Abstract. 1. The spatio-temporal approach was used to evaluate the environmental features influencing carabid beetle assemblages along a chronosequence of an Italian Alpine glacier foreland. The influence of environmental variables on species richness, morphology (wing and body length), and distribution along the chronosequence was tested.
2. Species richness was found to be a poor indicator of habitat due to weak influences by environmental variables. It seems that the neighbouring habitats of a glacier foreland are not able to determine significant changes in carabid species richness.
3. Instead it appears that history (age since deglaciation) and habitat architecture of a glacier foreland are strongly correlated to species adaptive morphological traits, such as wing morphology and body length. Assemblages characterised by species with reduced wing size are linked to the older stages of the chronosequence, where habitat is more structured. Assemblages characterised by the largest species are linked to the younger sites near the glacier. These morphological differentiations are explained in detail.
4. Habitat age can therefore be considered the main force determining assemblage composition. On the basis of the relationship between morphological traits and environmental variables, it seems likely that age since deglaciation is the main variable influencing habitat structure (primary effect) on the Forni foreland. The strong relationship between carabid assemblages and habitat type indicates that site age has but a secondary effect on carabid assemblages. Fungal histidine kinases (HKs) have been implicated in different processes, such as the osmostress response, hyphal development, sensitivity to fungicides and virulence. Members of HK class III are known to signal through the HOG mitogen‐activated protein kinase (MAPK), but possible interactions with other MAPKs have not been explored. In this study, we have characterized fhk1, encoding a putative class III HK from the soil‐borne vascular wilt pathogen Fusarium oxysporum. Inactivation of fhk1 resulted in resistance to phenylpyrrole and dicarboximide fungicides, as well as increased sensitivity to hyperosmotic stress and menadione‐induced oxidative stress. The osmosensitivity of Δfhk1 mutants was associated with a striking and previously unreported change in colony morphology. The Δfhk1 strains showed a significant decrease in virulence on tomato plants. Epistatic analysis between Fhk1 and the Fmk1 MAPK cascade indicated that Fhk1 does not function upstream of Fmk1, but that the two pathways may interact to control the response to menadione‐induced oxidative stress. 相似文献

Forward genetic screens are efficient tools for the dissection of complex biological processes, such as fungal pathogenicity. A transposon tagging system was developed in the vascular wilt fungus Fusarium oxysporum f. sp. lycopersici by inserting the novel modified impala element imp160::gfp upstream of the Aspergillus nidulans niaD gene, followed by transactivation with a constitutively expressed transposase. A collection of 2072 Nia+ revertants was obtained from reporter strain T12 and screened for alterations in virulence, using a rapid assay for invasive growth on apple slices. Seven strains exhibited reduced virulence on both apple slices and intact tomato plants. Five of these were true revertants showing the re-insertion of imp160::gfp within or upstream of predicted coding regions, whereas the other two showed either excision without re-insertion or no excision. Linkage between imp160::gfp insertion and virulence phenotype was determined in four transposon-tagged loci using targeted deletion in the wild-type strain. Knockout mutants in one of the genes, FOXG_00016 , displayed significantly reduced virulence, and complementation of the original revertant with the wild-type FOXG_00016 allele fully restored virulence. FOXG_00016 has homology to the velvet gene family of A. nidulans . The high rate of untagged virulence mutations in the T12 reporter strain appears to be associated with increased genetic instability, possibly as a result of the transactivation of endogenous transposable elements by the constitutively expressed transposase. 相似文献

The sarcosaprophagous fauna plays a key role in organic matter decomposition. Moreover, the biological, ecological and behavioral specificities of the taxa are useful to reconstruct the decay history of a corpse or carcass, often back to the lethal event. Here we report the seasonal succession of the insect fauna on a pig carcass exposed in a rural area in Calabria (southern Italy) during summer 2007 and 2008. The aim is to identify and qualitatively assess the major taxa of forensic importance in this region. The principal fly invaders were Lucilia caesar (L.), L. sericata (Meigen, 1826), Chrysomya albiceps (Wiedemann, 1819), Sarcophaga (Meigen, 1826) spp. and Amobia (Robineau‐Desvoidy, 1830) spp., Musca domestica (L.) and Muscina stabulans (Fallen, 1817). The primary beetle species collected in summer belonged to Dermestidae, Dermestes maculatus (De Geer, 1774) and Cleridae, Necrobia rufipes (De Geer, 1775). This paper also examined the ecological role of ants in the insect succession and describes the evidence of skin injuries directly inflicted by the acrobat ant Crematogaster (Acrocoelia) scutellaris (Olivier, 1791) (Hymenoptera Formicidae) while feeding on pig carrion. Ants belonging to two other species were also collected: Camponotus aethiops (Latreille, 1798) and Tetramorium semilaeve (André, 1881). Ants can invade carcasses and corpses directly, disrupting blowfly egg laying or preying on their larvae. Our data on the carrion faunal composition and role of ants as invaders should be useful for further forensic cases in Calabria (southern Italy). This is among the few reports of ants as forensically relevant species. 相似文献

The regulatory effects of the combined treatment of tumour necrosis factorα (TNFα), interleukin-1α (IL-1α) and interferonα(IFNα) on the growth and differentiation of Daudi lymphoma cells were investigated. By means of anti-BrdU monoclonal antibodies and [3H-thymidine] incorporation a reduced proliferation rate was shown both through a combi-nation of TNFα with either IL-1α or IFNα and, above all, through simultaneous treatment with the three cytokines. In parallel, the degree of differentiation was evaluated via morphological criteria and detection of Fc receptors (FcR) and appeared higher after treatment with the three cytokines. Our results provide evidence of the increased sensitivity of this cell line to this combined treatment supporting the existence of a synergistic interaction in inducing the antiproliferative and differentiative effects. 相似文献
