Barnea caridida oocytes release acid (1.35 pmole H+ /oocyte) upon fertilization. After artificial activation by an excess of KCl, germinal vesicle breakdown (GVBD) occurs normally and a quite similar, but not identical, acid release is recorded (1.10 pmole H+ /oocyte). KCl activation of Barnea oocytes is completely inhibited in 100 mM sodium-acetate sea water at pH 6.5 and fertilization does not result in activation when the oocytes are transferred after one minute into 100 mM sodium-acetate sea water at pH 6.3. When D–600, a calcium transmembrane fluxes inhibitor, is added 20 seconds after fertilization, GVBD is inhibited but a normal acid release is recorded. The presence of at least 10 mM sodium ions in the external medium is required for 100% activation of these oocytes by an excess of KCl. These results suggest that while an intracellular pH increase may be a requisite for GVBD, this can not be a sufficient condition to trigger it unless a calcium influx is allowed to occur. Moreover, the acid release does not result from a Ca++ -H+ exchange transport but appears more likely to be due to a Na+ -H* exchange as it has been demontrated in sea urchin eggs. 相似文献

In the context of the development of an ultrasound scanner where a probe turns around the part of the body to be studied, a new tomographic technique has been developed: the Ultrasound Reflection-mode Tomography Using Radial Image Processing (URTURIP Technique). This technique is used when a unique B-scan image is insufficient to correctly describe a cross-section. It utilises B-scan images obtained under different angles of view in the same plane to reconstruct a better cross-sectional image. Before using the URTURIP Technique, a calibration step is required to accurately determine the centre of rotation of the probe. To solve the calibration problem, a reference method has been developed, but it is very time-consuming. Two chromosomal races of house mice are present in Tunisia, one represented by mice carrying the 40-acrocentric standard karyotype and the other by a Robertsonian race (2re = 22) homozygous for nine centric fusions (Rb). A comparative summary on allozyme divergence, geographical distribution and level of reproductive isolation in the Tunisian and European Rb races is presented, to which new data on mitochondrial DNA and morphological divergence are added. The Tunisian 22Rb race revealed unique features not matched by the European chromosomal races, such as a decrease in allozymic variability, a higher level of genetic and morphological differentiation and a mosaic geographical distribution. The mtDNA analysis argued in favour of a local origin of the chromosomal divergence suggesting that the higher level of differentiation between the Tunisian races resulted from the older age of the 22Rb race and/or from a severe botdeneck. The decrease in fertility of chromosomal hybrids between the Tunisian races was compatible with the limited genetic introgression between diem. Furthermore, data on the restricted distribution of hybrid populations suggested that premating reproductive barriers may be evolving. The Tunisian 22Rb race is thus an appropriate model to investigate a chromosomally-mediated speciation event. 相似文献

ABSTRACT Alpha- and beta-tubulin cDNA were selected from a Euglenaλgt11 expression library, recloned and either sequenced (α-tubulin cDNA) or hybridized to Euglena RNA and DNA (α- and β-tubulin cDNA). RNA for hybridization was extracted at 30 minute intervals after flagellar amputation and quantitated for cDNA binding. Unlike previous reports on most other flagellates, no net increase in either α- or β-tubulin RNA could be detected during regeneration—suggesting steady state or constitutive tubulin RNA synthesis. Incubation of the cDNA with genomic DNA after restriction digestion produced patterns of hybridization consistent with the presence of one to two kinds each of the α- and β-tubulin genes. The deduced amino acid sequence of the α-tubulin cDNA was more than 90% identical to the α-tubulins of Trypanosoma, Chlamydomonas, Naegleria, Tetrahymena and higher plants. The carboxy terminus of the α-tubulin cDNA and the previously sequenced β-tubulin of Euglena showed greatest identity to the carboxy terminus of the tubulins from Trypanosoma brucei. The sequence data for α and β-tubulins of Euglena provides direct evidence for the similarity of two gene products from euglenas and trypanosomes and adds support to earlier suggestions that these organisms are phylogenetically related. 相似文献

