SYNOPSIS. Mice were used as a model system to demonstrate theeffects of increased loading on the trabecular bone in the developingproximal femur. A two-dimensional free body diagram revealedthat because the femur is normally held in a horizontal positionduring support, functional loading resulted in a net posteriorbending moment and a muscle generated axial compressive loadthat was 7.7 times the normal support weight for that limb.Experimental treatment consisted of four times normal gravityfor 10 min, six times per day for 30 days in addition to unrestrainednormal exercise. Unrestrained normal exercise served as thecontrol. An animal whose foot was amputated by his mother shortlyafter birth was used as a less than normal load example forcomparison with the hypergravity and control groups. All groupswere treated for 30 days. Two areas of trabecular bone wereexamined: a proximal central webbing area that may be underpredominantly tensile loading much of the time, and a more distalarea near the third trochanter that presumably experiences bendingstresses when the animal is exposed to normal activity or hypergravity.Results indicate that: 1) The central webbing area appears tohave a similar mass of bone regardless of loading conditions;however, with increased loading the trabeculae become orientedin directions that approximate directions of presumed principaltensile stresses. 2) There is no genetically predetermined amountof bony tissue in the third trochanter area; when the limb isrelatively unloaded no trabeculae are present, as the loadingincreases trabecular bone is formed. In the hypergravity group,trabeculae in the area of the third trochanter increased inlength and complexity and became more centrally positioned inthe lumen of the femur, a position that presumably would allowthe femur to better resist bending moments. These results demonstratethat short duration, high intensity functional loading can stimulategrowth of trabecular bone, and further, specific types of functionalloading can be correlated with specific architectural changes.  相似文献

Ultrastructural examination of crayfish superficial (tonic)and deep (phasic) abdominal extensor muscles reveals a distributionand quantitative difference in glycogen between these muscles.Both superficial and deep fibers have a dense accumulation ofglycogen in the interfibrillar sarcoplasm. In addition, thesuperficial extensors, but not the deep extensors, contain glycogenin the I band region. The glycogen granules are of the ßtype and can be removed enzymatically. The superficial medialand lateral fibers contain more glycogen than the medial andlateral deep fibers. A possible functional role for this differenceis suggested.  相似文献

Conidia of the aphid pathogen Erynia neoaphidis on detached bean leaves and glass coverslips maintained at 20°C and at humidities from 40 to 100% r.h. lost their infectivity for pea aphids at a rate dependent on the humidity. Infectivity declined most rapidly in inocula kept on leaves at 70% r.h. and persisted longest in those at 40 and 50% r. h. That of inocula on coverslips declined most rapidly at 77% r.h. and persisted longest in those kept at 40% r.h. Even after 21 days a little infectivity was retained by conidia stored at 50% r.h. on leaves and 40% r.h. on coverslips.  相似文献

The surface proteins and glycoproteins on red cells from normal and Babesia bovis-infected calf blood have been compared. Several radiolabeling probes were used to label specifically external membrane molecules which were then separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and identified by autoradiography or fluorography. No differences were observed among the Coomassie Blue-stained membrane proteins of erythrocytes from individual uninfected calves. Comparison of red cells from these animals also indicated no qualitative differences in the surface proteins with accessible tyrosyl residues labeled by lactoperoxidase-catalyzed radioiodnation, although some quantitative variation in the uptake of radioactivity into particular proteins was observed. the major radioiodinated bands on normal bovine erythrocytes had Mr of 165, 130, 90, and 45 kiloDaltons. However, labeling of surface glycoproteins by the periodate/[3H]NaBH4 and galactose oxidase (± neuraminidase)/[3H]NaBH4 methods showed significant differences in the surface proteins of red cells from individual uninfected calves. of 14 animals tested, 5 had major labeled glycoproteins of unique Mr. No changes were observed in radioiodinated surface proteins of total red cell samples from infected calves with 0.5-6% parasitemia. Radioiodination of concentrated infected red cells from the same samples (concentrated by selective hypotonic lysis of uninfected erythrocytes in KC1) resulted in the labeling of 3 new surface proteins, with Mr of 118, 115, and 60 kiloDaltons. the same new 125I-labeled bands were identified on infected cells from 3 avirulent strains of B. bovis used in vaccine production. Furthermore, in concentrated infected cells there was very poor radiolabeling of major bands strongly labeled on uninfected cells (Mr 165, 130, and 90 kiloDaltons), suggesting parasite-induced loss of these proteins. Although there were some differences in 3H-labeled surface glycoproteins of red cells from normal and. B. bovis -infected blood, they were restricted to minor labeled bands and were not seen consistently. the labeled surface glycoproteins of concentrated infected cells were very similar to those of the uninfected red blood cells from infected blood.  相似文献

