛋白量的25%。用离子交换层析、凝胶过滤层析从菌体裂解液中纯化了hCNTF。SDS-PAGEhCNTF的分子量约为24kd,N端氨基酸序列分析结果与依基因核苷酸推导疗列相符。  相似文献

The Friedreich ataxia (FA) mutation has recently been identified as an unstable trinucleotide GAA repeat present 7-22 times in the normal population but amplified as many as > 1,000 times in FA. Since it is an autosomal recessive disease, FA does not show typical features observed in other dynamic mutation disorders, such as genetic anticipation. We have analyzed the GAA repeat in 104 FA patients and 163 carrier relatives previously defined by linkage analysis. The GAA expansion was detected in all patients, most (94%) of them being homozygous for the mutation. We have demonstrated that clinical variability in FA is related to the size of the expanded alleles: milder forms of the disease-late-onset FA and FA with retained reflexes-are associated with shorter expansions, especially with the smaller of the two expanded alleles. Absence of cardiomyopathy is also associated with shorter alleles. Dynamics of the GAA repeat has been investigated in 212 parent-offspring pairs. Meiotic instability showed a sex bias: paternally transmitted alleles tend to decrease in a linear way that depends on the paternal expansion size, whereas maternal alleles can either increase or decrease. A different pattern of intergenerational variation was also observed, depending on the genetic status of the sib: patients had shorter expansions than were seen in heterozygous carriers. This finding has been interpreted as a postzygotic event. Finally, we have observed that the size of the expansion remains constant in the population through carriers.  相似文献

The effect of N-1 -naphthylphthalamic acid (NPA), indole-3-aceticacid (IAA) and kinetin on callus growth and bud formation wasstudied mainly by a tobacco callus culture method. Callus producedfrom Nicotiana tabacum var. Wisconsin 38 was used as the testplant material. Callus growth on nutrient agar containing 2mg/liter of IAA was promoted by NPA added at a concentrationof 0.5 mg/liter with 0.4 mg/liter of kinetin or by NPA addedat 5 mg/liter in the absence of kinetin. At a high concentrationof 50 mg/liter, however, NPA inhibited growth on the mediumcontaining 2 mg/liter IAA and no kinetin. Kinetin reduced thisNPA inhibition. In the presence of 0.4 mg/liter kinetin and2 mg/liter IAA, when the concentration of NPA was 50 mg/liter,buds were initiated after calluses were grown on the test mediumfor 7 weeks in dim light, but no buds formed when NPA was omittedfrom the above medium. The control of callus growth and bud initiation is based onthe active ratio of auxin (IAA) to cytokinin (kinetin) in themedium and NPA added to the medium can promote or inhibit callusgrowth and induce bud formation. Therefore, it is proposed thatNPA can itself reduce auxin activity or enhance cytokinin activityand hence change the active ratio of the two regulators. Stable degradation products, obtained by digestion with endogenous and V8 proteases of calf thymus SS-B (La) antigenic protein, have been studied. The most characteristic fragments have molecular masses of 47, 30, 23 and 17 kDa. The 47-kDa and 30-kDa fragments are complex and are constituted of a number of species of different isoelectric points, as has been described for the SS-B protein molecule from other sources. Degradation products from the entire SS-B nuclear antigen still contain the 30-kDa SS-B fragment, suggesting that the 30-kDa region of the SS-B protein molecule is firmly attached to the RNA moiety. A model is presented that implies the presence of two hinge regions sensitive to proteases and three structural domains that correspond to segments of 30 kDa, 17 kDa and 5-6 kDa.  相似文献

病毒是危害人体健康的主要病原体之一,病毒感染和传播造成的传染性疾病严重威胁人类健康。目前,艾滋病、病毒性肝炎等发病率高、治愈率低的病毒性疾病仍在全球蔓延,流感病毒、冠状病毒等呼吸道病毒不断发生变异,2019年以来,新冠病毒引起的全球疫情对世界各国产生巨大影响,疫情走向还存在很大不确定性,开发安全有效的抗病毒药物成为应对病毒性疾病的重要手段。拟在总结全球抗病毒药物研发整体现状的基础上,分析抗艾滋病病毒、肝炎病毒、新冠病毒等重点领域的新药研发进展,提出抗病毒药物的发展建议,为未来研发更加高效的抗病毒药物提供指引和参考。  相似文献