Pollution by heavy metals is one of the strongest environmental constraints in human-altered environments that only a handful of species can cope with. Identifying the genes conferring to those species the ability to grow in polluted areas is a first step towards a global understanding of the evolutionary processes involved and will eventually improve phytoremediation practices. We used a genome-scan approach to detect loci under divergent selection among four populations of Arabidopsis halleri growing on either polluted or nonpolluted habitats. Based on a high density of amplified fragment length polymorphism (AFLP) markers (820 AFLP markers, i.e. ~1 marker per 0.3 Mb), evidence for selection was found for some markers in every sampled population. Four loci departed from neutrality in both metallicolous populations and thus constitute high-quality candidates for general adaptation to pollution. Interestingly, some candidates differed between the two metallicolous populations, suggesting the possibility that different loci may be involved in adaptation in the different metallicolous populations. 相似文献

Abstract. 1. Determinants of host-use patterns in plant-feeding insects have been extensively studied, usually within the framework of optimality theory. Comparatively, factors driving host selection in saprophagous insects have received little attention.
2. In this study, mechanisms creating occurrence peaks of saprophagous wood-borers (Cerambycidae: Coleoptera) in standing dead aspen in the middle and late stages of decay were investigated by correlating insect occurrence with variations in substrate-related nutritional and physical parameters. Twenty-four snags at four decay stages were dissected from a mature stand in western Quebec, Canada. Wood samples were taken to measure levels of nitrogen, non-structural carbohydrates, phenols, wood density, water content and snag age.
3. Several nutritional and physical parameters varied significantly along the decay gradient and were correlated with insect occurrence, but all significant parameters were also strongly correlated with snag age and wood density. Model selection using Akaike's second order information criteria was used to rank the different models; the model including snag age only performed best, with a wi of 0.873.
4. This importance of snag age gives support to a proposed hypothesis of host selection in which temporal autocorrelation in probability of insect occurrence explains peaks observed in the middle and late stages of decay. 1. Abundance and bacterial production (BP) of heterotrophic bacteria (HBact) were measured in the north and south basins of Lake Tanganyika, East Africa, during seasonal sampling series between 2002 and 2007. The major objective of the study was to assess whether BP can supplement phytoplankton particulate primary production (particulate PP) in the pelagic waters, and whether BP and particulate PP are related in this large lake. HBact were enumerated in the 0–100 m surface layer by epifluorescence microscopy and flow cytometry; BP was quantified using 3H‐thymidine incorporation, usually in three mixolimnion layers (0–40, 40–60 and 60–100 m). 2. Flow cytometry allowed three subpopulations to be distinguished: low nucleic acid content bacteria (LNA), high nucleic acid content bacteria (HNA) and Synechococcus‐like picocyanobacteria (PCya). The proportion of HNA was on average 67% of total bacterial abundance, and tended to increase with depth. HBact abundance was between 1.2 × 105 and 4.8 × 106 cells mL−1, and was maximal in the 0–40 m layer (i.e. roughly, the euphotic layer). Using a single conversion factor of 15 fg C cell−1, estimated from biovolume measurements, average HBact biomass (integrated over a 100‐m water column depth) was 1.89 ± 1.05 g C m−2. 3. Significant differences in BP appeared between seasons, especially in the south basin. The range of BP integrated over the 0–100 m layer was 93–735 mg C m−2 day−1, and overlapped with the range of particulate PP (150–1687 mg C m−2 day−1) measured in the same period of time at the same sites. 4. Depth‐integrated BP was significantly correlated to particulate PP and chlorophyll‐a, and BP in the euphotic layer was on average 25% of PP. 5. These results suggest that HBact contribute substantially to the particulate organic carbon available to consumers in Lake Tanganyika, and that BP may be sustained by phytoplankton‐derived organic carbon in the pelagic waters. 相似文献

During the last century, North African ungulate species have suffered from habitat loss and over‐hunting. Gazella dorcas (Antilopidae subfamily) and Ammotragus lervia (Caprinae subfamily) are among the ungulates that have suffered most. To help to protect these species, conservation programs and population genetics studies are being implemented. Here, we tested 30 published microsatellite primer‐pairs from Bovids (cattle, sheep and goat) on eight individuals from each species. From the 30 loci tested, 20 amplified well and showed moderate allelic richness (3.75 and 4.65 mean number of alleles per species, respectively, for G. dorcas and A. lervia), and moderatly high heterozygosity (0.53 and 0.63 per species, respectively). These 20 polymorphic markers will facilitate conservation and genetic studies in these two species, and promise to be widely useful across divergent ungulate taxa. 相似文献