SYNOPSIS. Glugea gasti sp. n., a microsporidan pathogen of Anthonomus grandis Boheman (the boll weevil), is described and a probable life cycle presented. The alimentary canal, and probably the mesenteron 1st, is the initial site of infection, altho the disease later becomes generalized thruout most body tissues. Binucleate sporoplasms initiate the 1st schizogonic phase, characterized by mono- and bi-nucleate schizonts. The 2nd schizogonic phase is characterized by mono-, bi- and quadrinucleate schizonts, by prolific multiplication, by the dense compact nuclei early in this phase, and late in this phase by larger schizonts with less dense vesicular nuclei. This phase terminates in formation of diplokarya. The sporogonic phase is characterized by combination of the 2 nuclei in the diplokaryon followed by nuclear divisions in a sequence closely resembling meiosis. Two sporoblasts are produced from each sporont. Mature spores in wet mounts by phase contrast were 4.3 ± 0.3 μ long by 2.3 ± 0.2 μ wide. The polar filament averaged 76 μ long. Mature spores were present about 24 hours after infection. Some observations are presented on an external filament extending from one pole of the spore to host tissue and other events during the process of spore morphogenesis.  相似文献

C-TYPE viruses have been established as the causal agents of leukaemia in murine and feline species and have been characterized1,2. C-type virus is also probably associated with fibrosarcoma in non-human primates3–6. To determine whether viruses with identical characteristics are associated with other neoplasms in simian species, we looked for C-type viruses in cases of leukaemia. A gibbon (Hylobates lar) with a disseminated tumour (later confirmed as lymphosarcoma) was made available to the Comparative Oncology Laboratory by Dr Malcolm Jones of the University of California, San Francisco Medical Center. The principal sites of involvement (lymph node, liver and bone marrow) were extensively overrun with massive neoplastic cells, which were predominantly prolymphocytic forms. Electron microscopy revealed C-type particles identical to those observed in vitro in sections from lymph nodes, liver, spleen and bone marrow.  相似文献

The existence of dieldrin resistance in Erioischia brassicae has been investigated in the main areas in England and Wales where brassicas are grown for human consumption. Dieldrin-susceptible and dieldrin-resistant flies were collected and bred as separate strains in the laboratory. In toxicity tests adult flies were treated with topically applied doses of insecticide in microdrops of cellosolve, and larvae were dipped in solutions of insecticide in 70% acetone and 30% water. Adults of the laboratory resistant strain showed a high level of resistance to dieldrin and aldrin and a lower level of resistance to γ-BHC. There was no cross-resistance with chlorfenvinphos or diazinon. Larvae of the resistant strain were shown to be resistant to dieldrin. Dieldrin resistance has been found in four areas in southern England, but not in a fifth major area, and resistant flies did not increase as rapidly in some areas as others. Sufficient migration of flies appeared to take place to produce similar percentages of resistant flies over areas of several square miles, but considerable differences were found in fly populations about 10 miles apart. It is suggested that the speed of development of insecticide resistance is affected by the proportion of brassica crops which are treated in an area, the amounts of insecticide applied to individual crops, and hence the insecticidal pressure applied to the E. brassicae population.  相似文献

Flow commenced in November 1970 and ceased in July 1971, a maximum flow of 1.51 m3 s?1 being recorded in January 1971.
