Pine pollen (Pinus mugo) grown in suspension cultures readily utilize exogenous carbohydrates for tube growth and starch synthesis: these processes are not influenced by β-indolylacetic acid, gibberellic acid, kinetin and abscisic acid. It appears that the free sugars of the female gametophyte, namely sucrose, raffinose, and stachyose and their monosaccharide constituents, are the best substrates for growth and polysaccharide synthesis. The oligosaccharides are hydrolysed to their monosaccharide constituents before they are taken up. A preferential uptake of fructose is noted. Non-metabolizable sugars are not taken up. The data presented establish that tube growth, except for the initial growth phase, can be determined by the availability of exogenous carbohydrates. Measurements of some of the key enzymes in carbohydrate metabolism show that the enzymes were present in the ungerminated pollen grain, and that the enzyme activity increased severalfold during tube growth. The increase in enzyme activity was prevented if inhibitors of protein synthesis were present in the medium.  相似文献

The rate of change in dormancy level and light requirement,induced during stratification at 3.2 °C, was investigatedin seeds ofStellaria media, Cerastium fontanum, Veronica agrestisandTaraxacumofficinale.Two stratification environments, soil and wet filterpaper in petri-dishes, were used. On nine occasions during a6 week stratification period, germination was tested under threelight conditions at 3.5/18.5 °C: (1) darkness; (2) light;and (3) short-term light exposure followed by darkness. Priorto stratification, germination in all species was 89% in lightand 53% in darkness. Within 2 weeks of stratification, germinabilityinV. agrestisandS. mediaseeds decreased in all treatments. InC.fontanumandT. officinaleseeds, germinability also decreasedafter 2 weeks but only in the dark treatment, indicating inductionof a light requirement. After two more weeks of stratification,the induced dormancy inS. mediaseeds stratified in soil becameweaker and the light requirement inT. officinaleseeds was lost.Differences between the two stratification environments and/orinteractions between light conditions, stratification environmentsand time were found for all species. These results suggest that:(1) the dormancy level and the light requirement of seeds maychange dramatically over relatively short-time periods duringstratification; and (2) germinability depends on the stratificationenvironment experienced by the seeds. Active uptake of boron (B) by excised barley roots is linear with time for at least 1.5 h. Although no evidence was found for accumulation of B against a concentration gradient. this component of B uptake does satisfy other criteria for an active transport process. Transport is inhibited by 0.05 mM 2,4-dinitrophenol, 0.05 mM azide, 5 mM arsenate and 5 mM dicoumarol. Also, uptake is temperature-sensitive, being nil at 2°C and maximal at 34 to 38°C. Boron uptake by barley roots increases with time when they are washed in aerated 0.5 mM CaSO4 solution. A double reciprocal plot of the B uptake data manifests a series of phases separated by sharp transitions or “jumps”, and is compatible with the concept of multiphasic uptake mechanisms. Kinetic constants and transition points for the various phases were calculated accordingly. The fit of these data was compared statistically to three other relevant models, viz, the dual model, the “single + diffusion” model (a Michaelis–Menten term and a diffusion term), and the negative cooperativity model. Multiphasic Uptake of Amino Acids by Barley Roots   总被引:10,自引:0,他引:10  
Concentration-dependence and other characteristics of uptake of 3H-labeled l -lysine, l -methionine and l -proline by excised roots of barley (Hordeum vulgare L.) were studied. Use of relatively short uptake and wash periods and low solute concentrations ensured good estimates of influx across the plasmalemma. Uptake in the range of 10?7M– 6.3 × 10?3M can be precisely represented by four or five phases of single, multiphasic mechanisms. The mechanisms appear to be relatively specific as judged from the competition by unlabeled analogues. Structural requirements for interaction of a compound with the uptake site for methionine are given, as are the effects of analogues on the phase pattern for this amino acid. There is no indication of separate uptake and transition sites for methionine or lysine. i.e. phase transitions seem in this case to be caused by binding of molecule(s) to the uptake site. Uptake, but not phase patterns, was highly pH-dependent. The optima were pH 5 for lysine, pH 3–5 (a broad peak) for methionine and about pH 5.5 for proline. Uptake of the three amino acids was strongly inhibited by 2,4-dinitrophenol. sulfhydryl reagents and deoxycholate.  相似文献

We developed 31 novel, polymorphic microsatellite loci in the marbled murrelet (Brachyramphus marmoratus), a critically endangered seabird. Variability was tested on 15 individuals from the Santa Cruz, California population, with each locus characterized by two to 12 alleles. Observed levels of heterozygosity ranged from 0.13 to 0.93. These loci provide a valuable means of assessing the population structure and demographic parameters of this species, which may be critical to its conservation across a fragmented habitat.  相似文献

Uptake of sulfate by excised barley roots increases upon their washing in aerated water or dilute CaCl2 solutions. Washing increases the values for Vmax and the sulfate concentrations required for transition between the lower phases, but the KM-values remain essentially constant. At low sulfate concentrations, phase transitions do not occur in the absence of calcium or other divalent cations. These ions are about equally effective in enhancing short-term sulfate uptake. Phase transitions were not principally altered by sulfhydryl or protein reagents. These concentration-dependent transitions appear unrelated to temperature-dependent phase transitions as evidenced by similar multiphasic patterns at low and high temperature.  相似文献

Mass estimates for a number of fossil proboscideans were computed using regression analyses on appendicular bones to body mass, for seven specimens of modern elephants, for which body masses had been recorded prior to death. The marked differences in physical proportions between extant Loxodonta and Elephas , implying substantial differences in body mass at any given shoulder height, were not present in their long bone parameters. Length and least circumferences proved to be the best parameters for prediction of body mass. Some extinct proboscideans, notably certain Mammuthus and Deinotherium , were much larger than extant elephants. Both the basal and the field metabolic rates of extant elephants are lower than predicted for a hypothetical mammal, in accordance with their body size and subsistence on low-quality foods.
