SYNOPSIS. A polymorphic allogromiid (our strain NF) was isolated in monoxenic culture but attempts at its axenic culture failed. Growth of monoxenic cultures was stimulated by various metals and vitamins. The morphology and life cycle of this allogromiid with a prominent collar have been studied in detail. It varied in form from ovoid to elongate bioral "Shepheardella-like" forms and to irregular polyoral organisms. The organisms ranged in length from 56–385 μ (118 μ± 50.54) and in width from 35–385 μ (99 μ± 39.34). The number of nuclei per organism averaged 8.9 ± 6.3 (range 1–40). Reproduction of " Shepheardella -like" forms was by binary fission. Three types of budding have been observed and, rarely, schizogony. No evidence for sexual reproduction has thus far been seen.  相似文献

Comparative biochemistry and physiology offer the advantageof specifically defining the functional parameters or traitsthat affect an organism's performance (e.g., amino acids thataffect Km, enzymes that affect metabolism). By combining thesefunctional determinations with both intraspecific and phylogeneticallyappropriate analyses, comparative biologists can indicate thata trait is biologically important by demonstrating that it isevolving by natural selection. An evolutionary approach maybenefit from the analysis of variation within and among closelyrelated species. The advantages of analyzing closely relatedspecies are that they allow one to identify more definitivelythe derived conditions and suggest why differences arose. Importantly,there is substantial variation in physiological and biochemicaltraits within and among closely related species. For example,among species within a single genus of teleost, Fundulus, thevariation in enzyme expression is similar to the variation seenamong most superorders of teleost. However, most of the variationwithin the genus Fundulus is most readily explained by evolutionarydistance, and thus there is no compelling reason for furtheradaptive hypotheses. Extending this observation, the greaterthe phylogenetic distance between taxa in a comparative study,the more likely there will be a statistically significant differencethat may only represent evolutionary time. The molecular mechanismsaffecting adaptive variation in enzyme expression appear tobe readily altered and may vary within a species or betweenacclimation conditions. Thus, studies among closely relatedorganisms are more likely to identify the specific molecularor biochemical changes responsible for adaptive variation.  相似文献

The tropical rainforest mesocosm within the Biosphere 2 Laboratory, a model system of some 110 species developed over 12 years under controlled environmental conditions, has been subjected to a series of comparable drought experiments during 2000–2002. In each study, the mesocosm was subjected to a 4–6 week drought, with well‐defined rainfall events before and after the treatment. Ecosystem CO2 uptake rate (Aeco) declined 32% in response to the drought, with changes occurring within days and being reversible within weeks, even though the deeper soil layers did not become significantly drier and leaf‐level water status of most large trees was not greatly affected. The reduced Aeco during the drought reflected both morphological and physiological responses. It is estimated that the drought‐induced 32% reduction of Aeco has three principal components: (1) leaf fall increased two‐fold whereas leaf expansion growth of some canopy dominants declined to 60%, leading to a 10% decrease in foliage coverage of the canopy. This might be the main reason for the persistent reduction of Aeco after rewatering. (2) The maximum photosynthetic electron transport rate at high light intensities in remaining leaves was reduced to 71% for three of the four species measured, even though no chronic photo‐inhibition occurred. (3) Stomata closed, leading to a reduced ecosystem water conductance to water vapour (33% of pre‐drought values), which not only reduced ecosystem carbon uptake rate, but may also have implications for water and energy budgets of tropical ecosystems. Additionally, individual rainforest trees responded differently, expressing different levels of stress and stress avoiding mechanisms. This functional diversity renders the individual response heterogeneous and has fundamental implications to scale leaf level responses to ecosystem dynamics.  相似文献

Abstract. A new model of photosynthesis published recently in this journal (H. Farazdaghi & G. E. Edwards, Plant, Cell and Environment (1988) 11 , 789–798; 799–809) clams to have a more complete mechanistic basis than currently used models based on the paper of G. D. Farquhar, S. von Caemmerer & J. A. Berry (Planta (1980) 149 , 78–90). In this paper, we examine the validity of the new kinetic expression for the rate of CO2 fixation by Rubisco, and the derivation of an equation for photosynthetic CO2 assimilation as a function of light intensity and CO2 concentration presented in the new model. In addition, we compare measured response curve of photosynthesis to CO2 and light with simulated curves using alternative models. We conclude that the new model is mechanistically misleading and, empirically, overestimates the extent to which light and CO2 co-limit the rate of photosynthesis under most physiological conditions.  相似文献

A simple, fast and sensitive method was developed to verify the presence of the sialyl Lewis(x) antigen on an N-linked glycoprotein. High performance liquid chromatography-electrospray mass spectrometry (HPLC-ESI/MS) was used to identify which of the five N-linked glycosylation sites of human plasma alpha1-acid-glycoprotein (orosomucoid, OMD) contain the sialyl Lewis(x) antigen. OMD was digested with proteolytic enzymes and analyzed by reversed phase chromatography coupled with on-line ESI/MS. A tandem mass spectrometry experiment was designed to detect the presence of the sialyl Lewis(x) antigen based on the observation of an 803 mass to charge ratio ( m/z ) ion produced in the intermediate pressure region of the ESI interface. The ESI/MS signal at m/z 803 is consistent with an oxonium ion for a glycan structure containing NeuAc, Gal, GlcNAc, and Fuc. The identity of the m/z 803 ion was confirmed by ESI/MS/MS analysis of the m/z 803 fragment ion and comparison with a sialyl Lewis(x) standard. The stereochemistry and linkage positions were assigned using previous NMR analysis but could be determined with permethylation analysis if necessary. The analysis of OMD gave a pattern showing signal for the sialyl Lewis(x) antigen coeluting with each of the five N-linked glycopeptides. The ability to monitor sialyl Lewis(x) expression at each of the five sites is of interest in the study of OMD's role in inflammatory diseases.   相似文献

Survival of vegetation on soil-capped mining wastes is often impaired during dry seasons due to the limited amount of water stored in the shallow soil capping. Growth and survival of Rhodes grass (Chloris gayana) during soil drying on various layered capping sequences constructed of combinations of topsoil, subsoil, seawater-neutralised residue sand and low grade bauxite was determined in a glasshouse. The aim was to describe the survival of Rhodes grass in terms of plant and soil water relationships. The soil water characteristic curve and soil texture analysis was a good predictor of plant survival. The combination of soil with a high water holding capacity and low soil water diffusivity (e.g. subsoil with high clay contents) with soil having a high water holding capacity and high diffusivity (e.g. residue sand) gave best survival during drying down (up to 88 days without water), whereas topsoil and low grade bauxite were unsuitable (plants died within 18–39 days). Clayey soil improved plant survival by triggering a water stress response during peak evaporative water demand once residue sand dried down and its diffusivity fell below a critical range. Thus, for revegetation in seasonally dry climates, soil capping should combine one soil with low diffusivity and one or more soils with high total water holding capacity and high diffusivity.  相似文献
