The use of aqueous alkali for the titration of esterolytic activity when the esters are dissolved in alcoholic solutions, results in an error due to changes in the ionization of the buffer. This is corrected by titrating with alkali in the same solvent as the substrate. Alcohols and other substances which change the dielectric strength of water modify the rate of hydrolysis of BAEE1 and TSAME by trypsin to an extent proportionate to their effect on the dielectric strength. The reaction rate increases with diminished dielectric strength and vice versa. At low concentrations of substance there seems to be no specific effect other than that derived of the variation in dielectric strength. At higher concentrations, the enzyme might be denatured. In addition, it is probable that specific effects of each substance might intervene. The Coulombic and thermic energies of activation were calculated for the two esters in various solvents. The plot of the logarithm of rate constant vs. reciprocal of dielectric constant yields a straight line with positive slope. This behavior is similar to that of a non-enzymatic positive ion-dipole reaction. Trypsin reacts like a positive ion. The possible influence of the dielectric strength on the regulation of the equilibria involved in the interconversion of the various forms of trypsin in solution (active, inactive, denatured) is discussed. 相似文献

Genes of the major histocompatibility complex (MHC) play a fundamental role in the vertebrate immune response and are amongst the most polymorphic genes in vertebrate genomes. It is generally agreed that the highly polymorphic nature of the MHC is maintained through host–parasite co‐evolution. Two nonexclusive mechanisms of selection are supposed to act on MHC genes: superiority of MHC heterozygous individuals (overdominance) and an advantage for rare MHC alleles. However, the precise mechanisms and their relative importance are still unknown. Here, we examined MHC dependent parasite load in European rabbits (Oryctolagus cuniculus) from a distinct population with low MHC diversity (three alleles, six genotypes). Using a multivariate approach, we tested for associations of individual MHC class II DRB constitution and the rabbits’ intestinal burden with nematodes and coccidia. Rabbits having a particular allele showed lower infestations with hepatic coccidia (E. stiedai). However, a comparison of all six genotypes in the population revealed that carriers of this allele only benefit when they are heterozygous, and furthermore, MHC heterozygosity in general did not affect individual parasite load. In conclusion, this study suggests an immunogenetic basis of European rabbit resistance to hepatic coccidiosis, which can strongly limit survival to maturity in this species. Our study gives a complex picture of MHC–parasite correlations, unveiling the limits of the classical hypotheses of how MHC polymorphism is maintained in natural systems. 相似文献

Diel (24-h) time courses of CO2 exchange, water relations, and microclimate of the foliose lichen, Sticta tomentosa (Swartz) Ach., and responses to experimentally manipulated conditions were measured at a forest edge in a lower montane rainforest in Panama.
Similar to earlier observations on two other rain forest lichens, daily desiccation suppressed net photosynthesis (NP) during the period when irradiation was highest. Not surprisingly, the light response curves of NP showed saturation at rather low light levels. Rehydration was associated with an initial resaturation burst of short duration, which could be demonstrated both under natural conditions and experimentally. This additional loss of CO2 seems too low to be ecologically relevant. Moreover, high thallus hydration was also detrimental to NP: at maximum water content net CO2 uptake was depressed by >50%. Although NP was well adapted to the prevailing high temperatures, the latter also stimulated dark respiration. On average, almost 60% of the diurnal carbon gain was lost during the night.
In spite of these limitations, the integrated 24-h C gain was quite high, on average 0·5% of the thallus C content. Whilst these figures were determined for horizontally exposed samples, we also assessed the role of different exposures on photosynthetic performance. Diel C gain was highest under conditions of semi-shade (westerly exposure), which allowed long periods of activity, whilst much higher irradiance at other exposures could not be utilized for photosynthetic production: easterly exposed thalli dried out even faster than horizontally exposed samples. 相似文献
