The properties of Elderberry latent virus (ELV) and Pelargonium ringspot virus (PelRSV) were compared. The viruses were largely indistinguishable in herbaceous host range and symptomatology, particle morphology, sedimentation coefficient and RNA profiles and size. They were also very closely related serologically with SDI differences in agarose gel double‐diffusion tests of 1 to 3. Purified virus particle preparations of each virus contained isometric particles c. 30 nm in diameter that sedimented as a major component with an sO20W of 112–115S. Purified virus particle preparations contained a major and a minor ssRNA species that in polyacrylamide gel electrophoresis (PAGE) had estimated sizes of c. 3.8 kb and c. 1.6 kb respectively. Plants of Chenopodium quinoa infected with ELV or PelRSV each contained three dsRNA species of c. 3.8, 2.6 and 1.8 kbp, although the smallest of these species was not evident in all preparations. Protein from purified virus particle preparations contained a major polypeptide that, in SDS‐PAGE, had an estimated Mr of 40 000 (40K). However, after storage of purified virus particles for 7–10 days, protein preparations from PelRSV particles also contained an additional major polypeptide of estimated Mr of 37 000 that is probably derived by degradation of the 40K protein; this additional component was not observed in freshly prepared preparations of ELV. Neither virus was found to be related serologically to 16 other viruses with isometric particles and similar properties. These data, together with the recent finding by other researchers that the smallest RNA species is a sub‐genomic RNA, suggests that both viruses are members of the genus Carmovirus, and that PelRSV is a minor variant of ELV. However, the taxonomic status of these two viruses is discussed in relation to recent brief reports comparing the nucleotide and amino acid sequences of these two viruses. 相似文献

Root nodule formation was studied in 11-day-old seedlings of Pisum sativum L. cv. Alaska, which were inoculated with Rhizobium leguminosarum at the time of planting. On intact plants an identical nodulation pattern was observed in the lateral roots attached to the two xylem strands connected to the cotyledons. When one cotyledon was removed before germination, however, a highly significant increase in nodultion was observed on the lateral roots attached to the xylem strand which no longer was connected to a cotyledon. Excision of one cotyledon also caused an alteration in the radial location of root nodules on the primary root. Under these conditions there was a distinct promotion of nodular proliferations in the root cortex opposite the primary phloem poles. The fact that removal of one cotyledon increased nodulation on the lateral roots but had no effect on the rhizobial infection of lateral root hairs suggested that a cotyledonary inhibitor acts at a step between the infection process and the appearance of a macroscopic nodule. 1. We investigated the spatial (longitudinal position and reach geomorphology) and seasonal (spring and autumn) influences on the variation of δ13C among organic matter sources and consumers in a forested Piedmont river, South Carolina, U.S.A. 2. Six sites were sampled along a continuum and varied in basin area from approximately 30 to 300 km2. Sites fell into two geomorphic categories (i) high‐gradient, rock bed (‘rock’) or (ii) low‐gradient, sand bed (‘sand’) sites. 3. Variation in δ13C was more strongly related to reach geomorphology than longitudinal position. δ13C of biofilm and consumers was consistently enriched at rock sites. Leaf litter (i.e. coarse particulate organic matter, CPOM) δ13C did not vary with bed type. There was significant δ13C enrichment at rock sites for biofilm, seston, fine benthic organic matter (FBOM), and eight of nine consumer trophic guilds (e.g. grazing invertebrates, insectivorous fishes). δ13C of biofilm and four trophic guilds was also positively correlated with drainage area, but the magnitude of enrichment was less than between bed types. 4. δ13C was generally enriched in spring, but this varied among organic matter types, consumers, and by bed type. CPOM and seston were enriched in spring, FBOM was enriched in autumn, and biofilm showed no trend. Five consumer guilds were enriched in spring, and only one fish guild, generalised carnivores, showed enrichment of muscle tissue in autumn. 5. Consumer δ13C enrichment at rock sites suggests greater reliance on algal carbon than for consumers at sand sites, but we also found δ13C enrichment of biofilm at rock sites. Thus, differences in consumer δ13C between bed types could be related to (i) increased consumption of biofilm at rock compared with sand sites, or (ii) consumption of biofilm at rock sites that is enriched relative to biofilm at sand sites or (iii) both mechanisms. 6. δ13C signatures in local food webs appear to respond to processes operating at multiple spatial scales. Overall downstream enrichment of biofilm and consumers was disrupted by strong local effects related to bed morphology. These results suggest that human alteration of channel habitat will have corresponding effects on stream food webs, as assessed by changes in δ13C. 相似文献

