本工作采用离体孵育技术,观察大鼠下丘脑薄片(含有室旁核和视上核)释放精氨酸加压素(AVP)和糖皮质激素(GC)及其他甾体激素对AVP释放的快速影响。结果如下:(1)大鼠下丘脑薄片经过90min的恢复之后,在长达6h的孵育过程中能够相当稳定地释放AVP,释放量为9.06±1.23pg/min;(2)皮质酮(B)在20min内可明显地抑制AVP的释放,在10-7—10-4mol/L范围内呈剂量-效应关系;(3)在同一剂量(10-6mol/L),皮质醇、17β-雌二醇和睾丸酮也可快速地抑制AVP的释放,而相同剂量的地塞米松、醛固酮、孕酮、RU486和胆固醇却无此效应;(4)RU486(10-7—10-3mol/L)对AVP的释放没有影响,但却能(10-5—10-3mol/L)部分地阻断B的快速抑制效应。这些结果表明,GC对大鼠下丘脑AVP的释放具有不通过传统的基因组机制的快速抑制效应,此种抑制效应可能与GC的负反馈调节作用有关。 相似文献

BackgroundInsulin-like growth factor 2 (IGF2), an essential component of the stem cell niche, has been reported to modulate the proliferation and differentiation of stem cells. Previously, a continuous expression of IGF2 in tissues was reported to maintain the self-renewal ability of several types of stem cells. Therefore, in this study, we investigated the expression of IGF2 in adipose tissues and explored the effects of IGF2 on adipose-derived stromal cells (ADSCs) in vitro.MethodsThe expression pattern of IGF2 in rat adipose tissues was determined by gene expression and protein analyses. The effect of IGF2 on proliferation, stemness-related marker expression and adipogenic and osteogenic differentiation was systematically investigated. Furthermore, antagonists of IGF2-specific receptors—namely, BMS-754807 and picropodophyllin—were added to explore the underlying signal transduction mechanisms.ResultsIGF2 levels displayed a tendency to decrease with age in rat adipose tissues. After the addition of IGF2, isolated ADSCs displayed higher proliferation and expression of the stemness-related markers NANOG, OCT4 and SOX2 and greater differentiation potential to adipocytes and osteoblasts. Additionally, both type 1 insulin-like growth factor receptor (IGF-1R) and insulin receptor (IR) participated in the IGF2-mediated promotion of stemness in ADSCs.ConclusionsOur findings indicate that IGF2 could enhance the stemness of rat ADSCs via IGF-1R and IR and may highlight an effective method for the expansion of ADSCs for clinical application. 相似文献

目的:观察人参皂苷Rg2对慢性坐骨神经损伤大鼠痛觉敏化、抑郁状态的影响。方法: 将50只 SD 大鼠随机分为 5组(n=10): 空白对照组(Normal+生理盐水腹腔注射)、假手术组(手术但不结扎+生理盐水腹腔注射) 、坐骨神经慢性压迫损伤(CCI)组(CCI +生理盐水腹腔注射) 、人参皂苷Rg2低剂量组(CCI+ Rg2 5 mg/kg腹腔注射)、人参皂苷Rg2高剂量组(CCI+ Rg2 10 mg/kg 腹腔注射)。CCI模型建立后,药物通过注射器进行腹腔内注射 5 ml/kg,每天1次,连续14 d。分别在术前1 d和术后 1、3、5、7、10、14 d测定大鼠的机械性缩足反射阈值(MWT)和热缩足潜伏期(TWL);术前1 d和术后第14日时检测明暗箱实验和强迫游泳试验。 结果:与假手术组比较,CCI组术后14 d机械痛阈值和热痛潜伏期明显缩短(P<0.01),明箱内停留时间明显缩短(P<0.01),穿梭次数明显减少(P<0.01),游泳潜伏期明显延长(P<0.01)。与CCI组比较,人参皂苷Rg2组术后14 d机械痛阈和热痛潜伏期明显增加(P<0.01),大鼠在明箱内时间明显延长(P<0.01),穿梭次数明显增多(P<0.01),且游泳潜伏期明显缩短(P<0.01)。结论:人参皂苷Rg2能抑制 CCI 大鼠的机械痛敏和热痛敏,同时改善其抑郁状态。 相似文献

A total of 10 polymorphic microsatellite loci from Hemibarbus labeo were isolated and characterized using an optimized protocol to construct a microsatellite‐enriched genomic library. The analysis of variability was performed in 24 specimens of mainland China. The mean number of alleles across loci was 3.10 ± 1.10 and the level of expected heterozygosity varied from 0.0417 to 0.7482. Frequencies of null alleles of the 10 loci are not significantly greater than zero. No linkage disequilibrium was detected between loci in either population. Five primer pairs cross‐amplify the microsatellites in other species, indicating transportability of the markers within the family Cyprinidae. 相似文献

With the expansion of saline land worldwide, it is essential to establish a model halophyte to study the salt‐tolerance mechanism. The salt glands in the epidermis of Limonium bicolor (a recretohalophyte) play a pivotal role in salt tolerance by secreting excess salts from tissues. Despite the importance of salt secretion, nothing is known about the molecular mechanisms of salt gland development. In this study, we applied RNA sequencing to profile early leaf development using five distinct developmental stages, which were quantified by successive collections of the first true leaves of L. bicolor with precise spatial and temporal resolution. Specific gene expression patterns were identified for each developmental stage. In particular, we found that genes controlling salt gland differentiation in L. bicolor may evolve in a trichome formation, which was also confirmed by mutants with increased salt gland densities. Genes involved in the special ultrastructure of salt glands were also elucidated. Twenty‐six genes were proposed to participate in salt gland differentiation. Our dataset sheds light on the molecular processes underpinning salt gland development and thus represents a first step towards the bioengineering of active salt‐secretion capacity in crops. 相似文献

We isolated eight microsatellite loci from genomic DNA in Elliot's pheasant Syrmaticus ellioti using streptavidin‐coated magnetic beads. In the analyses of 53 individuals sampled, these loci displayed polymorphism varying from six to nine alleles per locus and observed heterozygosities ranging from 0.174 to 0.430. The results suggested that these novel microsatellite markers could become useful molecular tools for genetic studies of S. ellioti. 相似文献

再生障碍性贫血(aplastic anemia,AA)是由于物理、化学、生物或不明因素作用使骨髓造血干细胞和骨髓微环境严重受损,造成骨髓造血功能降低或衰竭,以全血细胞减少为主要表现的一组综合征。间充质干细胞(MSC)是属于中胚层的一类多能干细胞,主要存在于结缔组织和器官间质中。不仅具有多向分化潜能,还有多种免疫调节作用。许多研究表明MSCs抑制同种异体效应T淋巴细胞增殖,还能下调T淋巴细胞表面的活化分子CD25、CD38、CD69的表达。MSCs对DCs的分化、成熟和活化具有抑制作用,改变DCs的细胞因子分泌,使成熟的DCl分泌TNF-a减少,DC2分泌IL-10增加,从而抑制T淋巴细胞增殖。MSCs能够抑制IL-2和IL-15介导的NK细胞增殖,使IL-2刺激NK细胞分泌的IFN-y减少。通过这些机制来干预再生障碍性贫血,同时研究发现MSCs免疫原性较低。尤其脐带MSC,因为其独特优势,目前已经成为干细胞干预治疗AA的研究热点。 相似文献
