ABSTRACT From Bombyx mori larvae, RT-PCR and cDNA library screening isolated masquerade-like serine proteinase homologue cDNA gene, proposed to be related to insect immunity and its characteristics were examined. The isolated gene is composed of 1.3 kb of nucleotide and 420 amino acid residues were encoded. According to the results of database search, the isolated gene showed high sequence homology with Holotrichia and Tenebrio's 45 kDa protein, Drosophila CG5390 gene. Moreover, it is composed of regulatory domain and catalytic domain, which is characteristic of serine proteinase that can be found in the insect immune reaction and embryonic development processes. Enzyme activation site by proteolytic cleavage and the sequence of three amino acids participate in the catalytic triad of enzyme and 14 cystein residues used in disulfide bridges are well conserved with the compared genes. The mRNA expression was increased following E. coli injection and constitutive expression was also observed before injection by Northern blot analysis. 相似文献

Levels of allozyme variation and intrapopulation spatial genetic structure of the two terrestrial clonal orchids Liparis kumokiri , a self-compatible relatively common species, and L. makinoana , a self-incompatible rare species, were examined for 17 ( N = 1875) and four ( N = 425) populations, respectively, in South Korea. Populations of L. makinoana harboured high levels of genetic variation ( H e = 0.319) across 15 loci. In contrast, L. kumokiri exhibited a complete lack of allozyme variation ( H e = 0.000). Considering the lack of genetic variability, it is suggested that current populations of L. kumokiri in South Korea originated from a genetically depauperate ancestral population. For L. makinoana , a significant deficit of heterozygosity (mean F IS = 0.198) was found in population samples excluding clonal ramets, suggesting that pollen dispersal is localized, generating biparental inbreeding. The significant fine-scale genetic structuring (≤ 2 m) found in a previous study, in addition to the moderate levels of population differentiation ( F ST = 0.107) and the significant relationship between genetic and geographical distances ( r = 0.680) found here, suggests a leptokurtic distribution of seed dispersal for L. makinoana . Although populations of L. makinoana harbour high levels of genetic variation, they are affected by a recent genetic bottleneck. The influences of illumination, temperature, and soil water potential during development on leaf thickness, mesophyll cell wall area per unit leaf area (Ames/A), and the cellular CO2, resistance expressed on a mesophyll cell wall area basis (rCO2cell,) were examined for Plectranthus parviflorus Henckel. Although the ranges of all three growth conditions caused at least 9-fold variations in the leaf biomass produced in 4 weeks, only the illumination had a major effect on internal leaf morphology, e.g. the thickness went from 279 to 831 μm and Ames/A from 10.5 to 34.8 as the photosynthetically active radiation was raised from 3 to 53 nEinsteins cm?2 s?1, while rCO2cell remained close to 154 s cm?1. Variations in the growth temperature, soil water potential, and the nutritional status of the plant, affected photosynthesis mainly by changes in rCO2cell. To compare the influence of internal leaf area on photosynthesis for other plants, especially those with low Ames/A values, the maximum rates of CO2 uptake at light saturation and photosynthetically optimal temperatures were also determined for a moss, Mnium ciliare (C. Muell.) Schimp., and two ferns, Adiantum decorum Moore and Alsophila australe R. Br. As Ames/A went from 2.00 for the moss to 3.8, 7.5, 11.7, and 20.8 for the fens, the illumination at light saturation and the maximum rate of photosynthesis both progressively increased. ABSTRACT. We describe Hicanonectes teleskopos n. g., n. sp., a heterotrophic flagellate isolated from low-oxygen marine sediment. Hicanonectes teleskopos has a ventral groove and two unequal flagella, and rapidly rotates during swimming. At the ultrastructural level H. teleskopos is a "typical excavate": it displays flagellar vanes, a split right microtubular root, "I,""B," and "C" fibres, a singlet microtubular root, and a possible composite fibre. Small subunit rRNA (SSU rRNA) gene phylogenies and an "arched" B fibre demonstrate that H. teleskopos belongs to Fornicata (i.e. diplomonads, retortamonads, and relatives). It forms a clade with the deep-branching fornicate Carpediemonas , with moderate-to-strong bootstrap support, although their SSU rRNA gene sequences are quite dissimilar. Hicanonectes differs from Carpediemonas in cell shape, swimming behaviour, number of basal bodies (i.e. 4 vs. 3), number of flagellar vanes (i.e. 2 vs. 3), anterior root organization, and by having a cytopharynx. Like Carpediemonas and Dysnectes, Hicanonectes has conspicuous mitochondrion-like organelles that lack cristae and superficially resemble the hydrogenosomes of parabasalids, rather than the mitosomes of their closer relatives the diplomonads (e.g. Giardia ). 相似文献

The present study reports isolation and characterization of eight polymorphic microsatellite markers for Zingiber officinale Rosc. (Ginger). A total of 34 alleles were detected across the 20 accessions, with an average of 4.3 alleles per locus. Values for observed and expected heterozygosities ranged from 0 to 1.0 and from 0.23 to 0.67, respectively. The heterozygote deficits were observed at three loci. At the significance threshold (P < 0.05) of the eight loci, seven were found to have deviated from Hardy–Weinberg equilibrium, whereas significant linkage disequilibria were observed between 10 pairs of loci. Our data indicate the existence of moderate level of genetic diversity among the ginger accessions genotyped with eight markers. 相似文献

Attached 2-month-old roots of the succulent plant, Opuntia ficus-indica,shrank 0.4% radially during periods of maximal transpirationunder wet conditions. In contrast, reversible decreases in diameterof nearly 20% occurred for these roots as their ambient waterpotential () in the vapour phase decreased from 0.01to 10 MPa over 8 d, the changes being slightly more rapidat 40 °C than at 10 °C. Below5 MPa for 1-month-old roots and below 2 MPa for12-month-old roots, the soil became the main limiter of waterloss.