Experiments have been carried out to assess the possibility of the destruction by composting of mycelium of Didymella lycopersici in diseased tomato haulms. The results of laboratory trials with agar cultures and with infested tomato stems and fruits suggest that the fungus is destroyed if kept under moist conditions at 35° C. for from 3 to 6 days. Agar cultures buried for approximately this length of time in composting material at this or higher temperatures were dead when recovered. When composts made from haulms of tomatoes affected by Didymella stem rot were tested by placing them round the stem bases of tomato plants, or by growing tomato plants in them, few of the test plants became diseased. Sexual reproduction is one of the most taxonomically conserved traits, yet sex‐determining mechanisms (SDMs) are quite diverse. For instance, there are numerous forms of environmental sex determination (ESD), in which an organism’s sex is determined not by genotype, but by environmental factors during development. Important questions remain regarding transitions between SDMs, in part because the organisms exhibiting unique mechanisms often make difficult study organisms. One potential solution is to utilize mutant strains in model organisms better suited to answering these questions. We have characterized two such strains of the model nematode Caenorhabditis elegans. These strains harbour temperature‐sensitive mutations in key sex‐determining genes. We show that they display a sex ratio reaction norm in response to rearing temperature similar to other organisms with ESD. Next, we show that these mutations also cause deleterious pleiotropic effects on overall fitness. Finally, we show that these mutations are fundamentally different at the genetic sequence level. These strains will be a useful complement to naturally occurring taxa with ESD in future research examining the molecular basis of and the selective forces driving evolutionary transitions between sex determination mechanisms. 相似文献

In this article, we describe the analysis of over 9000 expressed sequence tags (ESTs) from cDNA libraries obtained from various life cycle stages of Globodera pallida . We have identified over 50 G. pallida effectors from this dataset using bioinformatics analysis, by screening clones in order to identify secreted proteins up-regulated after the onset of parasitism and using in situ hybridization to confirm the expression in pharyngeal gland cells. A substantial gene family encoding G. pallida SPRYSEC proteins has been identified. The expression of these genes is restricted to the dorsal pharyngeal gland cell. Different members of the SPRYSEC family of proteins from G. pallida show different subcellular localization patterns in plants, with some localized to the cytoplasm and others to the nucleus and nucleolus. Differences in subcellular localization may reflect diverse functional roles for each individual protein or, more likely, variety in the compartmentalization of plant proteins targeted by the nematode. Our data are therefore consistent with the suggestion that the SPRYSEC proteins suppress host defences, as suggested previously, and that they achieve this through interaction with a range of host targets. 相似文献

As well as their direct ecological impacts on native taxa, invasive species can impose selection on phenotypic attributes (morphology, physiology, behaviour, etc.) of the native fauna. In anurans, body size at metamorphosis is a critical life‐history trait: for most challenges faced by post‐metamorphic anurans, larger size at metamorphosis probably enhances survival. However, our studies on Australian frogs (Limnodynastes convexiusculus) show that this pattern can be reversed by the arrival of an invasive species. When metamorph frogs first encounter invasive cane toads (Bufo marinus), they try to eat the toxic invader and, if they are able to do so, are likely to die from poisoning. Because frogs are gape‐limited predators, small metamorphs cannot ingest a toad and thus survive long enough to disperse away from the natal pond (and thus from potentially deadly toads). These data show that larger size at metamorphosis can reduce rather than increase anuran survival rates, because larger metamorphs are more easily able to ingest (and thus be poisoned by) metamorph cane toads. Our results suggest that patterns of selection on life‐history traits of native taxa (such as size and age at metamorphosis, seasonal timing of breeding and duration of pondside aggregation prior to dispersal) can be modified by the arrival of an invasive species. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 100 , 329–336. 相似文献
