Two modern cultivars [Yangmai16 (Y16) and Yangfumai 2 (Y2)] of winter wheat (Triticum aestivum L.) with almost identical phenology were investigated to determine the impacts of elevated ozone concentration (E‐O3) on physiological characters related to photosynthesis under fully open‐air field conditions in China. The plants were exposed from the initiation of tillering to final harvest, with E‐O3 of 127% of the ambient ozone concentration (A‐O3). Measurements of pigments, gas exchange rates, chlorophyll a fluorescence and lipid oxidation were made in three replicated plots throughout flag leaf development. In cultivar Y2, E‐O3 significantly accelerated leaf senescence, as indicated by increased lipid oxidation as well as faster declines in pigment amounts and photosynthetic rates. The lower photosynthetic rates were mainly due to nonstomatal factors, e.g. lower maximum carboxylation capacity, electron transport rates and light energy distribution. In cultivar Y16, by contrast, the effects of E‐O3 were observed only at the very last stage of flag leaf ageing. Since the two cultivars had almost identical phenology and very similar leaf stomatal conductance before senescence, the greater impacts of E‐O3 on cultivars Y2 than Y16 cannot be explained by differential ozone uptake. Proliferation in cardiac fibroblasts (CFs) can be induced by a wide variety of growth factors that recruit multiple signal transduction pathways, including mitogen-activated protein kinase, phosphatidylinositol 3-kinase and protein kinase C. As a family of dimeric phophoserine-binding proteins, 14-3-3s are associated with a multitude of proteins that regulate signal transduction, apoptosis and checkpoint control pathways. However, it remains unknown whether the 14-3-3 proteins play an active role in cardiac proliferation and alter their expression patterns in response to growth factors in CFs. R18 peptide, an isoform-independent 14-3-3 inhibitor, was used to disrupt 14-3-3 function by adenovirus-mediated transfer of R18-EYFP (AdR18). Our results demonstrate that the 14-3-3 isoforms gamma, zeta and epsilon were highly expressed in CFs and the expression of 14-3-3 epsilon was elevated following serum stimulation. Inhibition of 14-3-3 proteins by AdR18 potentiated mitogen-induced DNA synthesis in CFs. This potentiation was presumably due to the increased inactivated glycogen synthase kinase-3 beta by Ser9 phosphorylation and nuclear factor of activated T-cell nuclear accumulation. However, AdR18 had no effect on extracellular signal-regulated kinase phosphorylation and reduced p70 S6 kinase (p70S6K) phosphorylation upon mitogenic stimulation. Furthermore, though R18 can block 14-3-3 binding abilities, it did not affect the serum-induced upregulation of 14-3-3 epsilon protein. Collectively, these findings reveal that the expression of 14-3-3 epsilon can be upregulated by serum in CFs and 14-3-3s may exert an inhibitory effect on serum-induced proliferation. 相似文献

Tengia has been called a "natural peloria" in the family Gesneriaceae because it exhibits an almost perfect actinomorphic flower from whorl one to whorl three. It would be especially interesting to know whether or how CYC-like gene activities are related to this type of perfect actinomorphic flower. To address this, we have isolated four CYC-like TCP genes and carried out an investigation on their expression patterns in Tengia. TsCYC1C and TsCYC1D have similar expression patterns with strong signals being detected in all five petals and stamens, whereas TsCYC2A and TsCYC2B are only transiently expressed in the very early floral meristem. Our results suggest that the expansion of the expressions of TsCYC1C and TsCYC1D from the dorsal to the ventral petals is likely responsible for the evolutionary formation of the fully dorsalized actinomorphic corolla, that is, an expanded functional domain of CYC-like gene dorsal identity in Tengia corolla. However, the expressions of TsCYC1C and TsCYC1D are not correlated with stamen abortion; therefore, TsCYC genes do not functionally repress the stamen development in Tengia flowers.
